    Architecture of coatomer: Molecular characterization of delta-COP and protein interactions within the complex

    Copyright © 2011 by The Rockefeller University Press.Coatomer is a cytosolic protein complex that forms the coat of COP I-coated transport vesicles. In our attempt to analyze the physical and functional interactions between its seven subunits (coat proteins, [COPs] alpha-zeta), we engaged in a program to clone and characterize the individual coatomer subunits. We have now cloned, sequenced, and overexpressed bovine alpha-COP, the 135-kD subunit of coatomer as well as delta-COP, the 57-kD subunit and have identified a yeast homolog of delta-COP by cDNA sequence comparison and by NH2-terminal peptide sequencing. delta-COP shows homologies to subunits of the clathrin adaptor complexes AP1 and AP2. We show that in Golgi-enriched membrane fractions, the protein is predominantly found in COP I-coated transport vesicles and in the budding regions of the Golgi membranes. A knock-out of the delta-COP gene in yeast is lethal. Immunoprecipitation, as well as analysis exploiting the two-hybrid system in a complete COP screen, showed physical interactions between alpha- and epsilon-COPs and between beta- and delta-COPs. Moreover, the two-hybrid system indicates interactions between gamma- and zeta-COPs as well as between alpha- and beta' COPs. We propose that these interactions reflect in vivo associations of those subunits and thus play a functional role in the assembly of coatomer and/or serve to maintain the molecular architecture of the complex.This work was supported by The Deutsche Forschungsgemeinschaft (SFB 352), the Human Frontier Science Program, and the Swiss National Science Foundation No. 31-43366.95

    Continuous in-line monitoring of electrolyte concentrations in extracorporeal circuits for individualization of dialysis treatment

    One objective of dialysis treatment is to normalize the blood plasma electrolytes and remove waste products such as urea and creatinine from blood. However, due to a shift in plasma osmolarity, a rapid or excessive change of the electrolytes can lead to complications like cardiovascular instability, overhydrating of cells, disequilibrium syndrome and cardiac arrhythmias. Especially for critical ill patients in intensive care unit with sepsis or multi-organ failure, any additional stress has to be avoided. Since the exchange velocity of the electrolytes mainly depends on the concentration gradients across the dialysis membrane between blood and dialysate, it can be controlled by an individualized composition of dialysate concentrations. In order to obtain a precise concentration gradient with the individualized dialysate, it is necessary to continuously monitor the plasma concentrations. However, with in-line sensors, the required hemocompatibility is often difficult to achieve. In this work, we present a concept for continuous in-line monitoring of electrolyte concentrations using ion-selective electrodes separated from the blood flow by a dialysis membrane, and therefore meeting the fluidic requirements for hemocompatibility. First investigations of hemocompatibility with reconfigured human blood show no increased hemolysis caused by the measuring system. With this concept, it is possible to continuously measure the plasma concentrations with a relative error of less than 0.5&thinsp;%.</p

With the on-going international debate about educational performance highlighting the widening gender achievement gap between girl and boy pupils, the demand for more male teachers has become prevalent in educational discourse. Concerns have frequently been raised about the underachievement of boys, with claims that the lack of male ‘role models’ in schools has an adverse effect on boys’ academic motivation and engagement. Although previous research has examined ‘teaching’ as institutional talk, men’s linguistic behaviour in the classroom remains largely ignored, especially in regard to enacting discipline.     Dynamic generation of maximally entangled photon multiplets by adiabatic passage

    The adiabatic passage scheme for quantum state synthesis, in which atomic Zeeman coherences are mapped to photon states in an optical cavity, is extended to the general case of two degenerate cavity modes with orthogonal polarization. Analytical calculations of the dressed-state structure and Monte Carlo wave-function simulations of the system dynamics show that, for a suitably chosen cavity detuning, it is possible to generate states of photon multiplets that are maximally entangled in polarization. These states display nonclassical correlations of the type described by Greenberger, Horne, and Zeilinger (GHZ). An experimental scheme to realize a GHZ measurement using coincidence detection of the photons escaping from the cavity is proposed. The correlations are found to originate in the dynamics of the adiabatic passage and persist even if cavity decay and GHZ state synthesis compete on the same time scale. Beyond entangled field states, it is also possible to generate entanglement between photons and the atom by using a different atomic transition and initial Zeeman state.Comment: 22 pages (RevTeX), including 23 postscript figures. When developmental milestones are combined with additional challenges of growing up with a chronic rheumatic musculoskeletal disease (RMD), it can increase AYA's susceptibility to psychological problems. Emotional issues in adolescence can often persist into adulthood and negatively impact future health, social, and work outcomes.     Distortion Product Otoacoustic Emissions Evoked by Tone Complexes

