    Syntheses and Properties of Salts of Chromophores with Ferrocenyl Electron Donor Groups and Quaternary Nitrogen Acceptors

    A series of five new dipolar cations has been synthesized with ferrocenyl (Fc) electron donor groups connected to N-arylpyridinium, N-methylquinolinium, N-methylbenzothiazolium, or N-methylacridinium acceptors. Together with their known N-methylpyridinium analogue, these chromophores have been characterized as their PF_6^− salts by using various techniques including electronic absorption spectroscopy and cyclic voltammetry. Nine single-crystal X-ray structures have been determined, including two polymorphs of one salt obtained from a single crystallization experiment, and two of these are polar materials. A highly favorable degree of dipolar alignment for bulk NLO effects is observed in one case. Molecular quadratic nonlinear optical (NLO) responses have been determined by using femtosecond hyper-Rayleigh scattering (HRS) at 1300 nm and also via Stark (electroabsorption) spectroscopic studies on the intense π → π^* intraligand and d → π^* metal-to-ligand charge-transfer bands. A broad correlation between the electron acceptor strength and the HRS-derived first hyperpolarizabilities β and the static first hyperpolarizabilities β0 estimated from the Stark data is evident. This is the first time that meaningful (albeit indirectly determined) β_0 data have been reported for Fc compounds, allowing quantitative comparisons with the chromophore in the technologically important material (E)-4′-(dimethylamino)-N-methyl-4-stilbazolium (DAS) tosylate. The observed β_0 values are in several cases similar to that of [DAS]PF_6, and possibly even larger in one instance

    Pedological and geochemical investigations at the „Red Outcrop“ of Langenlois (Lower Austria)

    In einem Aufschluss von Löss-Paläoboden-Sequenzen über Rehberger Amphibolit NW von Langenlois wurden sechs Bodenprofile (Lois 1 bis Lois 6) beprobt und pedologisch, mineralogisch und geochemisch analysiert. Am Profil Lois 7 wurden bodenmikromorphologische Untersuchungen durchgeführt. Zwei Bodenprofile (Lois 1 und 2) haben sich über Amphibolit entwickelt, zwei über einer Amphibolit/Marmor-Wechsellagerung (Lois 5 und 6) und drei Bodenprofile sind Sequenzen von polyzyklischen Paläoböden mit fossilen Bodenhorizonten ohne unterlagerndes kristallines Gestein (Lois 3, 4 und 7). In den Profilen Lois 1–4 und Lois 7 konnten intensive Karbonatanreicherungen beobachtet werden. Diese hohen Mengen an Karbonat können nicht Produkt einer rezenten Bodenbildung sein, sondern sprechen für eine Infiltration von ursprünglich das Profil überlagernden kalzitreichen Sedimenten (Löss), die erodiert wurden. Zudem wurde in Profil Lois 2 ein Kalksinter angetroffen. Dieser Kalksinter könnte aus der Verwitterung der im Profil Lois 6 aufgeschlossenen Marmorlagen stammen. Die Kalksinter-Schicht scheint die Stoffflüsse zwischen unterliegendem Gestein und Solum in den Profilen Lois 1 und 2 mehr oder weniger zu unterbinden, was auch durch die Ergebnisse der geochemischen Analytik unterstrichen wird. In den fossilen Horizonten wurden auch ältere Anzeichen von Tonverlagerung in Form von Tonkutanen über den Aggregaten und darüber hinaus leichte Pseudovergleyungserscheinungen angetroffen. Die mächtigen Profile ohne aufgeschlossenes Grundgebirge (Lois 3, 4 und 7) weisen mehrere polyzyklische Sedimentationsphasen und dadurch mehrere Generationen von fossilen Horizonten auf. Aufgrund der bodenmikromorphologischen Analyse können die Böden vom „Roten Aufschluss“ altersmäßig im unteren bis mittleren Pleistozän oder älter angesiedelt werden.researc

    Due to its extensive use in various livestock production activities and its use in human health care, antibiotics have become an environmental problem that draws the attention of the scientific community. Several research works report their presence in different ecosystems compartments, as well as their impact in organisms that inhabit these ecosystems which are being investigated by this community; until now, it has been reported problems of bacterial resistance and damages at DNA level in living beings, among others; in this perspective it is necessary to perform monitoring in various environmental matrixes, in order to detect and quantify their presence, to have a better understanding of their long-term effects on living beings. In this sense, it addresses aspects that determine their presence in the ecosystem, as well as shows results of works evaluating their removal of contaminated water, in order to ensure their safety from an environmental perspective and taking into consideration the health of living beings and human beings.Debido a su amplio uso en diversas actividades de producción de ganado y su uso en el cuidado de la salud humana, los antibióticos, se han convertido en un problema ambiental que llama la atención de la comunidad científica. Diversos trabajos reportan su presencia en diversos compartimientos de los ecosistemas, así como su impacto a los organismos que habitan estos ecosistemas se encuentran siendo investigados por esta comunidad, hasta el momento se han reportado problemas de resistencia bacteriana y daños a nivel de ADN en seres vivos, entre otros, por lo que, se hace necesario realizar el monitoreo en diversas matrices ambientales, con el fin de detectar y cuantificar su presencia, para tener un mejor conocimiento de su efectos a largo plazo en seres vivos. En ese sentido, se abordan aspectos que determinan su presencia en el ecosistema, así como se muestra resultados de trabajos que evalúan su remoción de aguas contaminadas, con el fin de garantizar su seguridad desde una perspectiva ambiental y llevando en consideración la salud de seres vivos y del mismo ser humano

