    Hongos acuáticos de los estuarios en Puerto Rico: Boca del Río Manatí

    Aquatic fungi were isolated from sea foam, leaf litter, beach sand, and driftwood in an estuary known as "La Boca" (river mouth) of the Manatí River in Barceloneta, northern Puerto Rico. Observations of the baits (sterilized balsa wood) and incubated organic debris, revealed the presence of 28 species of aquatic fungi, 13 of which belong to the ascomycetes and 15 to the mitosporic fungi. The species Arenariomyces triseptatus, Corollospora cf. colossa, C. filiformis, Halosphaeria sp., Kirschsteiniothelia sp., Astrosphaeriella aff. mangrovei, Torpedospora radiata (Ascomycota), Brachiosphaera tropicalis, Campylospora sp., and Clavatospora bulbosa (Mitosporic fungi) were the most common in the samples. Eleven species are new records for Puerto Rico; six fungal isolates could not be identified. Se aislaron hongos acuáticos de la espuma de mar, hojarasca, arena playera y madera a la deriva de un estuario conocido como "La Boca" (boca de río) del Río Manatí en Barceloneta, al norte de Puerto Rico. Las observaciones de los cebos (madera de balsa esterilizada) y material orgánico incubado, revelaron la presencia de 28 especies de hongos acuáticos, de los cuales 13 pertenecen a los ascomicetos y 15 a los hongos mitospóricos. Las especies Arenariomyces triseptatus, Corollospora cf. colossa, C. filiformis, Halosphaeria sp., Kirschsteiniothelia sp., Astrosphaeriella aff. mangrovei, Torpedospora radiata (Ascomycota), Brachiosphaera tropicalis, Campylospora sp. y Clavatospora bulbosa (Hongos Mitospóricos) fueron las más comunes en las muestras.     Caracterización química y cuantificación del rendimiento de extracción de pigmento en siete accesiones mexicanas de Bixa orellana

    Achiote (Bixa orellana) is a plant used for obtaining a natural dye rich on carotenoids (mainly bixin and norbixin); it is also the plant species with the highest content of tocotrienols in nature. In the present work, the pigment extraction yield of seven Mexican accessions of Bixa orellana was quantified. Also color parameters and content of tocotrienols, tocopherols, norbixin, bixin, total phenolic compounds and antioxidant capacity were evaluated in the corresponding annatto extracts. The highest percentage of pigment extraction yield was obtained with KOH (4.84%). Accessions 43 (L*= 4.01 ± 0.79, C*= 7.33 ± 1.07, h= 25.76 ± 6.35) and 50 (L*= 3.17 ± 0.64, C*= 6.81 ± 0.53, h= 26.41 ± 4.41) had the lowest color values, meaning these accessions had a darker and redder color. Four accessions showed the highest content of bixin: accession 48 (3.1%), 45 (2.6%) 43 (2.4%) and 47 (2.2%). Accession 50 had showed the highest content of total phenolic compounds and of tocotrienols (T3), mainly the isoform δ-T3 (5.03 ± 0.64 mg g−1 Seed Dry Weight), as well as the highest antioxidant capacity.El achiote (Bixa orellana) es una planta utilizada para obtener un colorante natural rico en carotenoides (principalmente bixina y norbixina); además, es la especie vegetal con el mayor contenido de tocotrienoles. En este trabajo, se determinó el rendimiento de extracción de pigmento de siete accesiones mexicanas de Bixa orellana. También se evaluaron los parámetros de color y el contenido de tocotrienoles, tocoferoles, norbixina, bixina, compuestos fenólicos totales y la capacidad antioxidante en extractos de annato. El mayor porcentaje de rendimiento de extracción de pigmento fue obtenido con KOH (4.847905%). Las accesiones 43 (L*= 4.01 ± 0.79, C*= 7.33 ± 1.07, h= 25.76 ± 6.35) y 50 (L*= 3.17 ± 0.64, C*= 6.81 ± 0.53, h= 26.41 ± 4.41) presentaron los valores más bajos de los párametros de color, lo que significa que estas accesiones tuvieron un color más oscuro y más rojo. Cuatro accesiones mostraron el mayor contenido de bixina: accesión 48 (3.1%), 45 (2.6%) 43 (2.4%) and 47 (2.2%).     Infrared luminescence and application of a vibronic-coupling Hamiltonian to the level structure of CdTe:Fe<sup>2+</sup>

