    The regulation of lysyl oxidase by bFGF and TGF-B1 in osteoblastic MC3T3-E1 Cells

Goldman School of Graduate Dentistry, 1995 (Oral Biology).Includes bibliographical references: (leaves 57-73).Lysyl oxidase catalyzes the enzymatic step that is limiting for collagen and elastin crosslinking. As a crosslinked collagenous extracellular matrix is required for bone formation, mineralization and structural integrity, regulation of lysyl oxidase in osteoblastic cells, although not previously studied, is likely to have relevance to bone synthesis and repair. The goals of this study were to determine whether lysyl oxidase, like its type I collagen substrate, is regulated by basic fibroblast growth factor (bFGF) and transforming growth factor beta-1(TGF-[beta] 1) in osteoblastic MC3T3-E1 cells. We also established the degree of posttranscriptional control by bFGF. Steady-state lysyl oxidase mRNA levels were decreased to 30% of control after 24 hours of treatment with 1 nM and 10 nM bFGF. This regulation was time-dependent. Similarly, the COL1A1 mRNA level decreased to less than 10% of control after 24 hours of treatment. Media lysyl oxidase activity decreased in accordance with the steady-state mRNA changes in bFGF-treated cultures that were refed after 24 hours of growth factor treatment. Treatment of MC3T3-E1 cells with 0.01 – 0.1 nM bFGF for 24 hours, and treatment with 1 nM bFGF for up to 12 hours resulted in a modest stimulation of lysyl oxidase gene expression and enzyme activity. Fifty per cent of the down-regulation of lysyl oxidase was shown to be posttranscriptional by measuring bFGF-dependent-differences in decay rates of lysyl oxidase mRNA in the presence of 5,6-dichloro- 1 [Beta] D-ribofuranoaylbenzimidazole. New protein synthesis was not required for the down-regulation by bFGF, but cycloheximide did increase constitutive lysyl oxidase mRNA levels 2.5-fold. GAPDH or COL1A1 mRNA levels were not affected by cycloheximide. TGF-[Beta] 1 increased the levels of steady-state lysyl oxidase mRNA, proenzyme synthesis and secretion, and enzyme activity in a dose- and time-dependent manner. A discrepancy between the synthesis of lysyl oxidase proenzyme and enzyme activity indicated that the posttranstational maturation of lysyl oxidase may limit full activation of lysyl oxidase enzyme activity. TGF-[Beta] 1-mediated effects on steady-state lysyl oxidase mRNA levels are greater than th e effects on steady-state COL1A1 mRNA levels, suggesting that lysyl oxidase is a major target for TGF-[Beta]1 in this cell culture model.     Fulminant Nonocclusive Mesenteric Ischemia Just after Hip Arthroplasty

    Nonocclusive mesenteric ischemia (NOMI) is not a rare clinical entity in intensive medicine, and it can be a consequence of several clinical or surgical situations. This pathology results from reduced intestinal microvascular blood supply associated with an acute inflammatory process, culminating with bowel necrosis. This is a case on a female patient who developed immediate postsurgical NOMI following hip arthroplasty and died. Since diagnosis of this potentially fatal condition remains a dilemma, NOMI should always be considered an eventual postoperative complication in high-risk surgical patients such as elderly individuals with previous history of nicotine abuse, congestive heart failure, and essential hypertension. The present paper highlights the importance of early diagnosis and prompt adequate treatment of NOMI in subjects with diminished cardiac output and severe abdominal pain

