    Activation of Interferon Regulatory Factor 5 by Site Specific Phosphorylation

    The cellular defense to infection depends on accurate activation of transcription factors and expression of select innate immunity genes. Interferon regulatory factor 5 (IRF5), a risk factor for systemic lupus erythematosus, is activated in response to pathogen recognition receptor engagement and downstream effector molecules. We find the nucleotide-binding oligomerization domain containing protein 2 (NOD2) receptor to be a significant activator of IRF5. Phosphorylation is key to the regulation of IRF5, but the precise phosphorylation sites in IRF5 remained to be identified. We used mass spectrometry to identify for the first time specific residues that are phosphorylated in response to TANK-binding kinase-1 (TBK-1), tumor necrosis factor receptor-associated factor 6 (TRAF6), or receptor interacting protein 2 (RIP2). RIP2, a kinase known to function downstream of NOD2, was the most effective activator of IRF5-regulated gene expression. To determine if the phosphorylated residues are required or sufficient for IRF5 activity, aspartic acid phosphomimetic substitutions or inactivating alanine substitutions were tested. Phosphorylation of carboxyl serines 451 and 462 appear the primary trigger of IRF5 function in nuclear accumulation, transcription, and apoptosis. Results indicate polyubiquitination of IRF5 does not play a major role in its transcriptional activity, and that ubiquitination and phosphorylation are independent modifications

    Associations between alcohol, smoking, and cartilage composition and knee joint morphology: Data from the Osteoarthritis Initiative

    Summary: Objective: To determine the cross-sectional associations of alcohol consumption and smoking history with magnetic resonance imaging (MRI) measures of cartilage composition (T2) and joint structure using data from the Osteoarthritis Initiative (OAI). Design: Subjects with radiographic Kellgren Lawrence right knee grades 0–2 were selected from the OAI database, and those with previously analyzed MRI cartilage T2 and semi-quantitative joint morphology gradings (WORMS) were included (n ​= ​2061). Alcohol consumption was categorized as: no drinks to 7 drinks/week. Smoking history was categorized as none, current, or former. Linear regression was used to assess the relationships of alcohol consumption and smoking history with both WORMS scores and cartilage T2. Results: Subjects who consumed >7 drinks/week had significantly higher cartilage T2 than subjects who consumed 7 drinks/week was associated with elevated cartilage T2. Compared to non-smokers, current smokers had a more degenerated cartilage matrix as evidenced by greater cartilage T2

    Seasonal Variability of the Labrador Current and Shelf Circulation off Newfoundland

    Three-dimensional finite element models were established for the Newfoundland and Labrador Shelf to investigate climatological monthly mean wind- and density-driven circulation. The model was forced using wind stresses from the National Center for Environmental Prediction-National Center for Atmospheric Research reanalysis data prescribed at the sea surface, large-scale remote forcing determined from a North Atlantic model, monthly mean temperature and salinity climatology, and M2 tide on the open boundary. The model results were examined against various in situ observations (moored current meter, tide gauge, and vessel-mounted acoustic Doppler current profiler data) and satellite drift measurements and discussed together with literature information. The seasonal mean circulation solutions were investigated in terms of relative importance of wind to density forcing for the Labrador Current. The model results indicate significant seasonal and spatial variations, consistent generally with previous study results and in approximate agreement with observations for the major currents. The region is dominated by the equatorward flowing Labrador Current along the shelf edge and along the Labrador and Newfoundland coasts. The Labrador Current is strong in the fall/winter and weak in the spring/summer. The mean transport of the shelf edge Labrador Current is 7.5 Sv at the Seat Island transect and 5.5 Sv through the Flemish Pass. The seasonal ranges are 4.5 and 5.2 Sv at the two sections, respectively. Density- and wind-driven components are both important in the inshore Labrador Current. The density-driven component dominates the mean component of the shelf edge Labrador Current while the large-scale wind-forcing contributes significantly to its seasonal variability

The unique vulnerabilities for patients with mental illnesses such as depression, mania, anxiety, schizophrenia and/or substance use disorders* heighten the need for coordinated transitions and aftercare. In 2010, depression was the fourth diagnosis by volume for readmissions in Minnesota according to the Potentially Preventable Readmissions data collected by the Minnesota Hospital Association

    Clinical, radiologic, pathologic, and molecular characteristics of long-term survivors of diffuse intrinsic pontine glioma (DIPG): a collaborative report from the International and European Society for Pediatric Oncology DIPG registries

    Purpose Diffuse intrinsic pontine glioma (DIPG) is a brainstem malignancy with a median survival of < 1 year. The International and European Society for Pediatric Oncology DIPG Registries collaborated to compare clinical, radiologic, and histomolecular characteristics between short-term survivors (STSs) and long-term survivors (LTSs). Materials and Methods Data abstracted from registry databases included patients from North America, Australia, Germany, Austria, Switzerland, the Netherlands, Italy, France, the United Kingdom, and Croatia. Results Among 1,130 pediatric and young adults with radiographically confirmed DIPG, 122 (11%) were excluded. Of the 1,008 remaining patients, 101 (10%) were LTSs (survival ≥ 2 years). Median survival time was 11 months (interquartile range, 7.5 to 16 months), and 1-, 2-, 3-, 4-, and 5-year survival rates were 42.3% (95% CI, 38.1% to 44.1%), 9.6% (95% CI, 7.8% to 11.3%), 4.3% (95% CI, 3.2% to 5.8%), 3.2% (95% CI, 2.4% to 4.6%), and 2.2% (95% CI, 1.4% to 3.4%), respectively. LTSs, compared with STSs, more commonly presented at age < 3 or > 10 years (11% v 3% and 33% v 23%, respectively; P < .001) and with longer symptom duration ( P < .001). STSs, compared with LTSs, more commonly presented with cranial nerve palsy (83% v 73%, respectively; P = .008), ring enhancement (38% v 23%, respectively; P = .007), necrosis (42% v 26%, respectively; P = .009), and extrapontine extension (92% v 86%, respectively; P = .04). LTSs more commonly received systemic therapy at diagnosis (88% v 75% for STSs; P = .005). Biopsies and autopsies were performed in 299 patients (30%) and 77 patients (10%), respectively; 181 tumors (48%) were molecularly characterized. LTSs were more likely to harbor a HIST1H3B mutation (odds ratio, 1.28; 95% CI, 1.1 to 1.5; P = .002). Conclusion We report clinical, radiologic, and molecular factors that correlate with survival in children and young adults with DIPG, which are important for risk stratification in future clinical trials

    The focus of National Student Employment Association (formerly the National Association of Student Employment Administrators, or NASEA) publications has always been on students in transition. From the freshman moving from high school to higher education, to the senior attempting the transition to professional employment and financial independence, we always have explored how students can better accomplish these linking experiences. Student employment is a hybrid, serving as a bridge between work and school, and ultimately, a link between school and full-time work. Student employment links elements of financial aid, career development, academic learning, experiential education, and personal development. Student employment, in all of these ways, is a bridge, moving the student from point A to point B. Because of this variety, any publication on student employment must necessarily speak to diverse themes.