    Distortion product otoacoustic emissions (DPOAEs) are traditionally evoked by two-tone stimuli. In this study, emission data from Mongolian gerbils are reported that were obtained with stimuli consisting of six to 10 tones. The stimuli were constructed by replacing one of the tones of a tone pair by a narrowband multitone complex. This produced rich spectra of the ear canal sound pressure in which many of the third-order DPOAEs originated from the interaction of triplets of stimulus components. A careful choice of the stimulus frequencies ensured that none of these DPOAE components coincided. Three groups of DPOAEs are reported, two of which are closely related to DPOAEs evoked by tone pairs. The third group has no two-tone equivalent and only arises when using a multitone stimulus. We analyzed the relation between multitone-evoked DPOAEs and DPOAEs evoked by tone pairs, and explored the new degrees of freedom offered by the multitone paradigm

    The incidence of experimental smoking in school children: an 8-year follow-up of the child and adolescent behaviors in long-term evolution (CABLE) study

    <p>Abstract</p> <p>Background</p> <p>Studies have established that most regular adult smokers become addicted in their adolescent years. We investigated the incidence of and risk factors associated with initial experimental smoking among a group of school children who were followed for 8 years.</p> <p>Methods</p> <p>We used cohort data collected as part of the Child and Adolescent Behaviors in Long-term Evolution (CABLE) study, which selected nine elementary schools each from an urban area (Taipei City) and a rural area (Hsingchu county) in northern Taiwan. From 2002 to 2008, children were asked annually whether they had smoked in the previous year. An accelerated lifetime model with Weibull distribution was used to examine the factors associated with experimental smoking.</p> <p>Results</p> <p>In 2001, 2686 4<sup>th</sup>-graders participated in the study. For each year from 2002 to 2008, their incidences of trial smoking were 3.1%, 4.0%, 2.8%, 6.0%, 5.3%, 5.0% and 6.0%, respectively. There was an increase from 7<sup>th </sup>to 8<sup>th </sup>grade (6.0%). Children who were males, lived in rural areas, came from single-parent families, had parents who smoked, and had peers who smoked were more likely to try smoking earlier. The influence of parents and peers on experimental smoking demonstrated gradient effects.</p> <p>Conclusions</p> <p>This study used a cohort to examine incidence and multiple influences, including individual factors, familial factors, and community factors, on experimental smoking in adolescents. The findings fit the social ecological model, highlighting the influences of family and friends. School and community attachment were associated with experimental smoking in teenagers.</p

    Comparison of children's self-reports of depressive symptoms among different family interaction types in northern Taiwan

    <p>Abstract</p> <p>Background</p> <p>Previous research has shown that family interactions are associated with depressive symptoms in children. However, detailed classifications of family interaction types have not been studied thoroughly. This study aims to understand the types of family interactions children experience and to identify the specific types of family interactions that are associated with a higher risk of depressive symptoms in children.</p> <p>Methods</p> <p>Data used in the study was collected as part of the Child and Adolescent Behavior in Long term Evolution (CABLE) project in 2003. CABLE is a longitudinal cohort study that commenced in 2001 and collects data annually from children in Taipei city and Hsinchu county in northern Taiwan. The data analyzed in this study was that obtained from the sixth graders (aged 11 to 12 years old) in 2003. Of the 2,449 sixth graders, 51.2% were boys and 48.8% were girls. Factor analysis and cluster analysis were used to investigate the types of family interactions. One way ANOVA was used to establish the relationship between family interaction types and children's self-reports of depressive symptoms.</p> <p>Results</p> <p>Based on the results of factor analysis, the latent factors for family interactions included supporting activities, psychological control, parental discipline, behavioral supervision, and family conflict. After conducting cluster analysis using factor scores, four types of family interactions were revealed: supervised (29.66%), disciplined (13.56%), nurtured (40.96%) and conflict (15.82%). Children from the disciplined or conflict families were more likely to report depressive symptoms. Children from the nurtured families were least likely to report depressive symptoms.</p> <p>Conclusion</p> <p>Family interactions can be classified into four different types, which are related to children's self-reports of depressive symptoms. The creation of a family interaction environment that is beneficial for children's mental health is an important issue for health education and health promotion professionals.</p
