    Diagnosis i proposta d'intervenció de la masia Can Jan de Santa Maria de Palautordera

    L’objecte d’aquest projecte és realitzar un estudi minuciós sobre l’estat actual de la masia Can Jan de Santa Maria de Palautordera i oferir una resposta contundent a tota la problemàtica que pateix. La proposta no es limita només a solucionar els problemes actuals que pateix sinó a garantir la seva qualitat i durabilitat. Una de les motivacions per escollir aquesta masia ha estat la proposta per part de l’ajuntament de Santa Maria de Palautordera de rehabilitar certes masies del municipi incloses al Catàleg de Patrimoni de la Generalitat. Aquestes masies tenen un gran valor pel municipi pel seu valor arquitectònic i per la representació patrimonial del territori. Aquestes condicions m’han motivat a profunditzar en les característiques de les vivendes que feien possible el tipus de vida que la població del municipi ha anat desenvolupant segons l’època. Analitzar la compartimentació que els inquilins necessitaven per dur a terme la seva vida quotidiana de la millor manera possible. També això em permetrà investigar i comprendre l’estructura i construcció de les masies de l’època. Un altre factor a tenir en comte, dintre de les possibles masies a tractar, és l’accessibilitat a la propietat per poder realitzar les visites corresponents. L’únic inconvenient de tot aquest conjunt de masies és la impossibilitat de realitzar cales en cap d’elles. L’objectiu d’aquest projecte és posar en pràctica tots els coneixements adquirits durant el recorregut dels quatre anys d’estudi universitari. Tota la teoria s’ha de posar en pràctica per reflectir la capacitat d’acció envers una problemàtica per solucionar-la de la manera més eficaç, rapida i econòmica. Tots aquests motius m’han conduït a dur a terme aquesta diagnosi i la seva corresponent proposta d’intervenció

    Catalyst-Free Vapor-Phase Method for Direct Integration of Gas Sensing Nanostructures with Polymeric Transducing Platforms

    Tungsten oxide nanoneedles (NNs) are grown and integrated directly with polymeric transducing platforms for gas sensors via aerosol-assisted chemical vapor deposition (AACVD) method. Material analysis shows the feasibility to grow highly crystalline nanomaterials in the form of NNs with aspect ratios between 80 and 200 and with high concentration of oxygen vacancies at the surface, whereas gas testing demonstrates moderate sensing responses to hydrogen at concentrations between 10 ppm and 50 ppm, which are comparable with results for tungsten oxide NNs grown on silicon transducing platforms. This method is demonstrated to be an attractive route to fabricate next generation of gas sensors devices, provided with flexibility and functionality, with great potential in a cost effective production for large-scale applications

    The Second Wind in McArdle Patients: Fitness Matters

In this regard, an increase in the workload eliciting the SW could potentially translate into an improved patients’ exercise tolerance in daily life. We aimed to determine whether aerobic fitness and physical activity (PA) levels are correlated with the minimum workload eliciting the SW in McArdle patients—as well as with the corresponding heart rate value. We also compared the SW variables and aerobic fitness indicators in inactive vs. active patients. Methods: Fifty-four McArdle patients (24 women, mean ± SD age 33 ± 12 years) performed 12-min constant-load and maximum ramp-like cycle-ergometer tests for SW detection and aerobic fitness [peak oxygen uptake (VO2peak) and workload and ventilatory threshold] determination, respectively. They were categorized as physically active/inactive during the prior 6 months (active = reporting ≥150 min/week or ≥75 min/week in moderate or vigorous-intensity aerobic PA, respectively) and were also asked on their self-report of the SW. Results: Both peak and submaximal indicators of aerobic fitness obtained in the ramp tests were significantly correlated with the workload of the SW test, with a particularly strong correlation for the VO2peak and peak workload attained by the patients (both Pearson’s coefficients > 0.70). Twenty (seven women) and 24 patients (18 women) were categorized as physically active and inactive, respectively. Not only the aerobic fitness level [∼18–19% higher values of VO2peak (ml⋅kg–1⋅min–1)] but also the workload of the SW tests was significantly higher in active than in inactive patients. All the inactive patients reported that they experienced the SW during walking/brisk walking in daily life, whereas active patients only reported experiencing this phenomenon during more strenuous activities (very brisk walking/jogging and bicycling). Conclusion: A higher aerobic fitness and an active lifestyle are associated with a higher workload eliciting the so-called SW phenomenon in patients with McArdle disease, which has a positive impact on their exercise tolerance during daily living.Research by AL, CF-L, GN-G, MM, and TP was funded by the Instituto de Salud Carlos III (ISCIII) and Fondos Feder (grants PI18/00139, PI20/00645, PI18/00713, PI17/00093, and PI19/01313).     Heterologous Expression of the Phytochelatin Synthase CaPCS2 from Chlamydomonas acidophila and Its Effect on Different Stress Factors in Escherichia coli

