    Involvement of carbon dioxide in the aerobic biodegradation of ethylene oxide, ethene, and vinyl chloride

    The involvement of a carboxylase in metabolism of C-2 alkenes by Ochrobactrum sp. strain TD and Pseudomonas putida strain AJ was examined. With resting cells of strain TD grown on vinyl chloride, ethene, and ethylene oxide, the maximum specific rate of ethylene oxide consumption decreased significantly in the absence of external CO2 in comparison to cells provided with room air or added CO2. The amount of 14CO2 incorporated into biomass by resting cells of strain TD grown on ethylene oxide increased more than 13-fold when the assay substrate was ethylene oxide versus acetate. These results indicate that strain TD uses a carboxylase. Similar experiments were performed with strain AJ with the results suggesting that a carboxylase is not involved. In this regard, strain AJ is more similar to various Mycobacterium isolates that also do not appear to use a carboxylase during metabolism of vinyl chloride and ethene.Department of Biomedical and Biomolecular Sciences, Sheffield Hallam University

    Biodegradation of Dichloromethane and Its Utilization as a Growth Substrate under Methanogenic Conditions

    Biodegradation of dichloromethane (DCM) to environmentally acceptable products was demonstrated under methanogenic conditions (35 degrees C). When DCM was supplied to enrichment cultures as the sole organic compound at a low enough concentration to avoid inhibition of methanogenesis, the molar ratio of CH4 formed to DCM consumed (0.473) was very close to the amount predicted by stoichiometric conservation of electrons. DCM degradation was also demonstrated when methanogenesis was partially inhibited (with 0.5 to 1.5 mM 2-bromoethanesulfonate or approximately 2 mM DCM) or completely stopped (with 50 to 55.5 mM 2-bromoethanesulfonate). Addition of a eubacterial inhibitor (vancomycin, 100 mg/liter) greatly reduced the rate of DCM degradation. 14CO2 was the principal product of [14C]DCM degradation, followed by 14CH4 (when methanogenesis was uninhibited) or 14CH3COOH (when methanogenesis was partially or completely inhibited). Hydrogen accumulated during DCM degradation and then returned to background levels when DCM was consumed. These results suggested that nonmethanogenic organisms mediated DCM degradation, oxidizing a portion to CO2 and fermenting the remainder to acetate; acetate formation suggested involvement of an acetogen. Methanogens in the enrichment culture then converted the products of DCM degradation to CH4. Aceticlastic methanogens were more easily inhibited by 2-bromoethanesulfonate and DCM than were CO2-reducing methanogens. When DCM was the sole organic-carbon and electron donor source supplied, its use as a growth substrate was demonstrated. The highest observed yield was 0.085 g of suspended organic carbon formed per g of DCM carbon consumed.     Reduction and Acetylation of 2,4-Dinitrotoluene by a Pseudomonas aeruginosa Strain

    Aerobic and anoxic biotransformation of 2,4-dinitrotoluene (DNT) was examined by using a Pseudomonas aeruginosa strain isolated from a plant treating propellant manufacturing wastewater. DNT biotransformation in the presence and absence of oxygen was mostly reductive and was representative of the type of cometabolic transformations that occur when a high concentration of an easily degradable carbon source is present. P. aeruginosa reduced both nitro groups on DNT, with the formation of mainly 4-amino-2-nitrotoluene and 2-amino-4-nitrotoluene and small quantities of 2,4-diaminotoluene. Acetylation of the arylamines was a significant reaction. 4-Acetamide-2-nitrotoluene and the novel compounds 2-acetamide-4-nitrotoluene, 4-acet-amide-2-aminotoluene, and 2,4-diacetamidetoluene were identified as DNT metabolites. The biotransformation of 2,4-diaminotoluene to 4-acetamide-2-aminotoluene was 24 times faster than abiotic transformation. 2-Ni-trotoluene and 4-nitrotoluene were also reduced to their corresponding toluidines and then acetylated. How-ever, the yield of 4-acetamidetoluene was much higher than that of 2-acetamidetoluene, demonstrating that acetylation at the position para to the methyl group was favored