    This article aims to harmonize the criteria for adoption of public policies in the area of innovation within the Institutions of Science and Technology of Brazil (ICT) to a post-positivistic interpretation of legal norms regarding the topic. From Ronald Dworkin ‘s theory of Integrity and Habermas´s theory of Communicative Action applied the law, the intent is to rebuild arguably the institutional framework of innovation in the country so that it serves as normative support for the adoption of public policies that aim to mitigate the effects of an unfair planning in the sector. For this, it is argued that the adoption of public policies by Institutions of Science and Technology should distance itself from a purely utilitarian conception of innovation, that is, a conception oriented exclusively by the quantitative balance of aspects such as time and cost. Takes place, from this new perspective, a case study on innovation policy of the Federal University of Juiz de Fora, namely the Innovation Incentive Program (IIP). In conclusion, it is assumed that the policy adopted by the University should be changed according to the proposed theoretical understanding. Although this study constitutes an acidic critique on methods of adoption of public policies in the area of innovation in Brazil, it was not able to restore, due to the early stage of the research, specific criteria to propose public policies by ICT. The originality of the work consists in the problematization of the unquestioned practice, however morally and legally deficient.O presente artigo objetiva compatibilizar os critérios de adoção de políticas públicas da área de inovação no âmbito das Instituições de Ciência e Tecnologia do Brasil (ICT) com uma interpretação pós-positivista das normas jurídicas referentes ao tema. A partir da teoria da Integridade de Ronald Dworkin e da teoria da Ação Comunicativa aplicada ao Direito de Habermas, pretende-se reconstruir argumentativamente o quadro institucional da inovação no país de modo que ele sirva de suporte normativo à adoção de políticas públicas com viés ético-político tendente a atenuar os efeitos de um planejamento simbolicamente injusto no setor. Para tanto, sustenta-se que a adoção de políticas públicas por parte das Instituições de Ciência e Tecnologia deve se distanciar de uma concepção de inovação meramente utilitária, isto é, que se orienta exclusivamente pelo balanço quantitativo de aspectos como tempo e custo. Realiza-se, a partir dessa nova perspectiva, um estudo de caso sobre a política de inovação da Universidade Federal de Juiz de Fora, a saber, o Programa de Incentivo à Inovação (PII) e conclui-se que a política adotada pela autarquia federal deve ser reformulada segundo o entendimento teórico proposto. Apesar de o trabalho constituir uma crítica ácida aos métodos de adoção de políticas públicas da área de inovação no Brasil, ele não foi capaz de restabelecer, em razão da fase inicial da pesquisa, critérios específicos para a proposição de políticas públicas por parte das ICT.     Didactically describe the orbitozygomatic craniotomy made in three pieces. Method This approach was performed, from 2002 to 2011, in 49 patients admitted at Beneficência Portuguesa of São Paulo Hospital. Results Twenty-seven patients had vascular lesions and twenty-two suffered for intracranial skull base tumors. The vascular lesions varied from cavernous angiomas inside the mesencephalum, high bifurcation basilar tip aneurysms, superior cerebellar arteries aneurysms and arteriovenous malformations in the interpeduncular cistern. Skull base tumors as meningiomas, interpeduncular hamartomas and third ventricle floor gliomas were among the neoplastic lesions approached. We had no permanent injuries and minimal transient complications had occurred. Tumores da base do crânio como meningeomas, hamartomas interpedunculares e gliomas no assoalho do terceiro ventrículo estão entre as lesões neoplásicas abordadas.     Objective: To study the ideal patient’s head positioning for the anterior circulation aneurysms microsurgery. Method: We divided the study in two parts. Firstly, 10 fresh cadaveric heads were positioned and dissected in order to ideally expose the anterior circulation aneurysm sites. Afterwards, 110 patients were submitted to anterior circulation aneurysms microsurgery. During the surgery, the patient’s head was positioned accordingly to the aneurysm location and the results from the cadaveric study. The effectiveness of the position was noted. Results: We could determine mainly two patterns for head positioning for the anterior circulation aneurysms. Conclusion: The best surgical exposure is related to specific head positions.     Morphological characteristics from the insula s lobe in patients with medial temporal lobe epilepsy

    The temporal medial sclerosis (TMS) is characterized by hippocampal sclerosis in temporal and by distinghished grades of injury near to other neurological structures such as: amygdaloid nucleus, parahippocampal girus and entorhinal region. The study analyzed 40 patients with TMS and 40 people from the control cluster. All the cases were appreciated by one method for measurement of insula's cortex (E-Film) and another method to calculate the insula's volume (Neuroline). There is no variation statistical between the insula's volume and insula's measurement for the two clusters. This paper didn't show the insula's morphological variation when these two groups were compared.A esclerose medial temporal (EMT) é caracterizada pela esclerose hipocampal e diferentes graus de acometimento das estruturas vizinhas como amígdala, giro parahipocampal e córtex entorrinal. O estudo avaliou 40 indivíduos com EMT e 40 do grupo controle. Os casos foram avaliados por um método para as medidas da ínsula (E-Film) e outro método para o cálculo do volume (Neuroline). Não houve diferença estatística de alteração de volume e das medidas do lobo da ínsula nos pacientes portadores de EMT. O estudo não demonstrou alteração morfológica da ínsula quando comparado os dois grupos.63964
