    ArtinM offers new perspectives in the development of antifungal therapy

    The thermally dimorphic fungus Paracoccidioides brasiliensis is the causative agent of paracoccidioidomycosis (PCM), the most frequent systemic mycosis that affects the rural populations in Latin America. Despite significant developments in antifungal chemotherapy, its efficacy remains limited since drug therapy is prolonged and associated with toxic side effects and relapses. In response to these challenges, it is now recognized that several aspects of antifungal immunity can be modulated to better deal with fungal infections. A common idea for halting fungal infections has been the need to activate a cell-based, pro-inflammatory Th1 immune response to improve the fungal elimination. ArtinM, a D-mannose binding lectin from Artocarpus heterophyllus, has the property of modulating immunity against several intracellular pathogens. Here, we review the immunomodulatory activity of ArtinM during experimental PCM in mice. Both prophylactic and therapeutic protocols of ArtinM administration promotes a Th1 immune response balanced by IL-10, which outstandingly reduces the fungal load in organs of the treated mice while maintaining a controlled inflammation at the site of infection. A carbohydrate recognition-based interaction of ArtinM with Toll-like receptor 2 (TLR2) accounts for initiating the immunomodulatory effect of the lectin. The precise identification of the TLR2 N-glycan(s) targeted by ArtinM may support novel basis for the development of antifungal therapy

    A utilização racional de energia (URE) visa proporcionar o mesmo nível de produção de bens, serviços e de conforto através de tecnologias que reduzem os consumos face a soluções convencionais. A URE pode conduzir a reduções substanciais do consumo de energia e das emissões de poluentes associadas à sua conversão. Em muitas situações a URE pode também conduzir a uma elevada economia nos custos do ciclo de vida dos equipamentos utilizadores de energia (custo inicial mais custo de funcionamento ao longo da vida útil). Embora geralmente sejam mais dispendiosos, em termos de custo inicial, os equipamentos mais eficientes consomem menos energia, conduzindo a custos de funcionamento mais reduzidos e apresentando outras vantagens adicionais. Os motores elétricos são de longe as cargas mais importantes na indústria e no sector terciário. A figura 1 mostra a importância relativa da força motriz nesses sectores. A iluminação aparece como a carga mais importante no sector terciário, sendo na indústria a segunda carga mais relevante. Os motores elétricos são utilizados numa vasta gama de aplicações, principalmente na movimentação de fluidos em bombas, compressores e ventiladores

    Nucleotide-Binding Oligomerization Domain-1 and -2 Play No Role in Controlling Brucella abortus Infection in Mice

    Nucleotide-binding oligomerization domain proteins (NODs) are modular cytoplasmic proteins implicated in the recognition of peptidoglycan-derived molecules. Further, several in vivo studies have demonstrated a role for Nod1 and Nod2 in host defense against bacterial pathogens. Here, we demonstrated that macrophages from NOD1-, NOD2-, and Rip2-deficient mice produced lower levels of TNF-α following infection with live Brucella abortus compared to wild-type mice. Similar reduction on cytokine synthesis was not observed for IL-12 and IL-6. However, NOD1, NOD2, and Rip2 knockout mice were no more susceptible to infection with virulent B. abortus than wild-type mice. Additionally, spleen cells from NOD1-, NOD2-, and Rip2-deficient mice showed unaltered production of IFN-γ compared to C57BL/6 mice. Taken together, this study demonstrates that NOD1, NOD2 and Rip2 are dispensable for the control of B. abortus during in vivo infection

    Non-interactive effects drive multiple stressor impacts on the taxonomic and functional diversity of atlantic stream macroinvertebrates

    Freshwaters are considered among the most endangered ecosystems globally due to multiple stressors, which coincide in time and space. These local stressors typically result from land-use intensification or hydroclimatic alterations, among others. Despite recent advances on multiple stressor effects, current knowledge is still limited to manipulative approaches minimizing biological and abiotic variability. Thus, the assessment of multiple stressor effects in real-world ecosystems is required. Using an extensive survey of 50 stream reaches across North Portugal, we evaluated taxonomic and functional macroinvertebrate responses to multiple stressors, including marked gradients of nutrient enrichment, flow reduction, riparian vegetation structure, thermal stress and dissolved oxygen depletion. We analyzed multiple stressor effects on two taxonomic (taxon richness, Shannon-diversity) and two trait-based diversity indices (functional richness, functional dispersion), as well as changes in trait composition. We found that multiple stressors had additive effects on all diversity metrics, with nutrient enrichment identified as the most important stressor in three out of four metrics, followed by dissolved oxygen depletion and thermal stress. Taxon richness, Shannon-diversity and functional richness responded similarly, whereas functional dispersion was driven by changes in flow velocity and thermal stress. Functional trait composition changed along a major stress gradient determined by nutrient enrichment and oxygen depletion, which was positively correlated with organisms possessing fast-living strategies, aerial respiration, adult phases, and gathering-collector feeding habits. Overall, our results reinforce the need to consider complementary facets of biodiversity to better identify assembly processes in response to multiple stressors. Our data suggest that stressor interactions may be less frequent in real-word streams than predicted by manipulative experiments, which can facilitate mitigation strategies.     SummaryManipulation of auditory stimuli affect the ABR evoked potentials and aid the diagnosis, particularly in auditory neuropathy patients. Some patients with auditory neuropathy lose evoked otoacoustic emissions over time; in these cases, comparing responses to rarefaction and condensation clicks, and decreasing the stimulus rate can show an extended cochlear microphonism or yield an improved electric potential record.AimTo analyze the effect of these click manipulations on the records of potentials of patients with hearing loss as a form of improving the diagnosis.Study designA clinical prospective study.Patients and Method59 patients with hearing loss underwent ABR recording using rarefaction and condensation clicks at a stimulus rate of 27.7/sec, and rarefaction clicks at a stimulus rate of 3.3/sec. The records were compared to the otoacoustic evoked emission.ResultsEight (13.53%) patients showed changes in the recorded ABR potentials as a result of manipulating the characteristics of clicks, such as extended cochlear microphonism or an improved record of electric potentials. Five patients had no otoacoustic evoked emissions.ConclusionManipulation of click stimuli can improve the topographic diagnosis of hearing loss, particularly in the group of auditory neuropathy patients with no otoacoustic evoked emissions, where usually, the diagnosis is only possible through the method described above

    Climate change and impact on renewable energies in the Azores strategic visions for sustainability

    The energy sector is the largest contributor to global greenhouse gas emissions, but could also be seriously affected by climate change, calling into question society’s current consumption patterns. In this communication, climate projections based on a set of numerical models of global circulation are used to simulate the climate until the end of the century and keep in mind the alternative scenarios of pollutant emissions. Apart from solar energy, the results for the Azores region show a negative impact on the production and consumption of renewable energies. In the regional context, this issue assumes special relevance, given the geographical constraints, such as territorial discontinuity and insularity. Based on these assumptions, measures and recommendations are pointed out for the sectors that most penalize greenhouse gas emissions, considering the energy sustainability in the Azores and the commitments and goals assumed under international agreements.info:eu-repo/semantics/publishedVersio
