Control of the pinewood nematode Bursaphelenchus xylophilus by essential oils and extracts obtained from plants: a review.
The pinewood nematode (PWN), Bursaphelenchus xylophilus, is a serious threat to
forest ecosystems at a global scale. The nematode has become a major quarantine
problem due to its capability to completely destroy Pinus spp. trees, with great damage to
the wood industry. Controlling the nematode inside a living tree is quite difficult, the
techniques used being often ineffective and quite expensive. In the coming years, most
chemicals used to control nematodes will be banned and replaced by safer and
environmentally friendly products. As so, chemicals naturally produced by plants will play
an important role in controlling diseases such as pine wilt. Plants, particularly aromatic
ones, are commonly used due to the chemical properties of their secondary metabolites.
Among these, essential oils and/or extracts are highly employed and are being tested as
possible control of some organisms, like nematodes. Recent publications have evaluated
essential oils derived from different plant species as natural nematicides [1; 2], antibacterial
[3], anti-fungal [4] as well as insecticidal [5]. Concerning control of the PWN, a
significant amount of information on plants tested, results obtained and employed
techniques, is available. Our revision has extensively gathered this information, making it
easier to search, read and use. It may become useful information for future studies on the
subject, since it will be possible to check the plants already tested. Although numbers
aren´t definitive, so far, tested plants are distributed amongst 148 families. The extracts or
essential oils of plants belonging to the Asteraceae, Lamiaceae and Euphorbiaceae
families show promising results on controlling the pinewood nematode
Mineralization of sheep manure and its influence on lettuce production
Diversos resíduos orgânicos são utilizados na agricultura sem o adequado conhecimento da sua dinâmica de mineralização. Avaliou-se a mineralização de esterco de ovinos e sua influência na produção de alface. Utilizou-se o delineamento experimental de blocos ao acaso com três repetições. Foram utilizadas 25 t ha-1 como dose de esterco para cada um dos seguintes tratamentos: 1) esterco de ovinos que se alimentaram de feno de mandioca (PAM); 2) esterco de ovinos que se alimentaram de subproduto de ervilha (ERV); 3) esterco de ovinos que se alimentaram de feno de capim coast-cross (FCC); 4) esterco de ovinos que se alimentaram de subproduto de saccharina (SAC) e 5) solo sem aplicação de esterco (testemunha). Foi determinada semanalmente a respiração basal do solo, utilizada como indicador de mineralização da matéria orgânica. A massa fresca de alface foi avaliada como medida de produção. Os tratamentos ERV, FCC e SAC apresentaram ganhos de massa fresca na ordem de 68, 65 e 62% em relação à testemunha e de 43, 39 e 33% em relação ao PAM, respectivamente. A produção menor promovida pelo PAM, em relação às demais, pode ser explicada pela forma de mineralização da matéria orgânica que apresentou elevada respiração microbiana cinco dias após o transplantio, com acentuado declínio, nas medições subseqüentes, ao longo do ciclo da cultura. Os demais tratamentos apresentaram mineralização sincronizada com conseqüente aumento na produção de massa fresca.
____________________________________________________________________________________________________________ ABSTRACTSeveral organic wastes are used in agriculture with no precise knowledge about the mineralization dynamics of these materials. In this study the sheep manure mineralization and its influence on the lettuce production was evaluated. A randomized block design with three replications was used. Five treatments were studied using 25 t ha-1 as dose of manure: 1) sheep manure obtained from animals fed with cassava straw (PAM); 2) sheep manure obtained from animals fed with residue of pea crop (ERV); 3) sheep manure obtained from animals fed with Coast-Cross hay (FCC), 4) sheep manure obtained from animals fed with saccharin residue (SAC) and 5) soil without application of manure (control). Weekly the basal respiration was determined and used as an indicator of organic matter mineralization. Lettuce fresh mass was evaluated as a measure of production. Treatments ERV, FCC and SAC showed superior weight gains of 68, 65 and 62% compared to the control and 43, 39 and 33% compared to MAP, respectively. Lower production promoted by the MAP in relation to the other treatments can be explained by organic matter mineralization that showed a high microbial respiration five days after transplanting, with marked decline in subsequent measurements during the crop cycle. Patent Human Infections with the Whipworm, Trichuris trichiura, Are Not Associated with Alterations in the Faecal Microbiota
Background: The soil-transmitted helminth (STH), Trichuris trichiura colonises the human large intestine where it may
modify inflammatory responses, an effect possibly mediated through alterations in the intestinal microbiota. We
hypothesised that patent T. trichiura infections would be associated with altered faecal microbiota and that anthelmintic treatment would induce a microbiota resembling more closely that observed in uninfected individuals.
