    Аналитически установлено, что высокоскоростное фрезерование располагает значительными технологическими возможностями с точки зрения уменьшения шероховатости поверхности при одновременном увеличении производительности обработки. Установлено также, что при шлифовании уменьшение шероховатости поверхности связано с уменьшением производительности. Наиболее прогрессивным методом шлифования, обеспечивающим одновременно увеличение производительности и уменьшение шероховатости поверхности, является глубинное шлифование с небольшой скоростью детали, которое характеризуется меньшей производительностью по сравнению с высокоскоростным фрезерованием.The paper presents the results of theoretical studies of the surface roughness during milling and grinding. It is shown that high-speed milling has significant technological capabilities in terms of reducing surface roughness, because cutting data parameters are included in the calculated dependencies obtained to determine surface roughness with higher degrees than during grinding. This applies in particular to the speed of rotation of the cutter. Therefore, with its increase, it becomes possible to significantly reduce the surface roughness while increasing the processing capacity, which opens up broad prospects for the practical use of high-speed milling. It is established that during grinding, a decrease in surface roughness is associated with a decrease in productivity, and this reduces the efficiency of processing. The most progressive method of grinding, providing b oth an increase in productivity and a reduction in surface roughness, is deep-grinding at a low speed of the part. However, it is characterized by lower productivity in comparison with high-speed milling

    Targeting class I histone deacetylases by the novel small molecule inhibitor 4SC-202 blocks oncogenic hedgehog-GLI signaling and overcomes smoothened inhibitor resistance

    Aberrant activation of Hedgehog (HH)/GLI signaling is causally involved in numerous human malignancies, including basal cell carcinoma (BCC) and medulloblastoma. HH pathway antagonists targeting smoothened (SMO), an essential effector of canonical HH/GLI signaling, show significant clinical success in BCC patients and have recently been approved for the treatment of advanced and metastatic BCC. However, rapid and frequent development of drug resistance to SMO inhibitors (SMOi) together with severe side effects caused by prolonged SMOi treatment call for alternative treatment strategies targeting HH/GLI signaling downstream of SMO. In this study, we report that 4SC-202, a novel clinically validated inhibitor of class I histone deacetylases (HDACs), efficiently blocks HH/GLI signaling. Notably, 4SC-202 treatment abrogates GLI activation and HH target gene expression in both SMOi-sensitive and -resistant cells. Mechanistically, we propose that the inhibition of HDACs 1/2/3 is crucial for targeting oncogenic HH/GLI signaling, and that class I HDAC inhibitors either in combination with SMOi or as second-line therapy may improve the treatment options for HH-associated malignancies with SMOi resistance

Vor dem Hintergrund des niedrigen Zinsumfelds suchen institutionelle Investoren nach alternativen Anlagevehikeln mit zufriedenstellenden Rendite-Risiko-Verhältnissen. Diese Arbeit untersucht die Marktakzeptanz des deutschen Erbbaurechts sowie mögliche Einflüsse aus Großbritannien als einem der höchstentwickelten Märkte für Erbbaurecht. Unsere Analyse zeigt, dass das Erbbaurecht ein sinnvolles Nischenprodukt für institutionelle Investoren zur Generierung von Mehrwerten darstellt. Der individuelle Nutzen für Investoren mit deutschem Investmentfokus hängt jedoch stark von der konkreten Vertragsgestaltung ab. Der britische Markt stellt für deutsche Immobilienfonds aktuell aufgrund der höheren Liquidität und Transparenz des Marktes eine geeignetere Anlageallokation dar

    Extent of posterolateral tibial plateau impaction fracture correlates with anterolateral complex injury and has an impact on functional outcome after ACL reconstruction

