    The watchdogs of 'Washminster' – parliamentary scrutiny of executive patronage in the UK

    The role of legislatures in scrutinising executive patronage has received scant attention in the context of parliamentary democracy. This article addresses this lacuna by focusing on the parliamentary scrutiny of public appointments in the UK. Presenting the results of an extensive programme of research, it reveals how select committees have accrued increasing powers to challenge ministerial appointments, and how this has resulted in a series of unintended consequences that raise critical concerns regarding the overall added-value of pre-appointment scrutiny. The article is therefore of comparative significance for theories of legislative scrutiny in particular and executive–legislature dynamics more broadly

    Epigenetic changes mediated by polycomb repressive complex 2 and E2a are associated with drug resistance in a mouse model of lymphoma

    Background: The genetic origins of chemotherapy resistance are well established; however the role of epigenetics in drug resistance is less well understood To investigate mechanisms of drug resistance we performed systematic genetic epigenetic and transcriptomic analyses of an alkylating agent-sensitive murine lymphoma cell line and a series of resistant lines derived by drug dose escalation   Methods: Dose escalation of the alkylating agent mafosfamide was used to create a series of increasingly drugresistant mouse Burkitt's lymphoma cell lines Whole genome sequencing DNA microarrays reduced representation bisulfite sequencing and chromatin immunoprecipitation sequencing were used to identify alterations in DNA sequence mRNA expression CpG methylation and H3K27me3 occupancy respectively that were associated with increased resistance   Results: Our data suggest that acquired resistance cannot be explained by genetic alterations Based on integration of transcriptional profiles with transcription factor binding data we hypothesize that resistance is driven by epigenetic plasticity We observed that the resistant cells had H3K27me3 and DNA methylation profiles distinct from those of the parental lines Moreover we observed DNA methylation changes in the promoters of genes regulated by E2a and members of the polycomb repressor complex 2 (PRC2) and differentially expressed genes were enriched for targets of E2a The integrative analysis considering H3K27me3 further supported a role for PRC2 in mediating resistance By integrating our results with data from the Immunological Genome Project (Immgenorg) we showed that these transcriptional changes track the B-cell maturation axis   Conclusions: Our data suggest a novel mechanism of drug resistance in which E2a and PRC2 drive changes in the B-cell epigenome; these alterations attenuate alkylating agent treatment-induced apoptosi

    The changing patterns of group politics in Britain

    Two interpretations of ways in which group politics in Britain have presented challenges to democracy are reviewed: neo-corporatism or pluralistic stagnation and the rise of single issue interest groups. The disappearance of the first paradigm created a political space for the second to emerge. A three-phase model of group activity is developed: a phase centred around production interests, followed by the development of broadly based 'other regarding' groups, succeeded by fragmented, inner directed groups focusing on particular interests. Explanations of the decay of corporatism are reviewed. Single issue group activity has increased as party membership has declined and is facilitated by changes in traditional media and the development of the internet. Such groups can overload the policy-making process and frustrate depoliticisation. Debates about the constitution and governance have largely ignored these issues and there is need for a debate

    VERITAS and Multiwavelength Observations of the BL Lacertae Object 1ES 1741+196

    We present results from multiwavelength observations of the BL Lacertae object 1ES 1741+196, including results in the very-high-energy Îł\gamma-ray regime using the Very Energetic Radiation Imaging Telescope Array System (VERITAS). The VERITAS time-averaged spectrum, measured above 180 GeV, is well-modelled by a power law with a spectral index of 2.7±0.7stat±0.2syst2.7\pm0.7_{\mathrm{stat}}\pm0.2_{\mathrm{syst}}. The integral flux above 180 GeV is (3.9±0.8stat±1.0syst)×10−8(3.9\pm0.8_{\mathrm{stat}}\pm1.0_{\mathrm{syst}})\times 10^{-8} m−2^{-2} s−1^{-1}, corresponding to 1.6% of the Crab Nebula flux on average. The multiwavelength spectral energy distribution of the source suggests that 1ES 1741+196 is an extreme-high-frequency-peaked BL Lacertae object. The observations analysed in this paper extend over a period of six years, during which time no strong flares were observed in any band. This analysis is therefore one of the few characterizations of a blazar in a non-flaring state.Comment: 8 pages, 5 figures. Accepted for publication in MNRA