Materials and Methods: School children in Ecuador were screened for STH infections and allocated to 3 groups: uninfected, T. trichiura only, and mixed infections with T. trichiura and Ascaris lumbricoides. A sample of uninfected children and those with T. trichiura infections only were given anthelmintic treatment. Bacterial community profiles in faecal samples were studied by 454 pyrosequencing of 16 S rRNA genes.
Results: Microbiota analyses of faeces were done for 97 children: 30 were uninfected, 17 were infected with T. trichiura, and 50 with T. trichiura and A. lumbricoides. Post-treatment samples were analyzed for 14 children initially infected with T. trichiura alone and for 21 uninfected children. Treatment resulted in 100% cure of STH infections. Comparisons of the microbiota at different taxonomic levels showed no statistically significant differences in composition between uninfected
children and those with T. trichiura infections. We observed a decreased proportional abundance of a few bacterial genera from the Clostridia class of Firmicutes and a reduced bacterial diversity among children with mixed infections compared to the other two groups, indicating a possible specific effect of A. lumbricoides infection. Anthelmintic treatment of children with T. trichiura did not alter faecal microbiota composition.
Discussion: Our data indicate that patent human infections with T. trichiura may have no effect on faecal microbiota but that A. lumbricoides colonisation might be associated with a disturbed microbiota. Our results also catalogue the microbiota of rural Ecuadorians and indicate differences with individuals from more urban industrialised societies
An apoplastic fluid extraction method for the characterization of grapevine leaves proteome and metabolome from a single sample
The analysis of complex biological systems keeps challenging
researchers. The main goal of systems biology is to decipher interactions
within cells, by integrating datasets from large scale analytical
approaches including transcriptomics, proteomics and metabolomics
andmore specialized ‘OMICS’ such as epigenomics and lipidomics. Studying
different cellular compartments allows a broader understanding of cell
dynamics. Plant apoplast, the cellular compartment external to the plasma
membrane including the cell wall, is particularly demanding to analyze.
Despite our knowledge on apoplast involvement on several processes from
cell growth to stress responses, its dynamics is still poorly known due to the
lack of efficient extraction processes adequate to each plant system.Analyzing
woody plants such as grapevine raises even more challenges. Grapevine is
among the most important fruit crops worldwide and awider characterization
of its apoplast is essential for a deeper understanding of its physiology and cellular
mechanisms. Here, we describe, for the first time, a vacuum-infiltrationcentrifugationmethod
that allows a simultaneous extraction of grapevine apoplastic
proteins and metabolites from leaves on a single sample, compatible
with high-throughput mass spectrometry analyses. The extracted apoplast
from two grapevine cultivars, Vitis vinifera cv ‘Trincadeira’ and ‘Regent’, was
directly used for proteomics and metabolomics analysis. The proteome was
analyzed by nanoLC-MS/MS and more than 700 common proteinswere identified,
with highly diverse biological functions. The metabolome profile
through FT-ICR-MS allowed the identification of 514 unique putative compounds

Future directions in managing aniridia-associated keratopathy
Congenital aniridia is a panocular disorder that is typically characterized by iris hypoplasia and aniridia-associated keratopathy (AAK). AAK results in the progressive loss of corneal transparency and thereby loss of vision. Currently, there is no approved therapy to delay or prevent its progression, and clinical management is challenging because of phenotypic variability and high risk of complications after interventions; however, new insights into the molecular pathogenesis of AAK may help improve its management. Here, we review the current understanding about the pathogenesis and management of AAK. We highlight the biological mechanisms involved in AAK development with the aim to develop future treatment options, including surgical, pharmacological, cell therapies, and gene therapies

Schistosoma haematobium Treatment in 1–5 Year Old Children: Safety and Efficacy of the Antihelminthic Drug Praziquantel
Urogenital schistosomiasis is an important, but neglected, infectious disease affecting over 100 million people, mainly in Africa. Children carry the heaviest burden of infection with children as young as 1 year old showing signs of infection. Children aged 5 years and below are currently excluded from schistosome control programmes for several reasons, including operational difficulties associated with accessing preschool children, misconceptions about their level of exposure to infective water and lack of safety data on the drug of choice for schistosome control, praziquantel, in children aged 5 years and below. This study was one of a small number of studies recently funded by the World Health Organization to investigate the need for praziquantel treatment in preschool children (aged 1–5 years) and to subsequently assess the safety and efficacy of the drug praziquantel in this age group. This study confirmed that preschool children carry significant levels of schistosome infection, exceeding those carried by their parents/guardians, highlighting the urgent need for their immediate inclusion in schistosome control programmes. The study also showed that praziquantel treatment is as safe and efficacious in children aged 1–5 years as it is in older children aged 6–10 years who are currently the target for mass drug administration
