Paradoxical exploitation of protected fishes as bait for anglers: evaluating the lamprey bait market in Europe and developing sustainable and ethical solutions
A reoccurring conservation problem is the resolution of consumptive use of threatened wildlife and is especially difficult to defend when it occurs for recreational practices. We explored the commercial capture and supply of threatened European river lamprey (Lampetra fluviatilis) to anglers, to determine the extent of exploitation and seek opportunities for improved conservation. The trade began in 1995 from England, but by 2012 involved sale of lamprey from England, The Netherlands and Estonia, including from protected populations. Lamprey are sold frozen for the capture of predatory fish, mostly in freshwater. In the year 2011/2012 9 tonnes (>90,000 lampreys) of river lamprey were supplied, almost exclusively to British anglers. Although annual catches in the main English lamprey fishery (River Ouse) have varied widely since 1995, catch per unit effort did not decline between 2000 and 2012. Conservation actions since 2011 have included a cap on fishing licenses, catch quotas and restricted fishing seasons. Now, 86% of lamprey bait is imported to Britain. Most bait sellers interviewed would not stock lamprey if they knew they were from threatened populations; many felt their trade would not be impacted if lamprey were not stocked. This facilitates opportunities to enter into dialogue with anglers over alternative baits to threatened lamprey. The study emphasises the need to inform stakeholders about conservation species subjected to market-driven exploitation
Constructing Policy Success for UK Energy Feedback
Energy feedback tools are commonly used to promote energy saving. In the UK, energy feedback provision (currently via an In-Home Display) is part of the government-mandated roll-out of smart meters to all homes by 2020. A core assumption underlying this widespread provision is that information, or evidence, can lead to positive changes in action. This is analogous to assumptions underlying the notion of ‘Evidence-Based Policy’, which raises questions about how users, researchers and policymakers go about using evidence in aiming for a ‘successful’ outcome. In addition the ‘policy feedback’ research agenda has asked how policies alter the landscapes within which they operate by, for example, affecting relationships between actors. Via an in-depth review of DECC (now BEIS) policy literature over 2010-2016, the UK smart meter roll-out was analysed in terms of how its energy feedback focussed measures may be deemed as ‘successful’. Findings include that direct energy savings played a smaller role than might be expected, and translation from one success measure to another was repeatedly observed. A key conclusion is that acting on feedback requires an assessment of success, but such assessment is highly contextual, for consumers and policymakers alike. Generalized Buneman pruning for inferring the most parsimonious multi-state phylogeny
Accurate reconstruction of phylogenies remains a key challenge in
evolutionary biology. Most biologically plausible formulations of the problem
are formally NP-hard, with no known efficient solution. The standard in
practice are fast heuristic methods that are empirically known to work very
well in general, but can yield results arbitrarily far from optimal. Practical
exact methods, which yield exponential worst-case running times but generally
much better times in practice, provide an important alternative. We report
progress in this direction by introducing a provably optimal method for the
weighted multi-state maximum parsimony phylogeny problem. The method is based
on generalizing the notion of the Buneman graph, a construction key to
efficient exact methods for binary sequences, so as to apply to sequences with
arbitrary finite numbers of states with arbitrary state transition weights. We
implement an integer linear programming (ILP) method for the multi-state
problem using this generalized Buneman graph and demonstrate that the resulting
method is able to solve data sets that are intractable by prior exact methods
in run times comparable with popular heuristics. Our work provides the first
method for provably optimal maximum parsimony phylogeny inference that is
practical for multi-state data sets of more than a few characters.Comment: 15 page
At what age do normal weight Canadian children become overweight adults? Differences according to sex and metric