    Very-High-Energy Îł\gamma-Ray Observations of the Blazar 1ES 2344+514 with VERITAS

    We present very-high-energy Îł\gamma-ray observations of the BL Lac object 1ES 2344+514 taken by the Very Energetic Radiation Imaging Telescope Array System (VERITAS) between 2007 and 2015. 1ES 2344+514 is detected with a statistical significance above background of 20.8σ20.8\sigma in 47.247.2 hours (livetime) of observations, making this the most comprehensive very-high-energy study of 1ES 2344+514 to date. Using these observations the temporal properties of 1ES 2344+514 are studied on short and long times scales. We fit a constant flux model to nightly- and seasonally-binned light curves and apply a fractional variability test, to determine the stability of the source on different timescales. We reject the constant-flux model for the 2007-2008 and 2014-2015 nightly-binned light curves and for the long-term seasonally-binned light curve at the >3σ> 3\sigma level. The spectra of the time-averaged emission before and after correction for attenuation by the extragalactic background light are obtained. The observed time-averaged spectrum above 200 GeV is satisfactorily fitted (χ2/NDF=7.89/6{\chi^2/NDF = 7.89/6}) by a power-law function with index Γ=2.46±0.06stat±0.20sys\Gamma = 2.46 \pm 0.06_{stat} \pm 0.20_{sys} and extends to at least 8 TeV. The extragalactic-background-light-deabsorbed spectrum is adequately fit (χ2/NDF=6.73/6{\chi^2/NDF = 6.73/6}) by a power-law function with index Γ=2.15±0.06stat±0.20sys\Gamma = 2.15 \pm 0.06_{stat} \pm 0.20_{sys} while an F-test indicates that the power-law with exponential cutoff function provides a marginally-better fit (χ2/NDF\chi^2/NDF = 2.56/52.56 / 5 ) at the 2.1σ\sigma level. The source location is found to be consistent with the published radio location and its spatial extent is consistent with a point source.Comment: 7 pages, 2 figures.     Climate change curricula for adult audiences in agriculture and forestry: A review

    Agricultural and forestry advisers and other technical service providers play an important role in supporting farmers and foresters to adapt to climate change. However, not all agricultural and forestry advisers are comfortable talking about climate change with land managers. While there is a demonstrated interest related to climate related professional development, few examples of curricula developed with the express purpose of serving this audience and a systematic review of these curricula has not been conducted. To address this gap, we reviewed 12 curricula which were developed and implemented between 2001 and 2017. The goal of this review is to apply the lessons learned from a range of climate change-focused curricula to new, regionally or sector-specific educational programs targeting both agricultural advisers and innovative farmers. Our findings suggest that developers of future educational programs consider the following: (a) the specific needs of their audience, including topical interests and learning needs; (b) the use of interdisciplinary teams for curricula development; (c) trade-offs associated with inclusivity and depth of course content; and (d) the advantages of project-based education approaches suited for adult learning audiences. By applying these concepts to future curricula, these curricula are likely to have the greatest level of impact

    Very-high-energy observations of the binaries V 404 Cyg and 4U 0115+634 during giant X-ray outbursts

    Transient X-ray binaries produce major outbursts in which the X-ray flux can increase over the quiescent level by factors as large as 10710^7. The low-mass X-ray binary V 404 Cyg and the high-mass system 4U 0115+634 underwent such major outbursts in June and October 2015, respectively. We present here observations at energies above hundreds of GeV with the VERITAS observatory taken during some of the brightest X-ray activity ever observed from these systems. No gamma-ray emission has been detected by VERITAS in 2.5 hours of observations of the microquasar V 404 Cyg from 2015, June 20-21. The upper flux limits derived from these observations on the gamma-ray flux above 200 GeV of F <4.4×10−12< 4.4\times 10^{-12} cm−2^{-2} s−1^{-1} correspond to a tiny fraction (about 10−610^{-6}) of the Eddington luminosity of the system, in stark contrast to that seen in the X-ray band. No gamma rays have been detected during observations of 4U 0115+634 in the period of major X-ray activity in October 2015. The flux upper limit derived from our observations is F <2.1×10−12< 2.1\times 10^{-12} cm−2^{-2} s−1^{-1} for gamma rays above 300 GeV, setting an upper limit on the ratio of gamma-ray to X-ray luminosity of less than 4%.Comment: Accepted for publication in the Astrophysical Journa
