Using Y-chromosome capture enrichment to resolve haplogroup H2 shows new evidence for a two-Path Neolithic expansion to Western Europe
Uniparentally-inherited markers on mitochondrial DNA (mtDNA) and the non-recombining regions of the Y chromosome (NRY), have been used for the past 30 years to investigate the history of humans from a maternal and paternal perspective.Researchers have preferred mtDNA due to its abundance in the cells, and comparatively high substitution rate. Conversely, the NRY is less susceptible to back mutations and saturation, and is potentially more informative than mtDNA owing to its longer sequence length. However, due to comparatively poor NRY coverage via shotgun sequencing, and the relatively low and biased representation of Y-chromosome variants on capture arrays such as the 1240K, ancient DNA studies often fail to utilize the unique perspective that the NRY can yield.Here we introduce a new DNA enrichment assay, coined YMCA (Y-mappable capture assay), that targets the “mappable” regions of the NRY. We show that compared to low-coverage shotgun sequencing and 1240K capture, YMCA significantly improves the coverage and number of sites hit on the NRY, increasing the number of Y-haplogroup informative SNPs, and allowing for the identification of previously undiscovered variants.To illustrate the power of YMCA, we show that the analysis of ancient Y-chromosome lineages can help to resolve Y-chromosomal haplogroups. As a case study, we focus on H2, a haplogroup associated with a critical event in European human history: the Neolithic transition. Bullet impacts and built heritage damage 1640–1939
© 2018, The Author(s). Conflict damage to heritage has been thrust into the global spotlight during recent conflict in the Middle East. While the use of social media has heightened and enhanced public awareness of this ‘cultural terrorism’, the occurrence of this type of vandalism is not new. In fact, as this study demonstrates, evidence of the active targeting of sites, as well as collateral damage when heritage is caught in crossfire, is widely visible around Europe and further afield. Using a variety of case studies ranging from the 1640s to the 1930s, we illustrate and quantify the changing impact of ballistics on heritage buildings as weaponry and ammunition have increased in both energy and energy density potential. In the first instance, this study highlights the increasing threats to heritage in conflict areas. Second, it argues for the pressing need to quantify and map damage to the stonework in order to respond to these challenges
Phenolic and furanic compounds of Portuguese chestnut and French, American and Portuguese oak wood chips
Botanical species used on aging process must
be wisely and judiciously chosen, and for this selection, a
basic knowledge of the chemical composition of woods is
warranted. Aiming to contribute to extend the knowledge
of the chemical composition of several wood species useful
for enological purposes, we have focused our studies on
Portuguese chestnut and French, American and Portuguese
oak chips. The profile of low molecular weight phenolic
composition of these chips was achieved, using an optimized
extraction method based on pressurized liquid
extraction, followed by the quantification of phenolic acids,
phenolic aldehydes and furanic derivatives by high-performance
liquid chromatography (HPLC-DAD). The
identification of those compounds was also confirmed by
LC-DAD/ESI-MS. This study allowed the determination of
the low molecular phenolic composition of Portuguese
chestnut and French, American and Portuguese oak wood.
According to our results, the influence of the botanical
species seems to be more relevant than the geographic
origin of the wood species