    Magnetization transfer ratio relates to cognitive impairment in normal elderly

    Magnetization transfer imaging (MTI) can detect microstructural brain tissue changes and may be helpful in determining age-related cerebral damage. We investigated the association between the magnetization transfer ratio (MTR) in gray and white matter (WM) and cognitive functioning in 355 participants of the Austrian stroke prevention family study (ASPS-Fam) aged 38-86 years. MTR maps were generated for the neocortex, deep gray matter structures, WM hyperintensities, and normal appearing WM (NAWM). Adjusted mixed models determined whole brain and lobar cortical MTR to be directly and significantly related to performance on tests of memory, executive function, and motor skills. There existed an almost linear dose-effect relationship. MTR of deep gray matter structures and NAWM correlated to executive functioning. All associations were independent of demographics, vascular risk factors, focal brain lesions, and cortex volume. Further research is needed to understand the basis of this association at the tissue level, and to determine the role of MTR in predicting cognitive decline and dementia

    Cultivation and Immortalization of Human B-Cells Producing a Human Monoclonal IgM Antibody Binding to MDA-LDL: Further Evidence for Formation of Atherogenic MDA-LDL Adducts in Humans In Vivo

    Oxidatively modified low-density lipoprotein (oLDL) is firmly believed to play an important role in the initiation and development of atherosclerosis, and malonic dialdehyde (MDA) is one of the major lipid peroxidation breakdown products involved in this process. In recent decades, antibodies against MDA-LDL have been detected in human and animal sera. In our study, human B-cells from the peripheral blood of a healthy female donor were fused with the SP2/0 mouse myeloma cell line. Antibody-producing hybridomas were detected by MDA-LDL-IgG/IgM enzyme-linked immunosorbent assays (ELISA) and Cu++-oxidized LDL IgG/IgM (oLAb) ELISA. Cells with supernatants emitting positive signals for antibodies were then cloned and after sufficient multiplication frozen and stored under liquid nitrogen. Due to the loss of antibody-producing ability, we established an MDA-LDL-IgM-producing cell line by recloning. This allowed isolation and immortalization of several human B-cells. The human donor had not been immunized with MDA-modified proteins, thus obviously producing MDA-LDL antibodies in vivo. Furthermore, using these antibodies for in vitro experiments, we were able to demonstrate that MDA epitopes are among the epitopes generated during Cu++-LDL oxidation as well. Finally, these antibodies compete in ELISA and cell culture experiments with MDA as a challenging toxin or ligand

    The Mitochondrial T16189C Polymorphism Is Associated with Coronary Artery Disease in Middle European Populations

    BACKGROUND: The pivotal role of mitochondria in energy production and free radical generation suggests that the mitochondrial genome could have an important influence on the expression of multifactorial age related diseases. Substitution of T to C at nucleotide position 16189 in the hypervariable D-loop of the control region (CR) of mitochondrial DNA (mtDNA) has attracted research interest because of its suspected association with various multifactorial diseases. The aim of the present study was to compare the frequency of this polymorphism in the CR of mtDNA in patients with coronary artery disease (CAD, n = 482) and type 2 diabetes mellitus (T2DM, n = 505) from two study centers, with healthy individuals (n = 1481) of Middle European descent in Austria. METHODOLOGY AND PRINCIPAL FINDINGS: CR polymorphisms and the nine major European haplogroups were identified by DNA sequencing and primer extension analysis, respectively. Frequencies and Odds Ratios for the association between cases and controls were calculated. Compared to healthy controls, the prevalence of T16189C was significantly higher in patients with CAD (11.8% vs 21.6%), as well as in patients with T2DM (11.8% vs 19.4%). The association of CAD, but not the one of T2DM, with T16189C remained highly significant after correction for age, sex and body mass index (BMI) and was independent of the two study centers. CONCLUSIONS AND SIGNIFICANCE: Our results show for the first time a significant association of T16189C with CAD in a Middle European population. As reported in other studies, in patients with T2DM an association with T16189C in individuals of European decent remains questionable

    Mitochondrial DNA haplogroup T is associated with coronary artery disease and diabetic retinopathy: a case control study

    <p>Abstract</p> <p>Background</p> <p>There is strong and consistent evidence that oxidative stress is crucially involved in the development of atherosclerotic vascular disease. Overproduction of reactive oxygen species (ROS) in mitochondria is an unifying mechanism that underlies micro- and macrovascular atherosclerotic disease. Given the central role of mitochondria in energy and ROS production, mitochondrial DNA (mtDNA) is an obvious candidate for genetic susceptibility studies on atherosclerotic processes. We therefore examined the association between mtDNA haplogroups and coronary artery disease (CAD) as well as diabetic retinopathy.</p> <p>Methods</p> <p>This study of Middle European Caucasians included patients with angiographically documented CAD (n = 487), subjects with type 2 diabetes mellitus with (n = 149) or without (n = 78) diabetic retinopathy and control subjects without clinical manifestations of atherosclerotic disease (n = 1527). MtDNA haplotyping was performed using multiplex PCR and subsequent multiplex primer extension analysis for determination of the major European haplogroups. Haplogroup frequencies of patients were compared to those of control subjects without clinical manifestations of atherosclerotic disease.</p> <p>Results</p> <p>Haplogroup T was significantly more prevalent among patients with CAD than among control subjects (14.8% vs 8.3%; p = 0.002). In patients with type 2 diabetes, the presence of diabetic retinopathy was also significantly associated with a higher prevalence of haplogroup T (12.1% vs 5.1%; p = 0.046).</p> <p>Conclusion</p> <p>Our data indicate that the mtDNA haplogroup T is associated with CAD and diabetic retinopathy in Middle European Caucasian populations.</p