    Task Dependence of Visual and Category Representations in Prefrontal and Inferior Temporal Cortices

    Visual categorization is an essential perceptual and cognitive process for assigning behavioral significance to incoming stimuli. Categorization depends on sensory processing of stimulus features as well as flexible cognitive processing for classifying stimuli according to the current behavioral context. Neurophysiological studies suggest that the prefrontal cortex (PFC) and the inferior temporal cortex (ITC) are involved in visual shape categorization. However, their precise roles in the perceptual and cognitive aspects of the categorization process are unclear, as the two areas have not been directly compared during changing task contexts. To address this, we examined the impact of task relevance on categorization-related activity in PFC and ITC by recording from both areas as monkeys alternated between a shape categorization and passive viewing tasks. As monkeys viewed the same stimuli in both tasks, the impact of task relevance on encoding in each area could be compared. While both areas showed task-dependent modulations of neuronal activity, the patterns of results differed markedly. PFC, but not ITC, neurons showed a modest increase in firing rates when stimuli were task relevant. PFC also showed significantly stronger category selectivity during the task compared with passive viewing, while task-dependent modulations of category selectivity in ITC were weak and occurred with a long latency. Finally, both areas showed an enhancement of stimulus selectivity during the task compared with passive viewing. Together, this suggests that the ITC and PFC show differing degrees of task-dependent flexibility and are preferentially involved in the perceptual and cognitive aspects of the categorization process, respectively.National Institute of Mental Health (U.S.) (5R01MH065252-12)Alfred P.     Frenetic: A High-Level Language for OpenFlow Networks

    Network administrators must configure network devices to simultaneously provide several interrelated services such as routing, load balancing, traffic monitoring, and access control. Unfortunately, most interfaces for programming networks are defined at the low level of abstraction supported by the underlying hardware, leading to complicated programs with subtle bugs. We present Frenetic, a high-level language for OpenFlow networks that enables writing programs in a declarative and compositional style, with a simple "program like you see every packet" abstraction. Building on ideas from functional programming, Frenetic offers a rich pattern algebra for classifying packets into traffic streams and a suite of operators for transforming streams. The run-time system efficiently manages the low-level details of (un)installing packet-processing rules in the switches.     The Luminosity of SN 1999by in NGC 2841 and the Nature of `Peculiar' Type Ia Supernovae

