    Freezing for control of stored-product psocids

    A series of studies was conducted by exposing young and old eggs, nymphs, and adults of the psocids Liposcelis bostrychophila (Badonnel), L. paeta (Pearman), L. decolor (Pearman), and L. entomophila (Enderlein) to -18°C for various time intervals. Survival was assessed as initial and final, at different times depending on the life stage. Young eggs of L. bostrychophila were the most tolerant life stage of any of the species, with scattered survival out to 120 h of exposure to -18°C. Eggs were the most tolerant life stage for each species, requiring 24, 12, and 2 h of exposure for complete kill of L. paeta, L. decolor, and L. entomophila, respectively. Nymphs and adults of all species were far more susceptible than eggs, with no final survival after two hours of exposure. Results show the extreme variation between different psocid life stages and species to cold temperatures, and provide guidelines for using cold as a control strategy for psocids. Our results show that 24 h at -18°C is sufficient to kill all life stages of the psocid species tested, except for young L. bostrychophila eggs which will require at least 128 h of exposure at -18°C for complete mortality

    Identification of hydroxyapatite spherules provides new insight into subretinal pigment epithelial deposit formation in the aging eye.

    Accumulation of protein- and lipid-containing deposits external to the retinal pigment epithelium (RPE) is common in the aging eye, and has long been viewed as the hallmark of age-related macular degeneration (AMD). The cause for the accumulation and retention of molecules in the sub-RPE space, however, remains an enigma. Here, we present fluorescence microscopy and X-ray diffraction evidence for the formation of small (0.5-20 μm in diameter), hollow, hydroxyapatite (HAP) spherules in Bruch's membrane in human eyes. These spherules are distinct in form, placement, and staining from the well-known calcification of the elastin layer of the aging Bruch's membrane. Secondary ion mass spectrometry (SIMS) imaging confirmed the presence of calcium phosphate in the spherules and identified cholesterol enrichment in their core. Using HAP-selective fluorescent dyes, we show that all types of sub-RPE deposits in the macula, as well as in the periphery, contain numerous HAP spherules. Immunohistochemical labeling for proteins characteristic of sub-RPE deposits, such as complement factor H, vitronectin, and amyloid beta, revealed that HAP spherules were coated with these proteins. HAP spherules were also found outside the sub-RPE deposits, ready to bind proteins at the RPE/choroid interface. Based on these results, we propose a novel mechanism for the growth, and possibly even the formation, of sub-RPE deposits, namely, that the deposit growth and formation begin with the deposition of insoluble HAP shells around naturally occurring, cholesterol-containing extracellular lipid droplets at the RPE/choroid interface; proteins and lipids then attach to these shells, initiating or supporting the growth of sub-RPE deposits

    Cold temperature disinfestation of bagged flour

    We conducted studies using a commercial freezer maintained at -17.8°C to determine the time needed to kill Tribolium castaneum eggs in a pallet of flour. Each bag weighed 22.7 kg, and there were 5 bags in each of 10 layers. The dimensions of the pallet were 109-cm wide by 132-cm long by 123-cm tall, and the weight of the stacked pallet was approximately 1152 kg. We conducted tests for nine internal goal temperatures of -12, -10, -8, -6, -4, -2, 0, 4 and 8°C. Internal temperatures in the most central location of the flour pallet reached: -11.0, -9.4, -6.9, -5.0, -3.5, -1.6, -0.1, 3.3, and 5.6°C and were achieved after 11.0, 9.1, 8.9, 7.2, 6.7, 5.8, 5.5, 5.2, and 4.2 days, respectively. For treatments where the goal temperature for the center bag ranged from -12 to 4°C, egg mortality was 100% in bags located in both the periphery and in the center of the pallet. When the temperature goal for the center bag was 8°C, 7 ± 2.5% of the eggs survived in bags located near the center of the pallet. Our data showed that temperatures that follow the dynamic temperature curve that takes place over 24.2 days (cool down and warm up for the 0°C temperature goal) resulted in 100% mortality of T. castaneum eggs. The reason for the difference in mortality for a static compared to a dynamic temperature treatment may be due to the fact that the dynamic temperature treatment occurs over a much longer duration. The fact that the treatment only required 5.5 days in the freezer before it could be shipped makes it a practical method to disinfest pallets of flour, especially because the bags do not need to be removed from the pallet and no chemicals are used

