    A four component system was developed which consists of a particle size reduction mechanism, a pneumatic waste transport system, a rotating-paddle incinerator, and a catalytic afterburner to be integrated into a six-man, zero-g subsystem for processing human wastes on board spacecraft. The study included the development of different concepts or functions, the establishment of operational specifications, and a critical evaluation for each of the four components. A series of laboratory tests was run, and a baseline subsystem design was established. An operational specification was also written in preparation for detailed design and testing of this baseline subsystem

    Buckling behavior of Rene 41 tubular panels for a hypersonic aircraft wing

    The buckling characteristics of Rene 41 tubular panels for a hypersonic aircraft wing were investigated. The panels were repeatedly tested for buckling characteristics using a hypersonic wing test structure and a universal tension/compression testing machine. The nondestructive buckling tests were carried out under different combined load conditions and in different temperature environments. The force/stiffness technique was used to determine the buckling loads of the panel. In spite of some data scattering, resulting from large extrapolations of the data fitting curve (because of the termination of applied loads at relatively low percentages of the buckling loads), the overall test data correlate fairly well with theoretically predicted buckling interaction curves. Also, the structural efficiency of the tubular panels was found to be slightly higher than that of beaded panels

    A baseline laboratory prototype of an integrated, six man, zero-g subsystem for processing human wastes onboard spacecraft was investigated, and included the development of an operational specification for the baseline subsystem, followed by design and fabrication. The program was concluded by performing a series of six tests over a period of two weeks to evaluate the performance of the subsystem. The results of the tests were satisfactory, however, several changes in the design of the subsystem are required before completely satisfactory performance can be achieved

    Intrinsic tethering activity of endosomal Rab proteins.

    Rab small G proteins control membrane trafficking events required for many processes including secretion, lipid metabolism, antigen presentation and growth factor signaling. Rabs recruit effectors that mediate diverse functions including vesicle tethering and fusion. However, many mechanistic questions about Rab-regulated vesicle tethering are unresolved. Using chemically defined reaction systems, we discovered that Vps21, a Saccharomyces cerevisiae ortholog of mammalian endosomal Rab5, functions in trans with itself and with at least two other endosomal Rabs to directly mediate GTP-dependent tethering. Vps21-mediated tethering was stringently and reversibly regulated by an upstream activator, Vps9, and an inhibitor, Gyp1, which were sufficient to drive dynamic cycles of tethering and detethering. These experiments reveal a previously undescribed mode of tethering by endocytic Rabs. In our working model, the intrinsic tethering capacity Vps21 operates in concert with conventional effectors and SNAREs to drive efficient docking and fusion

    Factors deterring and prompting the decision to attempt suicide on the railway networks: findings from 353 online surveys and 34 semi-structured interviews

    Background There is a suicide on the British railways every 36 hours. However, the reasons why people choose to die by train are not well understood. Aims To explore factors influencing and discouraging the decision to attempt suicide on the railway networks. Method We conducted an online survey and qualitative interviews with individuals who had contemplated or attempted suicide by train. Results A total of 353 survey responders had considered and 23 had attempted suicide at rail locations (including railways and metro/underground); a third of these cases were impulsive. The most frequently reported motivations for contemplating or attempting suicide were perceptions of quick and certain lethality (54 and 37%, respectively) and easy access to rail settings (33 and 38%, respectively). The main factor discouraging people from rail suicide was its wider impact, especially on train drivers (19%). In qualitative interviews (N = 34) the desire to avoid intervention from others was also a common motivating factor for attempting suicide on the railway networks. Conclusions People attempt suicide by train because railway settings are easy to access and because of an inaccurate perception of certain and quick lethality. Tackling exaggerated perceptions of lethality may help reduce suicides by train

    Superluminous supernovae: No threat from Eta Carinae

    Recently Supernova 2006gy was noted as the most luminous ever recorded, with a total radiated energy of ~10^44 Joules. It was proposed that the progenitor may have been a massive evolved star similar to eta Carinae, which resides in our own galaxy at a distance of about 2.3 kpc. eta Carinae appears ready to detonate. Although it is too distant to pose a serious threat as a normal supernova, and given its rotation axis is unlikely to produce a Gamma-Ray Burst oriented toward the Earth, eta Carinae is about 30,000 times nearer than 2006gy, and we re-evaluate it as a potential superluminous supernova. We find that given the large ratio of emission in the optical to the X-ray, atmospheric effects are negligible. Ionization of the atmosphere and concomitant ozone depletion are unlikely to be important. Any cosmic ray effects should be spread out over ~10^4 y, and similarly unlikely to produce any serious perturbation to the biosphere. We also discuss a new possible effect of supernovae, endocrine disruption induced by blue light near the peak of the optical spectrum. This is a possibility for nearby supernovae at distances too large to be considered "dangerous" for other reasons. However, due to reddening and extinction by the interstellar medium, eta Carinae is unlikely to trigger such effects to any significant degree.Comment: 19 pages, 2 figures; Revised version as accepted for publication in Astrobiolog

    Neuropilins 1 and 2 mediate neointimal hyperplasia and re-endothelialization following arterial injury

    AIMS: Neuropilins 1 and 2 (NRP1 and NRP2) play crucial roles in endothelial cell migration contributing to angiogenesis and vascular development. Both NRPs are also expressed by cultured vascular smooth muscle cells (VSMCs) and are implicated in VSMC migration stimulated by PDGF-BB, but it is unknown whether NRPs are relevant for VSMC function in vivo. We investigated the role of NRPs in the rat carotid balloon injury model, in which endothelial denudation and arterial stretch induce neointimal hyperplasia involving VSMC migration and proliferation. METHODS AND RESULTS: NRP1 and NRP2 mRNAs and proteins increased significantly following arterial injury, and immunofluorescent staining revealed neointimal NRP expression. Down-regulation of NRP1 and NRP2 using shRNA significantly reduced neointimal hyperplasia following injury. Furthermore, inhibition of NRP1 by adenovirally overexpressing a loss-of-function NRP1 mutant lacking the cytoplasmic domain (ΔC) reduced neointimal hyperplasia, whereas wild-type (WT) NRP1 had no effect. NRP-targeted shRNAs impaired, while overexpression of NRP1 WT and NRP1 ΔC enhanced, arterial re-endothelialization 14 days after injury. Knockdown of either NRP1 or NRP2 inhibited PDGF-BB-induced rat VSMC migration, whereas knockdown of NRP2, but not NRP1, reduced proliferation of cultured rat VSMC and neointimal VSMC in vivo. NRP knockdown also reduced the phosphorylation of PDGFα and PDGFβ receptors in rat VSMC, which mediate VSMC migration and proliferation. CONCLUSION: NRP1 and NRP2 play important roles in the regulation of neointimal hyperplasia in vivo by modulating VSMC migration (via NRP1 and NRP2) and proliferation (via NRP2), independently of the role of NRPs in re-endothelialization
