    Isolation and characterization of an antifungal agent active against human pathogenic fungi, produced by Pseudomonas fluorescens FSJ-3

    An antifungal agent, produced by Pseudomonas fiuorescens strain FSJ-3 originating from Moroccan soil (Fès), was isolated and characterized. It was extracted from Sabouraud's glucose broth culture by n-butanol and purified by gel filtration and silica gel chromatography giving yellow crystals. Its structure was assigned to 1-phenazinecarboxamide by analysing IH and 13 C-NMR and mass spectral data. It shOwed excellent activity against several species ofbacteria, yeasts and human pathogenic filamentous fungi , including Candida albicans, Cryptococcus neoformans, Aspergillus fumigatus and caused morphological modifications on Arthroderma simii hyphae. I-phenazinecarboxamide was non toxicwhen tested on rat leucocytes up to the highest concentration (200 pg/ml).Isolement et caractérisation d'une substance antifongique élaborée par Pseudomonas fluorescens FSJ-3, active contre des champignons pathogènes de l'hommeLa souche FSJ-3 de Pseudomonas fZuorescens douée d'une forte activité antifongique contre les champignons pathogènes de l'homme a été isolée à partir du sol (Fès, Maroc). La substance active, élaborée par cette souche, a été extraite du milieu de culture liquide, avec du n-butanol et purifiée par chromatographie sur gel de filtration et sur gel de silice. Le I-phénazinecarboxamide, obtenu sous forme de cristaux jaunes, a été analysé par RMN -IH et I3C et par spectrométrie de masse.     Development of polyamine levels in flower buds, flowers and young berries of Vitis vinifera L. (cv. Cabernet Sauvignon) infected by eutypiosis fungus, Eutypa lata Effects of eutypiosis on polyamines (PAs) and tyramine contents in flower buds, flowers and young berries of grapevine were studied to determine the relationship between these components and disease development. The study was performed using organs (1) from healthy vines, (2) healthy appearing organs from vines with one symptomless arm and one diseased arm, and (3) diseased organs. Free PAs, conjugated polyamines ( PAs-fpm) and wall-bound polyamines (PAs-mm) were analysed in Vitis vinifera L. cv. Cabernet Sauvignon which is sensitive to eutypiosis. An increase of PAs-mm and a decrease of free PAs were observed in diseased organs; particularly putrescine and diaminopropane were concerned. On the other hand these substances were not affected by eutypiosis in the healthy appearing organs. Alterations of free PAs and PAs-mm in diseased organs were correlated with alteration of the flowering process. In contrast, the PAsfpm levels decreased in diseased organs in response to eutypiosis whereas they increased in healthy appearing organs. This group of PAs seems to play an important role in the response to Eutypa lata attacks to prevent the expression of symptoms. Accumulation of the conjugated tyramine in diseased and healthy appearing organs as a response to eutypiosis might be used as marker of Eutypa lata infection

    Detection of Diarrheic Shellfish Poisoning and Azaspiracid Toxins in Moroccan Mussels: Comparison of the LC-MS Method with the Commercial Immunoassay Kit

    Diarrheic shellfish poisoning (DSP) is a recurrent gastrointestinal illness in Morocco, resulting from consumption of contaminated shellfish. In order to develop a rapid and reliable technique for toxins detection, we have compared the results obtained by a commercial immunoassay-“DSP-Check” kit” with those obtained by LC-MS. Both techniques are capable of detecting the toxins in the whole flesh extract which was subjected to prior alkaline hydrolysis in order to detect simultaneously the esterified and non esterified toxin forms. The LC-MS method was found to be able to detect a high level of okadaic acid (OA), low level of dinophysistoxin-2 (DTX2), and surprisingly, traces of azaspiracids 2 (AZA2) in mussels. This is the first report of a survey carried out for azaspiracid (AZP) contamination of shellfish harvested in the coastal areas of Morocco. The “DSP-Check” kit was found to detect quantitatively DSP toxins in all contaminated samples containing only OA, provided that the parent toxins were within the range of detection and was not in an ester form. A good correlation was observed between the two methods when appropriate dilutions were performed. The immunoassay kit appeared to be more sensitive, specific and faster than LC-MS for determination of DSP in total shellfish extract