    PURPOSE The impact of posterolateral tibial plateau impaction fractures (TPIF) on posttraumatic knee stability in the setting of primary anterior cruciate ligament (ACL) tear is unknown. The main objective was to determine whether increased bone loss of the posterolateral tibial plateau is associated with residual rotational instability and impaired functional outcome after ACL reconstruction. METHODS A cohort was identified in a prospective enrolled study of patients suffering acute ACL injury who underwent preoperative standard radiographic diagnostics and clinical evaluation. Patients were included when scheduled for isolated single-bundle hamstring autograft ACL reconstruction. Exclusion criteria were concurrent anterolateral complex (ALC) reconstruction (anterolateral tenodesis), previous surgery or symptoms in the affected knee, partial ACL tear, multi-ligament injury with an indication for additional surgical intervention, and extensive cartilage wear. On MRI, bony (TPIF, tibial plateau, and femoral condyle morphology) and ligament status (ALC, concomitant collateral ligament, and meniscus injuries) were assessed by a musculoskeletal radiologist. Clinical evaluation consisted of KT-1000, pivot-shift, and Lachman testing, as well as Tegner activity and IKDC scores. RESULTS Fifty-eight patients were included with a minimum follow-up of 12 months. TPIF was identified in 85% of ACL injuries (n = 49). The ALC was found to be injured in 31 of 58 (53.4%) cases. Pearson analysis showed a positive correlation between TPIF and the degree of concomitant ALC injury (p < 0.001). Multiple regression analysis revealed an increased association of high-grade TPIF with increased lateral tibial convexity (p = 0.010). The high-grade TPIF group showed worse postoperative Tegner scores 12 months postoperatively (p = 0.035). CONCLUSION Higher degrees of TPIFs are suggestive of a combined ACL/ALC injury. Moreover, patients with increased posterolateral tibial plateau bone loss showed lower Tegner activity scores 12 months after ACL reconstruction. LEVEL OF EVIDENCE III

    Cultivation of the PHB-producing cyanobacterium Synechococcus leopoliensis in a pilot-scale open system using nitrogen from waste streams

    PHB-producing cyanobacteria may provide the raw material for bio-based and biodegradable plastics. To commercialize photoautotrophic PHB production, their cultivation needs to be scaled up in open systems and to reduce costs and increase sustainability, nutrients must be obtained from waste streams. Here, the feasibility of these steps was verified. Different PHB-producing cyanobacteria were compared in laboratory-scale cultivations using either water from recirculating aquaculture systems or pre-processed liquid digestate as nutrient sources. Then, Synechococcus leopoliensis was cultivated in an open thin-layer photobioreactor (18 m2, 200 L), were growth in mineral Z-medium was again compared to said waste streams. All cultivations were successful. Cultivation in mineral medium resulted in both the highest final biomass yield (6 g L−1) and productivity (0.7 g L−1 d−1). Both waste stream-based media showed lower biomass yields and productivities (2 g L−1 and 0.25–0.3 g L−1 d−1). However, due to differences in the cultivation conditions (e.g., temperature, nutrient supply), final biomass yield and productivity do not represent the performance of the cultivations adequately. Relative parameters such as nitrogen and energy conversion ratios indicate that cultivation with aquaculture water suffered from insufficient nitrogen supply to the culture, whereas the use pre-processed liquid digestate resulted in a substantial loss of nitrogen due to volatilization. All cultivations in the open system were continued in the laboratory, where cultures were starved for ten days under nutrient-depleted conditions (without nitrogen, phosphorus, or both). While PHB accumulation occurred, concentrations were comparatively low (< 1%dw). The comparison of the results suggests that PHB yields were influenced more by the initial cultivation condition than by the specific type of nutrient depletion. Thus, while the cultivation with waste streams in an open system is feasible, environmental parameters seem to influence PHB yields considerably and must be considered for the optimization of the complete process

    AbstractObjective: The present cross sectional study was performed to test the hypothesis that in osteoarthritis (OA) of the knee severity of this disease is related to local levels of inflammatory metabolites and their corresponding enzymes.Methods: From 41 patients with OA of the knee (age range 45–79 years) undergoing arthroscopy blood, synovial fluid (SF) and synovial membrane (SM) were collected. Clinical conditions were primarily assessed by the WOMAC-index and radiographic grading (K&L-grade). Concentrations of PGE2, TxB2and NO2/3and that of IL-6, IL-1α, IL-1β, TNFα, COX-2 and iNOS were determined in SF and SM, respectively.Results: With advancing age K&L-grade and COX-2 in SM increased significantly (P=0.005 and P=0.01, respectively). TNFα and IL-1α were not detectable in SM samples. Apart from a correlation between PGE2and WOMAC-index (r=0.36, P=0.035) no significant relationships could be found between the various inflammatory parameters and any of the assessed clinical signs.Conclusions: Apparently no direct relationships exist between the measured markers of inflammation (e.g. PGE2, NO2/3) or the involved enzymes (e.g. COX-2, iNOS) and the severity of OA of the knee. The degenerative condition of this disease might be due to the more local, mainly mechanical injury with little systemic upset. However, further longitudinal studies are needed to clarify whether the assessed biochemical markers could serve as predictors for the progression of OA