    We present UBVRIJHK photometry and optical spectroscopy of the so-called 'peculiar' Type Ia supernova 1999by in NGC 2841. The observations began one week before visual maximum light which is well-defined by daily observations. The light curves and spectra are similar to those of the prototypical subluminous event SN 1991bg. We find that maximum light in B occurred on 1999 May 10.3 UT (JD 2,451,308.8 +/- 0.3) with B=13.66 +/- 0.02 mag and a color of B_max-V_max=0.51 +/- 0.03 mag. The late-time color implies minimal dust extinction from the host galaxy. Our photometry, when combined with the recent Cepheid distance to NGC 2841 (Macri et al. 2001), gives a peak absolute magnitude of M_B=-17.15 +/- 0.23 mag, making SN 1999by one of the least luminous Type Ia events ever observed. We estimate a decline rate parameter of dm15(B)=1.90 mag, versus 1.93 for SN 1991bg, where 1.10 is typical for so-called 'normal' events. We compare SN 1999by with other subluminous events and find that the B_max-V_max color correlates strongly with the decline rate and may be a more sensitive indicator of luminosity than the fading rate for these objects. We find a good correlation between luminosity and the depth of the spectral feature at 580 nm, which had been attributed solely to Si II. We show that in cooler photospheres the 580 nm feature is dominated by Ti II, which provides a simple physical explanation for the correlation. Using only subluminous Type Ia supernovae we derive a Hubble parameter of H_0=75 +12 -11 km/s Mpc, consistent with values found from brighter events.Comment: 36 preprint pages including 18 figures.     Apolipoprotein L1 gene (APOL1) G1 and G2 coding variants are strongly associated with chronic kidney disease (CKD) in African Americans (AAs). Here APOL1 association was tested with baseline estimated glomerular filtration rate (eGFR), urine albumin:creatinine ratio (UACR), and prevalent cardiovascular disease (CVD) in 2571 AAs from the Systolic Blood Pressure Intervention Trial (SPRINT), a trial assessing effects of systolic blood pressure reduction on renal and CVD outcomes. Logistic regression models that adjusted for potentially important confounders tested for association between APOL1 risk variants and baseline clinical CVD (myocardial infarction, coronary, or carotid artery revascularization) and CKD (eGFR under 60 ml/min per 1.73 m(2) and/or UACR over 30 mg/g). AA SPRINT participants were 45.3% female with a mean (median) age of 64.3 (63) years, mean arterial pressure 100.7 (100) mm Hg, eGFR 76.3 (77.1) ml/min per 1.73 m(2), and UACR 49.9 (9.2) mg/g, and 8.2% had clinical CVD. APOL1 (recessive inheritance) was positively associated with CKD (odds ratio 1.37, 95% confidence interval 1.08-1.73) and log UACR estimated slope (β) 0.33) and negatively associated with eGFR (β -3.58), all significant. APOL1 risk variants were not significantly associated with prevalent CVD (1.02, 0.82-1.27). Thus, SPRINT data show that APOL1 risk variants are associated with mild CKD but not with prevalent CVD in AAs with a UACR under 1000 mg/g

    Observational constraints on the spectral index of the cosmological curvature perturbation

    We evaluate the observational constraints on the spectral index nn, in the context of the Λ\LambdaCDM hypothesis which represents the simplest viable cosmology. We first take nn to be practically scale-independent. Ignoring reionization, we find at a nominal 2-σ\sigma level n1.0±0.1n\simeq 1.0 \pm 0.1. If we make the more realisitic assumption that reionization occurs when a fraction f105f\sim 10^{-5} to 1 of the matter has collapsed, the 2-σ\sigma lower bound is unchanged while the 1-σ\sigma bound rises slightly. These constraints are compared with the prediction of various inflation models.     The Carnegie Supernova Project: Analysis of the First Sample of Low-Redshift Type-Ia Supernovae

    We present the analysis of the first set of low-redshift Type Ia supernovae (SNe Ia) by the Carnegie Supernova Project. Well-sampled, high-precision optical (ugriBV) and near-infrared (NIR; YJHKs) light curves obtained in a well-understood photometric system are used to provide light-curve parameters, and ugriBVYJH template light curves. The intrinsic colors at maximum light are calibrated to compute optical--NIR color excesses for the full sample, thus allowing the properties of the reddening law in the host galaxies to be studied. A low value of Rv~1.7, is derived when using the entire sample of SNe. However, when the two highly reddened SNe in the sample are excluded, a value Galactic standard of Rv~3.2 is obtained. The colors of these two events are well matched by a reddening model due to circumstellar dust. The peak luminosities are calibrated using a two-parameter linear fit to the decline rates and the colors, or alternatively, the color excesses. In both cases, dispersions in absolute magnitude of 0.12--0.16 mag are obtained, depending on the filter-color combination. In contrast to the results obtained from color excesses, these fits give Rv~1--2, even when the two highly reddened SNe are excluded. This discrepancy suggests that, beyond the "normal" interstellar reddening produced in the host galaxies, there is an intrinsic dispersion in the colors of SNe Ia which is correlated with luminosity but independent of the decline rate. Finally, a Hubble diagram is produced by combining the results of the fits for each filter. The resulting scatter of 0.12 mag appears to be limited by peculiar velocities as evidenced by the strong correlation between the distance-modulus residuals among the different filters.