Background: The prevalence of overweight and obesity doubles between adolescence and young adulthood. However, the exact age, and appropriate metric to use, to identify when overweight develops is still debated. Aim: To examine the age of onset of overweight by sex and four metrics: body mass index (BMI), fat mass (%FM), waist circumference (WC) and waist-to-height ratio (WHtR). Methods: Between 1991 and 2017, serial measures of body composition, were taken on 237 (108 males) individuals (aged 8 to 40 years of age). Hierarchical random effects models were used to develop growth curves. Curves were compared to BMI, %FM and WC overweight age and sex-specific cut-points. Results: In males the BMI growth curve crossed the cut-point at 22.0 years compared to 23.5 and 26.5 years for WHtR and %FM respectively; WC cut-off were not reached until 36 years. In females the BMI growth curve, crossed the overweight cut-point at 21.5 years compared to 14.2 years for %FM and at 21.9 and 27.5 years for WC and WHtR respectively. Investigation of Biofuel as a Potential Renewable Energy Source
An accelerating global energy demand, paired with the harmful environmental effects of fossil fuels, has triggered the search for alternative, renewable energy sources. Biofuels are arguably a potential renewable energy source in the transportation industry as they can be used within current infrastructures and require less technological advances than other renewable alternatives, such as electric vehicles and nuclear power. The literature suggests biofuels can negatively impact food security and production; however, this is dependent on the type of feedstock used in biofuel production. Advanced biofuels, derived from inedible biomass, are heavily favoured but require further research and development to reach their full commercial potential. Replacing fossil fuels by biofuels can substantially reduce particulate matter (PM), carbon monoxide (CO) emissions, but simultaneously increase emissions of nitrogen oxides (NOx), acetaldehyde (CH3CHO) and peroxyacetyl nitrate (PAN), resulting in debates concerning the way biofuels should be implemented. The potential biofuel blends (FT-SPK, HEFA-SPK, ATJ-SPK and HFS-SIP) and their use as an alternative to kerosene-type fuels in the aviation industry have also been assessed. Although these fuels are currently more costly than conventional aviation fuels, possible reduction in production costs has been reported as a potential solution. A preliminary study shows that i-butanol emissions (1.8 Tg/year) as a biofuel can increase ozone levels by up to 6% in the upper troposphere, highlighting a potential climate impact. However, a larger number of studies will be needed to assess the practicalities and associated cost of using the biofuel in existing vehicles, particularly in terms of identifying any modifications to existing engine infrastructure, the impact of biofuel emissions, and their chemistry on the climate and human health, to fully determine their suitability as a potential renewable energy source

Membrane Hsp70 — a novel target for the isolation of circulating tumor cells after epithelial-to-mesenchymal transition
The presence of circulating tumor cells (CTCs) in the peripheral blood is a pre-requisite for progression, invasion, and metastatic spread of cancer. Consequently, the enumeration and molecular characterization of CTCs from the peripheral blood of patients with solid tumors before, during and after treatment serves as a valuable tool for categorizing disease, evaluating prognosis and for predicting and monitoring therapeutic responsiveness. Many of the techniques for isolating CTCs are based on the expression of epithelial cell surface adhesion molecule (EpCAM, CD326) on tumor cells. However, the transition of adherent epithelial cells to migratory mesenchymal cells (epithelial-to-mesenchymal transition, EMT)—an essential element of the metastatic process—is frequently associated with a loss of expression of epithelial cell markers, including EpCAM. A highly relevant proportion of mesenchymal CTCs cannot therefore be isolated using techniques that are based on the “capture” of cells expressing EpCAM. Herein, we provide evidence that a monoclonal antibody (mAb) directed against a membrane-bound form of Hsp70 (mHsp70)—cmHsp70.1—can be used for the isolation of viable CTCs from peripheral blood of tumor patients of different entities in a more quantitative manner. In contrast to EpCAM, the expression of mHsp70 remains stably upregulated on migratory, mesenchymal CTCs, metastases and cells that have been triggered to undergo EMT. Therefore, we propose that approaches for isolating CTCs based on the capture of cells that express mHsp70 using the cmHsp70.1 mAb are superior to those based on EpCAM expression

Immune-phenotyping and transcriptomic profiling of peripheral blood mononuclear cells from patients with breast cancer: identification of a 3 gene signature which predicts relapse of triple negative breast cancer
Background: Interactions between the immune system and tumors are highly reciprocal in nature, leading to speculation that tumor recurrence or therapeutic resistance could be influenced or predicted by immune events that manifest locally, but can be detected systemically.
