    Toll-like receptor 2 expression on c-kit + cells tracks the emergence of embryonic definitive hematopoietic progenitors

    International audienceHematopoiesis in mammalian embryos proceeds through three successive waves of hema-topoietic progenitors. Since their emergence spatially and temporally overlap and phenotypic markers are often shared, the specifics regarding their origin, development, lineage restriction and mutual relationships have not been fully determined. The identification of wave-specific markers would aid to resolve these uncertainties. Here, we show that toll-like receptors (TLRs) are expressed during early mouse embryogenesis. We provide phenotypic and functional evidence that the expression of TLR2 on E7.5 c-kit + cells marks the emergence of precursors of erythro-myeloid progenitors (EMPs) and provides resolution for separate tracking of EMPs from primitive progenitors. Using in vivo fate mapping, we show that at E8.5 the Tlr2 locus is already active in emerging EMPs and in progenitors of adult hematopoietic stem cells (HSC). Together, this data demonstrates that the activation of the Tlr2 locus tracks the earliest events in the process of EMP and HSC specification

    Inhalation of ZnO nanoparticles: Splice junction expression and alternative splicing in mice

    Despite the wide application of nanomaterials, toxicity studies of nanoparticles (NP) are often limited to in vitro cell models, and the biological impact of NP exposure in mammals has not been thoroughly investigated. Zinc oxide (ZnO) NPs are commonly used in various consumer products. To evaluate the effects of the inhalation of ZnO NP in mice, we studied splice junction expression in the lungs as a proxy to gene expression changes analysis. Female ICR mice were treated with 6.46 x 10(4) and 1.93 x 10(6) NP/cm(3) for 3 days and 3 months, respectively. An analysis of differential expression and alternative splicing events in 298 targets (splice junctions) of 68 genes involved in the processes relevant to the biological effects of ZnO NP was conducted using next-generation sequencing. Three days of exposure resulted in the upregulation of IL-6 and downregulation of BID, GSR, NF-kB2, PTGS2, SLC11A2, and TXNRD1 splice junction expression; 3 months of exposure increased the expression of splice junctions in ALDH3A1, APAF1, BID, CASP3, DHCR7, GCLC, GCLM, GSR, GSS, EHHADH, FAS, HMOX-1, IFN, NF-kB1, NQO-1, PTGS1, PTGS2, RAD51, RIPK2, SRXN1, TRAF6, and TXNRD1. Alternative splicing of TRAF6 and TXNRD1 was induced after 3 days of exposure to 1.93 x 10(6) NP/cm(3). In summary, we observed changes of splice junction expression in genes involved in oxidative stress, apoptosis, immune response, inflammation, and DNA repair, as well as the induction of alternative splicing in genes associated with oxidative stress and inflammation. As a valuation method was used the earnings valuation method based on discounted free cash flow to the company. Except of value of the company, there is determined internal value of private equity. Financial analysis of the company and strategic analysis are applied in the process of valuation. Process of valuation is based on financial plan and another input variables such as avarage cost of capital. Delivery of the statement of the company value is followed by valuation. A single-factor sensitivity analysis has been carried out too. It focuses on individual factors that affect the final value of the company

    Cílem této práce je navrhnout moderní design konstruktoru parserů a návrh poté realizovat. Výsledkem práce je programátorské dílo sestávající z programu, který ze vstupního souboru čte popis bezkontextové LALR(1) gramatiky a sémantických akcí. Do výstupních souborů generuje zdrojový kód syntaktického analyzátoru, který při parsování jazyka odpovídajícího dané gramatice vykonává dané sémantické akce. Součástí jsou šablony zdrojových kódů pro implementaci výsledného analyzátoru, a to pro několik cílových moderních objektových programovacích jazyků. The result is a program that reads description of some context-free LALR(1) grammar and semantic actions from an input file. To output files the program generates source codes in several target modern object-oriented programming languages for implementation of the syntax analyzer which, when parsing the language corresponding to the given grammar, executes the given semantic actions.     Diplomová práce se zabývá oceněním společnosti LINET spol. s.r.o. k datu 08. 12. 2012. Ocenění společnosti je provedeno výnosovou metodou diskontovaného peněžního toku pro vlastníky a věřitele. Kromě hodnoty podniku je stanovena i vnitřní hodnota vlastního kapitálu. Při procesu oceňování je provedena strategická a finanční analýza. Ocenění společnosti vychází z finančního plánu a dalších vstupních proměnných jako třeba průměrné náklady na kapitál. Po ocenění následuje výrok o hodnotě podniku.     The aim of bachelor study is focused on financial analysis of LINET company s r.o. and concentrate on financial situation of company. Analysis was made over the economy years 2003/2004-2007/2008. In the first part the study describes process of financial analysis, theoretical part describes instruments and methods of calculation which was used in this study and practical part describes brief description of the company and calculations of particular instruments. All attainable results always have verbal commentary and compare with average results in branch. Situation of the company is summarized in the end of this study and proposals on her improvement