    A Novel hAPP/htau Mouse Model of Alzheimer's Disease: Inclusion of APP With Tau Exacerbates Behavioral Deficits and Zinc Administration Heightens Tangle Pathology

    The brains of those with Alzheimer's disease have amyloid and tau pathology; thus, mice modeling AD should have both markers. In this study, we characterize offspring from the cross of the J20 (hAPP) and rTg4510 (htau) strains (referred to as dual Tg). Behavior was assessed at both 3.5 and 7 months, and biochemical differences were assessed at 8 months. Additionally, mice were placed on zinc (Zn) water or standard lab water in order to determine the role of this essential biometal. Behavioral measures examined cognition, emotion, and aspects of daily living. Transgenic mice (dual Tg and htau) showed significant deficits in spatial memory in the Barnes Maze at both 3.5 and 7 months compared to controls. At 7 months, dual Tg mice performed significantly worse than htau mice (p < 0.01). Open field and elevated zero maze (EZM) data indicated that dual Tg and htau mice displayed behavioral disinhibition compared to control mice at both 3.5 and 7 months (p < 0.001). Transgenic mice showed significant deficits in activities of daily living, including burrowing and nesting, at both 3.5 and 7 months compared to control mice (p < 0.01). Dual Tg mice built very poor nests, indicating that non-cognitive tasks are also impacted by AD. Overall, dual Tg mice demonstrated behavioral deficits earlier than those shown by the htau mice. In the brain, dual Tg mice had significantly less free Zn compared to control mice in both the dentate gyrus and the CA3 of the hippocampus (p < 0.01). Dual Tg mice had increased tangles and plaques in the hippocampus compared to htau mice and the dual Tg mice given Zn water displayed increased tangle pathology in the hippocampus compared to htau mice on Zn water (p < 0.05). The dual Tg mouse described here displays pathology reminiscent of the human AD condition and is impaired early on in both cognitive and non-cognitive behaviors. This new mouse model allows researchers to assess how both amyloid and tau in combination impact behavior and brain pathology

    Real-world clinical experience in the Connect® chronic lymphocytic leukaemia registry: a prospective cohort study of 1494 patients across 199 US centres.

    The clinical course of chronic lymphocytic leukaemia (CLL) is heterogeneous, and treatment options vary considerably. The Connect® CLL registry is a multicentre, prospective observational cohort study that provides a real-world perspective on the management of, and outcomes for, patients with CLL. Between 2010 and 2014, 1494 patients with CLL and that initiated therapy, were enrolled from 199 centres throughout the USA (179 community-, 17 academic-, and 3 government-based centres). Patients were grouped by line of therapy at enrolment (LOT). We describe the clinical and demographic characteristics of, and practice patterns for, patients with CLL enrolled in this treatment registry, providing patient-level observational data that represent real-world experiences in the USA. Fluorescence in situ hybridization (FISH) analyses were performed on 49·3% of patients at enrolment. The most common genetic abnormalities detected by FISH were del(13q) and trisomy 12 (45·7% and 20·8%, respectively). Differences in disease characteristics and comorbidities were observed between patients enrolled in LOT1 and combined LOT2/≥3 cohorts. Important trends observed include the infrequent use of genetic prognostic testing, and differences in patient characteristics for patients receiving chemoimmunotherapy combinations. These data represent experiences of patients with CLL in the USA, which may inform treatment decisions in everyday practice

    The influence of the physical environment on self-recovery after disasters in Nepal and the Philippines

    Following a disaster, the majority of families rebuild their homes themselves. In this paper, we consider how the physical environment influences such ‘self-recovery’ by investigating disasters in the Philippines (typhoons Haiyan in 2013 and Haima in 2016) and Nepal (the Gorkha earthquake - 2015). Despite the many differences in the disaster contexts, there are some common barriers to self-recovery (and building back better) in a substantially changed and dynamic multi-hazard, post-disaster environment. These are related to changes in water supply (shortage or surplus), impacts of post-disaster geohazard events on infrastructure (particularly affecting transport) and the availability of technical advice. People face a broad spectrum of challenges as they recover and tackling these ‘geo-barriers’ may help to create a more enabling environment for self-recovery. The findings point to what needs to be in place to support self-recovery in dynamic physical environments, including geoscience information and advice, and restoration of infrastructure damaged by natural hazard events. Further research is necessary to understand the issues this raises for the shelter and geoscience communities, particularly around availability of geoscience expertise, capacity and information at a local scale.<br/