    Investigating health impacts of natural resource extraction projects in Burkina Faso, Ghana, Mozambique, and Tanzania: protocol for a mixed methods study

    Natural resource extraction projects offer both opportunities and risks for sustainable development and health in host communities. Often, however, the health of the community suffers. Health impact assessment (HIA) can mitigate the risks and promote the benefits of development but is not routinely done in the developing regions that could benefit the most.; Our study aims to investigate health and health determinants in regions affected by extractive industries in Burkina Faso, Ghana, Mozambique, and Tanzania. The evidence generated in our study will inform a policy dialogue on how HIA can be promoted as a regulatory approach as part of the larger research initiative called the HIA4SD (Health impact assessment for sustainable development) project.; The study is a concurrent triangulation, mixed methods, multi-stage, multi-focus project that specifically addresses the topics of governance and policy, social determinants of health, health economics, health systems, maternal and child health, morbidity and mortality, and environmental determinants, as well as the associated health outcomes in natural resource extraction project settings across four countries. To investigate each of these health topics, the project will (1) use existing population-level databases to quantify incidence of disease and other health outcomes and determinants over time using time series analysis; (2) conduct two quantitative surveys on mortality and cost of disease in producer regions; and (3) collect primary qualitative data using focus groups and key informant interviews describing community perceptions of the impacts of extraction projects on health and partnership arrangements between the projects and local and national governance. Differences in health outcomes and health determinants between districts with and without an extraction project will be analyzed using matched geographical analyses in quasi-Poisson regression models and binomial regression models. Costs to the health system and to the households from diseases found to be associated with projects in each country will be estimated retrospectively.; Fieldwork for the study began in February 2019 and concluded in February 2020. At the time of submission, qualitative data collection had been completed in all four study countries. In Burkina Faso, 36 focus group discussions and 74 key informant interviews were conducted in three sites. In Ghana, 34 focus group discussions and 64 key informant interviews were conducted in three sites. In Mozambique, 75 focus group discussions and 103 key informant interviews were conducted in four sites. In Tanzania, 36 focus group discussions and 84 key informant interviews were conducted in three sites. Quantitative data extraction and collection is ongoing in all four study countries. Ethical approval for the study was received in all four study countries prior to beginning the fieldwork. Data analyses are underway and results are expected to be published in 2020 and 2021.; Disentangling the complex interactions of resource extraction projects with their host communities requires an integrative approach drawing on many methodologies under the HIA umbrella. By using complementary data sources to address the question of population health in project areas from several angles, bias and missing data will be reduced, generating high-quality evidence to aid countries in moving toward sustainable development.; DERR1-10.2196/17138

    Podoplanin-positive cells are a hallmark of encapsulating peritoneal sclerosis

    Background. Encapsulating peritoneal sclerosis (EPS) and simple peritoneal sclerosis are important complications of long-term peritoneal dialysis (PD). Podoplanin is expressed by mesothelial cells and lymphatic vessels, which are involved in inflammatory reactions in the peritoneal cavity. Methods. We studied 69 peritoneal biopsies from patients on PD (n = 16), patients with EPS (n = 18) and control biopsies taken at the time of hernia repair (n = 15) or appendectomy (n = 20). Immunohistochemistry was performed to localize podoplanin. Additionally, markers of endothelial cells, mesothelial cells, myofibroblasts (smooth muscle actin), proliferating cells, and double labelling for smooth muscle actin/podoplanin were used on selected biopsies. Results. Podoplanin was present on the endothelium of lymphatic vessels in the submesothelial fibrous tissue and on mesothelial cells. In patients on PD and in biopsies with appendicitis, the mesothelial cells demonstrated a cuboidal appearance and circumferential podoplanin staining, with gaps between the cells. The number of lymphatic vessels was variable, but prominent at sites of fibrosis. In patients with EPS, a diffuse infiltration of podoplanin-positive cells with a fibroblastic appearance was present in 15 out of 18 biopsies. This pattern was focally present in 3 out of 16 on PD and none in the 35 controls. The podoplanin-positive cells did not express the endothelial marker or the mesothelial marker (calretinin). Conclusions. EPS is characterized by a population of podoplanin and smooth muscle actin double-positive cells. Podoplanin might be a suitable morphological marker supporting the diagnosis and might be involved in the pathogenesis of EP
