    Vlasov simulation of laser-driven shock acceleration and ion turbulence

    We present a Vlasov, i.e. a kinetic Eulerian simulation study of nonlinear collisionless ion-acoustic shocks and solitons excited by an intense laser interacting with an overdense plasma. The use of the Vlasov code avoids problems with low particle statistics and allows a validation of particle-in-cell results. A simple original correction to the splitting method for the numerical integration of the Vlasov equation has been implemented in order to ensure the charge conservation in the relativistic regime. We show that the ion distribution is affected by the development of a turbulence driven by the relativistic "fast" electron bunches generated at the laser-plasma interaction surface. This leads to the onset of ion reflection at the shock front in an initially cold plasma where only soliton solutions without ion reflection are expected to propagate. We give a simple analytic model to describe the onset of the turbulence as a nonlinear coupling of the ion density with the fast electron currents, taking the pulsed nature of the relativistic electron bunches into account

    La presenza di un’importante stratificazione archeologica nella città di Milano è fatto noto da molto tempo e i cantieri che si sono succeduti, segnando la trasformazione urbanistica della città a partire dal secolo scorso, ne hanno dato ampia testimonianza, anche se spesso non accompagnata da adeguata valorizzazione. Le ricerche e gli studi hanno chiaramente individuato i capisaldi urbanistici di Mediolanum, sviluppatasi in vero e proprio centro urbano da un insediamento celtico, a partire dall’epoca augustea, quando furono eretti quegli edifici monumentali (il foro con i suoi annessi, il teatro, le mura, l’anfiteatro) necessari allo svolgimento della vita politica, religiosa, economica e sociale dell’epoca. Il panorama monumentale della città si arricchì ulteriormente alla fine del III secolo d.C. quando Mediolanum fu scelta da Massimiano quale sede imperiale, ruolo svolto fino agli inizi del V secolo d.C.: l’ampliamento delle mura, la costruzione del complesso palazzo imperiale – circo e l’edificazione di un imponente complesso termale (Fig. 2) – resero Milano uno delle più importanti città dell’impero romano d’Occidente

    Aspirin modulates LPS-induced nitric oxide release in rat glial cells

    Nitric oxide and prostaglandins are among the numerous substances released by activated glial cells. The aim of this study was to evaluate the effect of high-level aspirin on iNOS expression in cultured rat glial cells treated with lipopolysaccharide (LPS) as pathological stimulator. Using Western Blotting, we verified that aspirin enhanced LPS-induced iNOS expression and the presence of 15-deoxy-Delta(12,14)-prostaglandin (15d-PGJ(2)) suppressed this aspirin effect. However, the exposure of LPS-treated glial cells to aspirin resulted in a decrease of NO production. These results suggest that aspirin interferes with the cross-talk of prostaglandins and NO, blocking the endogenous negative control exerted by COX products on iNOS expression. On the other side, aspirin seems to act directly on iNOS reducing its activity, even if it does not completely block NO release by LPS-stimulated glial cells. Then aspirin could maintain homeostatic functions of NO, while it prevents toxic effects, corresponding to high NO concentrations. (c) 2005 Elsevier Ireland Ltd. All rights reserved

    Nutritionally enriched tomatoes (Solanum lycopersicum L.) grown with wood distillate: chemical and biological characterization for quality assessment