    Phytochelatins (PCs) are cysteine-rich small peptides, enzymatically synthesized from reduced glutathione (GSH) by cytosolic enzyme phytochelatin synthase (PCS). The open reading frame (ORF) of the phytochelatin synthase CaPCS2 gene from the microalgae Chlamydomonas acidophila was heterologously expressed in Escherichia coli strain DH5 alpha, to analyze its role in protection against various abiotic agents that cause cellular stress. The transformed E. coli strain showed increased tolerance to exposure to different heavy metals (HMs) and arsenic (As), as well as to acidic pH and exposure to UVB, salt, or perchlorate. In addition to metal detoxification activity, new functions have also been reported for PCS and PCs. According to the results obtained in this work, the heterologous expression of CaPCS2 in E. coli provides protection against oxidative stress produced by metals and exposure to different ROS-inducing agents. However, the function of this PCS is not related to HM bioaccumulation

    Salt resistance genes revealed by functional metagenomics from brines and moderate-salinity rhizosphere within a hypersaline environment

    Hypersaline environments are considered one of the most extreme habitats on earth and microorganisms have developed diverse molecular mechanisms of adaptation to withstand these conditions. The present study was aimed at identifying novel genes from the microbial communities of a moderate-salinity rhizosphere and brine from the Es Trenc saltern (Mallorca, Spain), which could confer increased salt resistance to Escherichia coli. The microbial diversity assessed by pyrosequencing of 16S rRNA gene libraries revealed the presence of communities that are typical in such environments and the remarkable presence of three bacterial groups never revealed as major components of salt brines. Metagenomic libraries from brine and rhizosphere samples, were transferred to the osmosensitive strain E. coli MKH13, and screened for salt resistance. Eleven genes that conferred salt resistance were identified, some encoding for well-known proteins previously related to osmoadaptation such as a glycerol transporter and a proton pump, whereas others encoded proteins not previously related to this function in microorganisms such as DNA/RNA helicases, an endonuclease III (Nth) and hypothetical proteins of unknown function. Furthermore, four of the retrieved genes were cloned and expressed in Bacillus subtilis and they also conferred salt resistance to this bacterium, broadening the spectrum of bacterial species in which these genes can function.     Reconstructing the Snow Avalanche of Coll de Pal 2018 (SE Pyrenees).

    Developments in mountain areas prone to natural hazards produce undesired impacts and damages. Thus, disaster assessment is mandatory to understand the physics of dangerous events and to make decisions to prevent hazardous situations. This work focusses on the practical implementation of methods and tools to assess a snow avalanche that affected a road at the Coll de Pal in 2018 (SE Pyrenees). This is a quite common situation in mountain roads and the assessment has to focus specially in the avalanche-road interaction, on the return periods considered and on the dynamics of the phenomena. This assessment presents the field recognition, snow and weather characterization and numerical modelling of the avalanche. Field campaigns revealed evidences of the avalanche triggering, runout trajectory and general behavior. An unstable situation of the snowpack due to a relatively large snowfall fallen some days before over a previous snowpack with weak layers, caused the avalanche triggering when an additional load was added by a strong wind-drift episode. A medium size (<2500 m3) soft slab avalanche, corresponding to a return period of 15-20 years, occurred and crossed the road of the Coll de Pal pass. The event was reproduced numerically by means of the 2D-SWE based numerical tool Iber aiming to analyze the avalanche behavior. Results of the simulation corresponded with the observations (runout trajectory and snow deposit); thus, relevant information about the avalanche dynamics could be obtained.     Virtual reality body exposure therapy for anorexia nervosa. A case report with follow-up results

