Vacuolar and plasma membrane stripping and autophagic elimination of Toxoplasma gondii in primed effector macrophages
Apicomplexan protozoan pathogens avoid destruction and establish a replicative niche within host cells by forming a nonfusogenic parasitophorous vacuole (PV). Here we present evidence for lysosome-mediated degradation of Toxoplasma gondii after invasion of macrophages activated in vivo. Pathogen elimination was dependent on the interferon γ inducible-p47 GTPase, IGTP, required PI3K activity, and was preceded by PV membrane indentation, vesiculation, disruption, and, surprisingly, stripping of the parasite plasma membrane. Denuded parasites were enveloped in autophagosome-like vacuoles, which ultimately fused with lysosomes. These observations outline a series of mechanisms used by effector cells to redirect the fate of a classically nonfusogenic intracellular pathogen toward a path of immune elimination

The synthesis and crystal and molecular structure of the
bis(trifluoroacetato)(trimesitylphospine)mercuryiII) dimer are reported.
Crystals are triclinic, space group Pl with one centrosymmetric
dimer in a unit cell of dimensions a = 12.854(3), b =
= 12.877(4), c = 12.405(2) A, a = 107.34(2), (J = 118.55(2), y = 63.65

• The structure was solved by the heavy atom method and
refined by full-matrix least-squares calculation, R = 0.051 for
1656 observed reflections measured by diffractometer. The mercury
coord!nation is characterized by three strong nearly coplanar
bonds (Hg-P 2.415(5), Hg-0(12) 2.29(2), Hg-0 2.18(2) A) and two
weaker bonds (Hg-0(22) 2.95(2) and Hg-0(11)\u27 2.66(2) A). The Pmes3
ligand has a regular propeller conformation (Hg-P-C-C torsion
angles 45-49 °), enlarged C-P-C angles (mean 112.6(9) 0
), decreased Hg-P-C angles (mean 106.2(6) 0
), and a maximum cone angle of 208 °. Bridging in the solid state utilizes both oxygens of the CFsC02 moiety; the complex becomes monomeric in dichloroethane solution. Infrared, 1H, and 31P NMR data are discussed
Galaxy Harassment and the Evolution of Clusters of Galaxies
Disturbed spiral galaxies with high rates of star formation pervaded clusters
of galaxies just a few billion years ago, but nearby clusters exclude spirals
in favor of ellipticals. ``Galaxy harassment" (frequent high speed galaxy
encounters) drives the morphological transformation of galaxies in clusters,
provides fuel for quasars in subluminous hosts and leaves detectable debris
arcs. Simulated images of harassed galaxies are strikingly similar to the
distorted spirals in clusters at observed by the Hubble Space

Attribute and technology value mapping for conceptual product design phase

The new test-bed has the ability to define a technology value map to assess multiple technical options as a function of its attributes, whose precise values can be determined at a given cost. A mathematical model that incorporates a highly adaptable, data-driven and multi-attribute value approach to product specification and conceptual design is developed, novel to the concept design process. Arabidopsis TFL2/LHP1 Specifically Associates with Genes Marked by Trimethylation of Histone H3 Lysine 27
TERMINAL FLOWER 2/LIKE HETEROCHROMATIN PROTEIN 1 (TFL2/LHP1) is the only Arabidopsis protein with overall sequence similarity to the HETEROCHROMATIN PROTEIN 1 (HP1) family of metazoans and S. pombe. TFL2/LHP1 represses transcription of numerous genes, including the flowering-time genes FLOWERING LOCUS T (FT) and FLOWERING LOCUS C (FLC), as well as the floral organ identity genes AGAMOUS (AG) and APETALA 3 (AP3). These genes are also regulated by proteins of the Polycomb repressive complex 2 (PRC2), and it has been proposed that TFL2/LHP1 represents a potential stabilizing factor of PRC2 activity. Here we show by chromatin immunoprecipitation and hybridization to an Arabidopsis Chromosome 4 tiling array (ChIP-chip) that TFL2/LHP1 associates with hundreds of small domains, almost all of which correspond to genes located within euchromatin. We investigated the chromatin marks to which TFL2/LHP1 binds and show that, in vitro, TFL2/LHP1 binds to histone H3 di- or tri-methylated at lysine 9 (H3K9me2 or H3K9me3), the marks recognized by HP1, and to histone H3 trimethylated at lysine 27 (H3K27me3), the mark deposited by PRC2. However, in vivo TFL2/LHP1 association with chromatin occurs almost exclusively and co-extensively with domains marked by H3K27me3, but not H3K9me2 or -3. Moreover, the distribution of H3K27me3 is unaffected in lhp1 mutant plants, indicating that unlike PRC2 components, TFL2/LHP1 is not involved in the deposition of this mark. Rather, our data suggest that TFL2/LHP1 recognizes specifically H3K27me3 in vivo as part of a mechanism that represses the expression of many genes targeted by PRC2
Early-type galaxies at large galactocentric radii - I. Stellar kinematics and photometric properties
We present the results of a combined analysis of the kinematic and
photometric properties at large galactocentric radii of a sample of 14
low-luminosity early-type galaxies in the Fornax and Virgo clusters. From
Gemini South GMOS long-slit spectroscopic data we measure radial profiles of
the kinematic parameters v_{rot}, sigma, h_{3}, and h_{4} out to ~ 1 - 3
effective radii. Multi-band imaging data from the HST/ACS are employed to
evaluate surface brightness profiles and isophotal shape parameters of
ellipticity, position angle and discyness/boxiness. The galaxies are found to
host a cold and old stellar component which extend to the largest observed
radii and that is the dominant source of their dynamical support. The
prevalence of discy-shaped isophotes and the radial variation of their
ellipticity are signatures of a gradual gas dissipation. An early star-forming
collapse appears to be the main mechanism acting in the formation of these
objects. Major mergers are unlikely to have occurred in these galaxies. We can
not rule out a minor merging origin for these galaxies, but a comparison of our
results with model predictions of different merger categories places some
constraints on the possible merger progenitors. These merger events are
required to happen at high-redshift (i.e., z > 1), between progenitors of
different mass ratio (at least 3:1) and containing a significant amount of gas
(i.e., > 10 percent). A further scenario is that the low-luminosity galaxies
were originally late-type galaxies, whose star formation has been truncated by
removal of gas and subsequently the disc has been dynamically heated by high

Multiple reassortment events in the evolutionary history of H1N1 influenza A virus since 1918
The H1N1 subtype of influenza A virus has caused substantial morbidity and mortality in humans, first documented in the global pandemic of 1918 and continuing to the present day. Despite this disease burden, the evolutionary history of the A/H1N1 virus is not well understood, particularly whether there is a virological basis for several notable epidemics of unusual severity in the 1940s and 1950s. Using a data set of 71 representative complete genome sequences sampled between 1918 and 2006, we show that segmental reassortment has played an important role in the genomic evolution of A/H1N1 since 1918. Specifically, we demonstrate that an A/H1N1 isolate from the 1947 epidemic acquired novel PB2 and HA genes through intra-subtype reassortment, which may explain the abrupt antigenic evolution of this virus. Similarly, the 1951 influenza epidemic may also have been associated with reassortant A/H1N1 viruses. Intra-subtype reassortment therefore appears to be a more important process in the evolution and epidemiology of H1N1 influenza A virus than previously realized
