    The OARSI histopathology initiative – recommendations for histological assessments of osteoarthritis in the horse

    SummaryObjectiveEquine models of osteoarthritis (OA) have been used to investigate pathogenic pathways of OA and evaluate therapeutic candidates for naturally occurring equine OA which is a significant clinical disease in the horse. This review focuses on the macroscopic and microscopic criteria for assessing naturally occurring OA in the equine metacarpophalangeal joint as well as the osteochondral fragment-exercise model of OA in the equine middle carpal joint.MethodsA review was conducted of all published OA studies using horses and the most common macroscopic and microscopic scoring systems were summarized. Recommendations regarding methods of OA assessment in the horse have been made based on published studies.ResultsA modified Mankin scoring system is recommended for semi-quantitative histological assessment of OA in horses due to its already widespread use and similarity to other scoring systems. Recommendations are also provided for histological scoring of synovitis and macroscopic lesions of OA as well as changes in the calcified cartilage and subchondral bone of naturally occurring OA.ConclusionsThe proposed system for assessment of equine articular tissues provides a useful method to quantify OA change. It is believed that addition of quantitative tracing onto plastic and macroscopic measurement as recently described would be an improvement for overall assessment of articular cartilage change

    Manure production on grower/finisher swine operations in the United States was examined using data from 184 grower/finisher swine operations that participated in the United States National Animal Health Monitoring System's 1995 National Swine Study. Two methods were used: one, assuming that pigs produced 8.4% of their body weight in manure each day; another using the difference between feed fed and weight gained as a proxy variable to study manure production. Using this latter approach, a production function was developed. The function exhibited diminishing returns to scale when food waste was not fed to pigs, but constant returns to scale when food waste was included in their diets. The difference between feed fed and weight gained was lower on operations that restricted entry to employees only.<br>A produção de dejetos em granjas de crescimento e terminação de suínos nos Estados Unidos foi avaliada utilizando dados de 184 granjas participantes de um estudo nacional de 1995 do "United States National Animal Health Monitoring System".     Despite the large and well-documented characterization of the microbial cell wall in terms of chemical composition, the determination of the mechanical properties of surface molecules in relation to their function remains a key challenge in cell biology.The emergence of powerful tools allowing molecular manipulations has already revolutionized our understanding of the surface properties of fungal cells. At the frontier between nanophysics and molecular biology, atomic force microscopy (AFM), and more specifically single-molecule force spectroscopy (SMFS), has strongly contributed to our current knowledge of the cell wall organization and nanomechanical properties. However, due to the complexity of the technique, measurements on live cells are still at their infancy.In this chapter, we describe the cell wall composition and recapitulate the principles of AFM as well as the main current methodologies used to perform AFM measurements on live cells, including sample immobilization and tip functionalization.The current status of the progress in probing nanomechanics of the yeast surface is illustrated through three recent breakthrough studies. Determination of the cell wall nanostructure and elasticity is presented through two examples: the mechanical response of mannoproteins from brewing yeasts and elasticity measurements on lacking polysaccharide mutant strains. Additionally, an elegant study on force-induced unfolding and clustering of adhesion proteins located at the cell surface is also presented
