    From Nelson Rockefeller to Eric Adams: The Evolving Politics of Crime and Punishment in New York

    Despite calls for the "defunding" of the police and the reimagining of policing following the death of George Floyd in 2020, many New York politicians, in response to rising rates of violent crime, have begun to embrace "law and order."  All of this bears a great similarity to the politics of crime and punishment during the governorship of Nelson Rockefeller.  Examining several documents in the gubernatorial records of Nelson Rockefeller at the Rockefeller Archive Center, newspaper articles, and public opinion, this report documents the political response to violence and drug addiction in the 1960s and 1970s and compares it to the present, reviewing contrasting arguments of influential Black leaders and "white liberals." It concludes that the present crime context, much like the one during the Rockefeller-era, has divided the left and Black leadership while solidifying Republican commitment to "law and order." It argues that the history of the Rockefeller drug laws illustrates that these divisions and the legitimate fears of working- and middle-class minorities can produce haphazard policies that harm rather than save these communities

    Cardiovascular and metabolic demands of the kettlebell swing using a Tabata interval versus a traditional resistance protocol

    International Journal of Exercise Science 7(3) : 179-185, 2014. Tabata (TAB) training, consisting of eight cycles of 20 seconds of maximal exercise followed by 10 seconds of rest, is time-efficient, with aerobic and anaerobic benefit. This study investigated the cardiovascular and metabolic demands of a TAB versus traditional (TRAD) resistance protocol with the kettlebell swing. Fourteen young (18-25y), non-obese (BMI 25.7±0.8 kg/m2) participants reported on three occasions. All testing incorporated measurements of HR, oxygen consumption, and blood lactate accumulation. Each participant completed Tabata kettlebell swings (male- 8kg, female- 4.5kg; 8 intervals; 20s maximal repetitions, 10s rest). On a subsequent visit (TRAD), the total swings from the TAB protocol were evenly divided into 4 sets, with 90s rest between sets. Outcome measures were compared using paired t-tests. The TAB was completed more quickly than the TRAD protocol (240.0±0.0 v. 521.5±3.3 sec, P\u3c0.01), at a higher perceived exertion (Borg RPE; 15.1±0.7 v. 11.7±0.9, P\u3c0.01). The TAB elicited a higher average VO2 value (33.1±1.5 v. 27.2±1.6 ml/kg/min, P\u3c0.01), percent of VO2peak achieved (71.0±0.3 v. 58.4±0.3%, P\u3c0.01), maximal HR (162.4±4.6 v. 145.6±4.8 bpm, P\u3c0.01), and post-exercise blood lactate concentration (6.4±1.1 v. 3.7±0.5 mmol/L, P\u3c0.01). Conclusion: The kettlebell swing demonstrated significantly greater cardiovascular and metabolic responses within a TAB vs. TRAD framework. Appropriate screening and risk stratification are advised before implementing kettlebell swings

    Measuring the Grafting Density of Nanoparticles in Solution by Analytical Ultracentrifugation and Total Organic Carbon Analysis

    Many of the solution phase properties of nanoparticles, such as their colloidal stability and hydrodynamic diameter, are governed by the number of stabilizing groups bound to the particle surface (i.e., grafting density). Here, we show how two techniques, analytical ultracentrifugation (AUC) and total organic carbon analysis (TOC), can be applied separately to the measurement of this parameter. AUC directly measures the density of nanoparticle–polymer conjugates while TOC provides the total carbon content of its aqueous dispersions. When these techniques are applied to model gold nanoparticles capped with thiolated poly(ethylene glycol), the measured grafting densities across a range of polymer chain lengths, polymer concentrations, and nanoparticle diameters agree to within 20%. Moreover, the measured grafting densities correlate well with the polymer content determined by thermogravimetric analysis of solid conjugate samples. Using these tools, we examine the particle core diameter, polymer chain length, and polymer solution concentration dependence of nanoparticle grafting densities in a gold nanoparticle–poly(ethylene glycol) conjugate system

    Glycolysis promotes caspase-3 activation in lipid rafts in T cells.

    Resting T cells undergo a rapid metabolic shift to glycolysis upon activation in the presence of interleukin (IL)-2, in contrast to oxidative mitochondrial respiration with IL-15. Paralleling these different metabolic states are striking differences in susceptibility to restimulation-induced cell death (RICD); glycolytic effector T cells are highly sensitive to RICD, whereas non-glycolytic T cells are resistant. It is unclear whether the metabolic state of a T cell is linked to its susceptibility to RICD. Our findings reveal that IL-2-driven glycolysis promotes caspase-3 activity and increases sensitivity to RICD. Neither caspase-7, caspase-8, nor caspase-9 activity is affected by these metabolic differences. Inhibition of glycolysis with 2-deoxyglucose reduces caspase-3 activity as well as sensitivity to RICD. By contrast, IL-15-driven oxidative phosphorylation actively inhibits caspase-3 activity through its glutathionylation. We further observe active caspase-3 in the lipid rafts of glycolytic but not non-glycolytic T cells, suggesting a proximity-induced model of self-activation. Finally, we observe that effector T cells during influenza infection manifest higher levels of active caspase-3 than naive T cells. Collectively, our findings demonstrate that glycolysis drives caspase-3 activity and susceptibility to cell death in effector T cells independently of upstream caspases. Linking metabolism, caspase-3 activity, and cell death provides an intrinsic mechanism for T cells to limit the duration of effector function

    Targeting mitochondrial oxidative stress with MitoQ reduces NET formation and kidney disease in lupus-prone MRL-lpr mice.