    Samples of crystalline CdTe doped with two different concentrations of iron were prepared by the vertical high-pressure Bridgman method. Absorption and emission spectra were recorded at liquid-helium temperature in the region of the 5T2(D)? 5E(D) infrared transitions of substitutional Fe2+(d6) ions. Especially in the range between 2200 and 2300 cm−1, a rich structure is resolved comprising more lines than predicted from plain crystal-field theory. The explanation of all the important lines is found after introducing a vibronic Jahn-Teller term to the Hamiltonian. A linear coupling between the double-degenerate vibrational mode ε (or γ3) to the electronic orbitals of the atomic multiplet of symmetry 5D leads to the diagonalization of the total Hamiltonian in a set of vibronic functions. Just one free parameter is used in the adjustment: the so-called Jahn-Teller energy representing the strength of the coupling. The corresponding value that we report here is 3 cm−1. The energies thus found are in good agreement with the positions of the observed lines in the spectra. With the final wave functions we can calculate the relative intensities of the most important transitions and approximate theoretical line shape. This is also in good agreement with the experiment. Using these same energies and wave functions a calculation was performed to explain data existing in the literature about far-infrared absorption for the system CdTe:Fe2+. Again, good agreement between experiment and theory is found

    The Effect of Endogenous Expression of HIV-1 gp120 on Glutamate Metabolism in Human Astrocytes

    Human immunodeficiency virus (HIV) infection is a global epidemic that targets the immune system. HIV infects white blood cells and spreads throughout the entire body via blood stream and makes its way to the brain. HIV infection in the brain may lead to HIV associated neurocognitive disorders (HAND). To be able to address this problem, we have to better understand how HIV infection damages neurons. We hypothesize that gp120 causes neurotoxicity in the cells by inhibiting the conversion of glutamate to glutamine by glutaminase. As a result, glutamate concentrations will build up both inside and outside the cell causing excitatory neurotoxicity. To better understand this process, we transfected human astrocytes (U87MG cells) with mock (control), an empty vector (control), and with gp120 plasmid. Seventy-two hours post transfection, the cells were collected and run through a series of tests including SDS-PAGE/Western Blot and qRT-PCR to assess protein and mRNA levels of glutaminase and gp120. We expect production of gp120 by astrocytes to lead to a decrease in expression of glutaminase. This would inhibit the process of converting glutamate to glutamine and explain how excess of glutamate accumulates inside and outside of the cell causing neurotoxicity and cell death. In conclusion, we expect to find a direct relationship between gp120 and the glutamate metabolism in human astrocytes. Understanding the effect gp120 has on neurons will help develop more effective treatments to better fight the virus

    Numerical and Experimental Analysis of Sound Suppressor for a 5.56 mm Calibre

    A sound suppressor is an internal or external device coupled to the barrel of a firearm. Its development has been historically related to the negative effects produced by the noise. This article presents the numerical and experimental analysis of a sound suppressor for a 5.56 mm caliber rifle. It was designed, manufactured, and tested inside a shooting tunnel for 911 m/s and 344 m/s velocities. Three geometric configurations with curved deflectors, conical deflectors, and finally with a reactive spiral capable of dissipating the acoustic wave were compared considering reactive and dissipative systems. The attenuation of the sound inside the silencer depends directly on the reduction of the projectile wave velocity and the deflagration of the gases at the instant of firing. Then the MIL-STD-1474E standard was used to carry out the experiments. The results in the computational numerical simulation show an average value of 143 dB for the considered three models, the Sound Pressure Level in the reactive core model decreased by 25% with respect to other proposals, which have an average value of 141 dB. These results can be useful to improve in the design of sound suppressors based on the needs of the users and under the specific characteristics of each weapon ballistic.&nbsp

    Large Charge Moment Change Lightning in an Oklahoma Mesoscale Convective System

    On 31 May 2013, a line of severe thunderstorms developed during the local afternoon in central Oklahoma, USA. One of the supercells produced the El Reno tornado, which caused significant damage and killed several people. During the 2300 UTC hour (during the mature supercell stage and just after the tornado began), the storm produced several positive cloud-to-ground (+CG) lightning strokes that featured large (> 100 C km) impulse charge moment changes (iCMCs; charge moment during the first 2 ms after the return stroke). These discharges occurred mainly in convection, in contrast to the typical pattern of large-CMC and sprite-parent +CGs occurring mainly in stratiform precipitation regions. After this time, the line of thunderstorms evolved over several hours into a large mesoscale convective system (MCS). By the 0700 UTC hour on 1 June 2013, the large-CMC pattern had changed markedly. Large-CMC negative CGs, which were absent early in the storm's lifetime, occurred frequently within convection. Meanwhile, large-CMC +CGs had switched to occurring mainly within the broad stratiform region that had developed during the intervening period. The evolution of the large-CMC lightning in this case will be examined using a mix of national mosaics of radar reflectivity, the Oklahoma Lightning Mapping Array (OKLMA), the Charge Moment Change Network (CMCN), and the National Lightning Detection Network (NLDN). A major goal of this study is understanding how storm structure and evolution affected the production of large-CMC lightning. It is anticipated that this will lead to further insight into how and why storms produce the powerful lightning that commonly causes sprites in the upper atmosphere