    Objective: Exposure-based therapies such as mirror exposure may help to improve the results of classic cognitive behavioral therapy in anorexia nervosa (AN). Virtual reality (VR)-based procedures provide interesting novelties for targeting body-related concerns. This study aimed to provide preliminary evidence of the usefulness of a VR body exposure therapy in a patient diagnosed with AN. Method: Fear of gaining weight (FGW), body anxiety, drive for thinness, body image disturbances, body mass index and body-related attentional bias were assessed before and after the intervention, as well as five months later. Five sessions of VR body exposure therapy were included within the standard course of cognitive behavioral therapy. The sessions involved a systematic and hierarchical exposure of the patient to a virtual representation of her own silhouette, with the body mass index of the avatar progressively increasing in subsequent sessions. Results: After the intervention, there was a clear reduction in AN symptoms such as the FGW, drive for thinness, body-related anxiety and dissatisfaction. Body mass index values rose continuously during the intervention and reached healthy levels. Finally, there was a notable change in the dysfunctional body-related attentional bias. Almost all these improvements were maintained after five months, except for the FGW. Conclusion: To the best of our knowledge, this study is the first to focus on treating the FGW and body-related concerns in AN using a VR-based paradigm. To pursue this study further and assess the effectiveness of this new VR software, larger controlled clinical trials are needed

    Novel Genes Involved in Resistance to Both Ultraviolet Radiation and Perchlorate From the Metagenomes of Hypersaline Environments

    Microorganisms that thrive in hypersaline environments on the surface of our planet are exposed to the harmful effects of ultraviolet radiation. Therefore, for their protection, they have sunscreen pigments and highly efficient DNA repair and protection systems. The present study aimed to identify new genes involved in UV radiation resistance from these microorganisms, many of which cannot be cultured in the laboratory. Thus, a functional metagenomic approach was used and for this, small-insert libraries were constructed with DNA isolated from microorganisms of high-altitude Andean hypersaline lakes in Argentina (Diamante and Ojo Seco lakes, 4,589 and 3,200 m, respectively) and from the Es Trenc solar saltern in Spain. The libraries were hosted in a UV radiation-sensitive strain of Escherichia coli (recA mutant) and they were exposed to UVB. The resistant colonies were analyzed and as a result, four clones were identified with environmental DNA fragments containing five genes that conferred resistance to UV radiation in E. coli. One gene encoded a RecA-like protein, complementing the mutation in recA that makes the E. coli host strain more sensitive to UV radiation. Two other genes from the same DNA fragment encoded a TATA-box binding protein and an unknown protein, both responsible for UV resistance. Interestingly, two other genes from different and remote environments, the Ojo Seco Andean lake and the Es Trenc saltern, encoded two hypothetical proteins that can be considered homologous based on their significant amino acid similarity (49%). All of these genes also conferred resistance to 4-nitroquinoline 1-oxide (4-NQO), a compound that mimics the effect of UV radiation on DNA, and also to perchlorate, a powerful oxidant that can induce DNA damage. Furthermore, the hypothetical protein from the Es Trenc salterns was localized as discrete foci possibly associated with damaged sites in the DNA in cells treated with 4-NQO, so it could be involved in the repair of damaged DNA. In summary, novel genes involved in resistance to UV radiation, 4-NQO and perchlorate have been identified in this work and two of them encoding hypothetical proteins that could be involved in DNA damage repair activities not previously described.Fil: Lamprecht Grandío, María.     Ultrasensitive mass sensor fully integrated with complementary metal-oxide-semiconductor circuitry

    Nanomechanical resonators have been monolithically integrated on preprocessed complementary metal-oxide-semiconductor(CMOS) chips. Fabricatedresonatorsystems have been designed to have resonance frequencies up to 1.5 MHz. The systems have been characterized in ambient air and vacuum conditions and display ultrasensitive mass detection in air. A mass sensitivity of 4ag/Hz has been determined in air by placing a single glycerine drop, having a measured weight of 57 fg, at the apex of a cantilever and subsequently measuring a frequency shift of 14.8 kHz.