    : Bio-based products are nowadays useful tools able to affect the productivity and quality of conventionally cultivated crops. Several bio-based products are currently on the market; one of the newest and most promising is the wood distillate (WD) derived from the pyrolysis process of waste biomass after timber. Its foliar application has been widely investigated and shown to promote the antioxidant profile of cultivated crops. WD was used here as additive for the cultivation of tomato (Solanum lycopersicum L.) plants. The application improved quality (chemical) parameters, minerals, polyphenols, and lycopene contents of tomato fruits. The extracts of WD-treated and untreated tomatoes have been chemically and biologically characterized. The 1 H-NMR and ESI-mass spectrometry analyses of the extracts revealed the presence of different fatty acids, amino acids and sugars. In particular, the WD-treated tomatoes showed the presence of pyroglutamic acid and phloridzin derivatives, but also dihydrokaempferol, naringenin glucoside, cinnamic acid, and kaempferol-3-O-glucoside. When tested in cells, the extracts showed a promising anti-inflammatory profile in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Furthermore, the extracts displayed a slight vasorelaxant activity on rat aorta rings (either endothelium-denuded or endothelium-intact) pre-contracted with phenylephrine or potassium chloride. PRACTICAL APPLICATION: Wood distillate has been used for tomato plant growth. Tomatoes showed improved nutritional parameters, and their extracts displayed antioxidant and anti-inflammatory activities

    Data from a three-wave complete longitudinal design survey on career calling and related constructs (N = 6368)

    This dataset provides de-identified raw responses to a non-anonymous three-wave online survey with a 12-month time lag. Data collection was part of a larger project on the development of career calling in Italian college students. The first wave was collected during the fall of 2014. Participants were bachelor's or master's students enrolled in 24 different study domains and 4 different universities. Sample sizes for each wave are NT1 = 5,886, NT2 = 1,700 and NT3 = 881, 434 participants provided valid responses at all the three waves. Consent form was electronic. Dataset and codebook can be found here: https://osf.io/v56du/. The sample is mainly composed of women (63.8%, at Time 1). Participants' mean age at Time 1 was 23.37 years (SD = 5.39). The survey was in Italian and included multiple-item measures of career calling, intrinsic and extrinsic motivation, social support, engaged learning, clarity of professional identity, and quality of mentorship. Socio-demographic information and academic performance indicators are provided. The dataset is necessary to reproduce previously published results (Vianello et al., 2018) and can be useful to 1) investigate cross-cultural differences between college students from Italy and other countries; 2) investigate person-level variability in predictors and consequences of change in the variables collected over time; 3) develop and/or validate new statistical models for longitudinal data; and 4) develop and/or test original theoretical hypotheses

    Ki67 and PR in Patients Treated with CDK4/6 Inhibitors: A Real-World Experience

    CDK4/6 inhibitors (CDK4/6i) are recommended in patients with estrogen receptor (ER)-positive, HER2-negative advanced breast cancer (ABC). Up to now, no prognostic biomarkers have been identified in this setting. We retrospectively analyzed the expression of progesterone receptor (PR) and Ki67, assessed by immunohistochemistry, in 71 ABC patients treated with CDK4/6i and analyzed the impact of these markers on progression-free survival (PFS). The majority of patients 63/71 (88.7%) received palbociclib, 4 (5.6%) received ribociclib, and 4 (5.6%) received abemaciclib. A higher median value of Ki67 was observed in cases undergoing second-line treatment (p= 0.047), whereas the luminal B subtype was more prevalent (p= 0.005). In the univariate analysis of the first-line setting, luminal A subtype showed a trend towards a correlation with a longer PFS (p= 0.053). A higher continuous Ki67 value led to a significantly shorter PFS. When the interaction between pathological characteristics and line of treatment was considered, luminal B subtype showed a significantly (p= 0.043) worse outcome (Hazard Ratio (HR) 2.84; 1.03-7.82 95% Confidence Interval (CI)). PFS in patients undergoing endocrine therapy plus CDK4/6i was inversely correlated with Ki67 expression but not with PR, suggesting that tumor proliferation has a greater impact on cell cycle inhibitors combined with endocrine therapy than PR expression

    Phase II study of liposomal doxorubicin, docetaxel and trastuzumab in combination with metformin as neoadjuvant therapy for HER2-positive breast cancer