    OBJECTIVES: Recent investigations in humans and mouse models with lupus have revealed evidence of mitochondrial dysfunction and production of mitochondrial reactive oxygen species (mROS) in T cells and neutrophils. This can provoke numerous cellular changes including oxidation of nucleic acids, proteins, lipids and even induction of cell death. We have previously observed that in T cells from patients with lupus, the increased mROS is capable of provoking oligomerisation of mitochondrial antiviral stimulator (MAVS) and production of type I interferon (IFN-I). mROS in SLE neutrophils also promotes the formation of neutrophil extracellular traps (NETs), which are increased in lupus and implicated in renal damage. As a result, in addition to traditional immunosuppression, more comprehensive treatments for lupus may also include non-immune therapy, such as antioxidants. METHODS: Lupus-prone MRL-lpr mice were treated from weaning for 11 weeks with the mitochondria-targeted antioxidant, MitoQ (200 µM) in drinking water. Mice were then assessed for ROS production in neutrophils, NET formation, MAVS oligomerisation, serum IFN-I, autoantibody production and renal function. RESULTS: MitoQ-treated mice manifested reduced neutrophil ROS and NET formation, decreased MAVS oligomerisation and serum IFN-I, and reduced immune complex formation in kidneys, despite no change in serum autoantibody . CONCLUSIONS: These findings reveal the potential utility of targeting mROS in addition to traditional immunosuppressive therapy for lupus

    An Innovative Option for Venous Reconstruction After Pancreaticoduodenectomy: the Left Renal Vein

    INTRODUCTION: Pancreatic ductal adenocarcinoma has a high mortality rate with limited treatment options. One option is pancreaticoduodenectomy, although complete resection may require venous resection. Pancreaticoduodenectomy with venous resection and reconstruction is becoming a more common practice with many choices for venous reconstruction. We describe the technique of using the left renal vein as a conduit for venous reconstruction during pancreaticoduodenectomy. METHODS: The technique for use of the left renal vein as an interposition graft for venous reconstruction during pancreaticoduodenectomy is described as well as outcomes for nine patients that have undergone the procedure. RESULTS: Nine patients, seven men, with a mean age of 57 years, have undergone the operation. There were eight interposition grafts and one patch graft. Mean operating time was 7.8 hours, and mean tumor size was 3.4 cm. Eight patients had node-positive disease, and six had involvement of the vein. Mean hospital stay was 14 days and perioperative morbidity included a superficial wound infection, delayed gastric emptying, ascites, and gastrointestinal bleeding in one patient each. Creatinine ranged from 0.8–1.1 mg/dl preoperatively and from 0.7–1.3 mg/dl at discharge. Mean follow-up was 6.8 months with normal creatinine values noted through the follow-up period. Two patients had died during follow-up from recurrent disease at 8.3 and 18.2 months after the operation. CONCLUSIONS: The left renal vein provides an additional choice for an autologous graft during pancreaticoduodenectomy with venous resection. The ease of harvesting the graft and maintenance of renal function distinguish its use

    Intensity-Frontier Antiproton Physics with The Antiproton Annihilation Spectrometer (TAPAS) at Fermilab

    The Fermilab Antiproton Source is the world's most intense source of antimatter. With the Tevatron program now behind us, this unique facility can help make the case for Fermilab's continued accelerator operations. The Antiproton Source can be used for unique, dedicated antimatter studies, including medium-energy {bar p}-annihilation experiments. We propose to assemble a powerful, yet cost-effective, solenoidal magnetic spectrometer for antiproton-annihilation events, and to use it at the Fermilab Antiproton Accumulator to measure the charm production cross section, study rare hyperon decays, search for hyperon CP asymmetry, precisely measure the properties of several charmonium and nearby states, and make the first measurements of the Drell-Yan continuum in medium-energy antiproton annihilation. Should the charm production cross section be as large as some have proposed, we will also be able to measure D{sup 0}-{bar D}{sup 0} mixing with high precision and discover (or sensitively limit) charm CP violation. The observation of charm or hyperon CP violation would be evidence for physics beyond the Standard Model, with possible implications for the origin of the baryon asymmetry of the universe - the question of what happened to all the antimatter that must have been produced in the Big Bang. The experiment will be carried out by an international collaboration and will require some four years of running time. As possibly the sole hadron experiment in progress at Fermilab during that time, it will play an important role in maintaining a broad particle physics program at Fermilab and in the U.S. It will thus help us to continue attracting creative and capable young people into science and technology, and introducing them to the important technologies of accelerators, detectors, and data acquisition and analysis - key roles in society that accelerator-based particle physics has historically played

    Investigation of Effects of L-Theanine on PTSD-induced Changes in Rat Neurobehavior

    Post-traumatic stress disorder (PTSD) is a devastating neuropsychological disorder that may develop in response to traumatic experiences. Symptoms include anxiety, hypervigilance, memory deficits, and depression. We investigated L-Theanine on neurobehavioral effects in a PTSD rodent model. Evaluation of the effects of L-Theanine as a sole agent and in combination with midazolam on neurobehavior was analyzed using the Elevated Plus-Maze, Morris Water Maze, and Forced Swim Test. Statistical analysis consisted of comparison of PTSD symptoms in PTSD vs. non-stressed groups. Data regarding weight gain between the 40 control (non-stressed) and 40 PTSD (stressed) rats were significantly different (p < 0.001), where the control rats gained an average of 55.4 grams compared to 37.4 grams for the PTSD rats over the 10 post stress days. This research did not show statistical significance with single dose administration of L-Theanine or in combination with midazolam. However, the theoretic framework and Post-Traumatic Stress Disease Induction Model were validated based on this research. This study establishes a solid framework for future investigation of PTSD treatments. Future studies of L-Theanine and other herbal therapies may use an extended dosing period to obtain a steady state for the period of time needed to alter neurobiology