Methods: Multi-parameter flow cytometry was used to examine the percentage and phenotype of natural killer (NK) cells, myeloid-derived suppressor cells (MDSCs), monocyte subsets and regulatory T (Treg) cells in the peripheral blood of of 85 patients with breast cancer (50 of whom were assessed before and after one cycle of anthracycline-based chemotherapy), and 23 controls. Transcriptomic profiles of peripheral blood mononuclear cells (PBMCs) in 23 patients were generated using a NanoString gene profiling platform.
Results: An increased percentage of immunosuppressive cells such as granulocytic MDSCs, intermediate CD14++CD16+ monocytes and CD127negCD25highFoxP3+ Treg cells was observed in patients with breast cancer, especially patients with stage 3 and 4 disease, regardless of ER status. Following neoadjuvant chemotherapy, B cell numbers decreased significantly, whereas monocyte numbers increased. Although chemotherapy had no effect on the percentage of Treg, MDSC and NK cells, the expression of inhibitory receptors CD85j, LIAR and NKG2A and activating receptors NKp30 and NKp44 on NK cells increased, concomitant with a decreased expression of NKp46 and DNAM-1 activating receptors. Transcriptomic profiling revealed a distinct group of 3 patients in the triple negative breast cancer (TNBC) cohort who expressed high levels of mRNA encoding genes predominantly involved in inflammation. The analysis of a large transcriptomic dataset derived from the tumors of patients with TNBC revealed that the expression of CD163, CXCR4, THBS1 predicted relapse-free survival.
Conclusions: The peripheral blood immunome of patients with breast cancer is influenced by the presence and stage of cancer, but not by molecular subtypes. Furthermore, immune profiling coupled with transcriptomic analyses of peripheral blood cells may identify patients with TNBC that are at risk of relapse after chemotherapy
Impression creep testing for evaluation of grade 22 ex-service hot reheat piping seam weld
U.S. electric power production is significantly dependent on the operation of coal-fired steam generation units and a large majority of these units are reaching ages over 50+ years with concerns for operating component integrity and remaining life. This paper discusses a small sample testing technique (impression creep) that was used to estimate the remaining life of a hot reheat seam welded piping system that saw about 322,000 hours of operation at nominally 4170 kPa (605psig) and 538°C (1000°F) steam conditions. Two different life assessments using experimental impression creep data are discussed and findings compared to a previous preliminary study of the same piping system using operational data, reported measured piping thickness values (from UT measurements), and published creep rupture data. Impression creep tests were conducted in unaffected base metal, weld metal and the heat-affected zone. Impression creep rates of the various zones showed no creep mismatch. Minimal creep mismatch, proper design, fabrication and operation, combined with proper metallurgy have successfully demonstrated that even low-alloy seam welds can operate 300,000+ hours and still exhibit useful remaining life
Acute effects of nicotine on visual search tasks in young adult smokers
Rationale Nicotine is known to improve performance on tests involving sustained attention and recent research suggests that nicotine may also improve performance on tests involving the strategic allocation of attention and working memory. Objectives We used measures of accuracy and response latency combined with eye-tracking techniques to examine the effects of nicotine on visual search tasks. Methods In experiment 1 smokers and non-smokers performed pop-out and serial search tasks. In experiment 2, we used a within-subject design and a more demanding search task for multiple targets. In both studies, 2-h abstinent smokers were asked to smoke one of their own cigarettes between baseline and tests. Results In experiment 1, pop-out search times were faster after nicotine, without a loss in accuracy. Similar effects were observed for serial searches, but these were significant only at a trend level. In experiment 2, nicotine facilitated a strategic change in eye movements resulting in a higher proportion of fixations on target letters. If the cigarette was smoked on the first trial (when the task was novel), nicotine additionally reduced the total number of fixations and refixations on all letters in the display. Conclusions Nicotine improves visual search performance by speeding up search time and enabling a better focus of attention on task relevant items. This appears to reflect more efficient inhibition of eye movements towards task irrelevant stimuli, and better active maintenance of task goals. When the task is novel, and therefore more difficult, nicotine lessens the need to refixate previously seen letters, suggesting an improvement in working memory