    Background:The aim of this study was to improve activity over single human epidermal growth factor receptor 2 (HER2)-blockade sequential neaodjuvant regimens for HER2-positive breast cancer, by exploiting the concomitant administration of trastuzumab, taxane and anthracycline, while restraining cardiac toxicity with use of liposomal doxorubicin, and by adding metformin, based on preliminary evidence of antitumor activity.Patients and methods:This multi-center, single-arm, two-stage phase II trial, assessed the safety and the activity of a new treatment regimen for HER2-positive, early or locally advanced breast cancer. Patients received six 21-day cycles of non-pegylated liposomal doxorubicin, 50 mg/m(2) intravenously (i.v.) on day 1, docetaxel, 30 mg/m(2) i.v. on days 2 and 9, trastuzumab, 2 mg/kg/week i.v. on days 2, 9, and 16 (with 4 mg/kg loading dose), in association with metformin 1000 mg orally twice daily. The primary endpoint was the rate of pathological complete response (pCR) in the breast and axilla (ypT0/is ypN0). A subgroup of patients performed a 3-deoxy-3-18F-fluorothymidine positron emission tomography (FLT-PET) at baseline and after one cycle.Results:Among 47 evaluable patients, there were 18 pCR [38.3%, 95% confidence interval (CI) 24.5-53.6%]. A negative estrogen-receptor status, high Ki67, and histological grade 3 were related with pCR, although only grade reached statistical significance. FLT-PET maximum standardized uptake value after one cycle was inversely related to pCR in the breast (odds ratio 0.29, 95% CI 0.06-1.30, p = 0.11). Toxicity included grade 3-4 neutropenia in 70% and febrile neutropenia in 4% of patients, grade 1-2 nausea/vomiting in 60%/38%, and grade 3 in 4%/2%, respectively, grade 1-2 diarrhea in 72%, and grade 3 in 6%. There were two cases of reversible grade 2 left-ventricular ejection-fraction decrease, and one case of sharp troponin-T increase.Conclusions:The concomitant administration of trastuzumab, liposomal doxorubicin, docetaxel, and metformin is safe and shows good activity, but does not appear to improve activity over conventional sequential regimens

    The role of the LISTANet Consortium in the European DEDIPAC-KH project

    Aim:To improve understanding of the determinants of dietary, physical activity (PA), and sedentary behaviours, the European multi-disciplinary consortium on “Determinants of Diet and Physical Activity Knowledge Hub” (DEDIPAC-KH) includes 46 consortia and organisations supported by joint programming grants from 12 countries across Europe (Lakerveld et al., 2014). Six Italian Universities (e.g., Cassino, Chieti-Pescara, Palermo, Roma Foro Italico, Roma Sapienza, and UCSC) participating in the LISTANet consortium supported by MIUR (B84G14000040008) contributed to the Thematic Area2 “Determinants of dietary, PA, and sedentary behaviours across the life course and in vulnerable groups”. In particular, the coordinator of LISTANet Prof Capranica and Prof. MacDonncha from the Irish Physical Activity and Health Consortium act as Work Package (WP) Leaders of PA determinants (WP2.2). Methods: A mix of methods has been used in identifying PA determinants by developing PA taxonomy and a European framework (EU-PAD), seven umbrella systematic literature reviews (e.g., behavioural, biological, economic, physical, policy, psychological, and socio-cultural), and identifying ongoing/recently completed European-funded projects and data sets for secondary data analyses. Results: LISTANet participated in DEDIPAC-KH meetings/seminars/courses/conferences, and organized two workshops dedicated to the EU-PAD framework and umbrella SLRs. Outcomes included internal reports, presentations to international conferences, and scientific papers submitted for publications. Conclusions: The DEDIPAC-KH project represents an excellent start in setting up a complex, cross-country, organisational structure to: 1) guide a European strategic plan for novel and multi-disciplinary research addressing the complexity of determinants of PA behaviours across the life course; and 2) identify key aspects for potential strategies and intervention programmes to implement multi-sectoral European policies in PA. Finally, the cumulated experience of LISTANet could be valuable to fully exploit effective research and actions to increase PA levels of Italian citizens
