    High-throughput chromatin accessibility profiling at single-cell resolution.

    Here we develop a high-throughput single-cell ATAC-seq (assay for transposition of accessible chromatin) method to measure physical access to DNA in whole cells. Our approach integrates fluorescence imaging and addressable reagent deposition across a massively parallel (5184) nano-well array, yielding a nearly 20-fold improvement in throughput (up to ~1800 cells/chip, 4-5ā€‰h on-chip processing time) and library preparationĀ cost (~81Ā¢ per cell) compared to prior microfluidic implementations. We apply this method to measure regulatory variation in peripheral blood mononuclear cells (PBMCs) and show robust, de novo clustering of single cells by hematopoietic cell type

    A Dynamic View of Response to Aphasia: The Emergence of Compensatory Adaptations

    This presentation employs two qualitative research methodologies, conversation analysis (CA) and phenomenology, to describe the evolution of eleven compensatory strategies within the conversational performance of a particular individual with aphasia. By comparing conversational data prior to the individualā€™s stroke with data samples 6 months, 12 months, and 24 months post onset, the evolution of his compensatory adaptations to aphasia are documented and analyzed. This provides a dynamic perspective on his conversational compensations and their utility. Theoretical and clinical implications are discussed

    Methylenetetrahydrofolate Reductase Gene Variant (MTHFR C677T) and Migraine: A Case Control Study and Meta-analysis

    Extent: 9p.Background: Migraine is a common disorder that often coexists with depression. While a functional polymorphism in methyleneterahydrofolate reductase gene (MTHFR C677T) has been implicated in depression; the evidence to support an association of MTHFR with migraine has been inconclusive. We aim to investigate the effect of this variant on propensity for migraine and to perform a systematic review and meta-analysis of studies of MTHFR and migraine to date. Methods: Individuals with migraine (n = 447) were selected from the Depression Case Control (DeCC) study to investigate the association between migraine and MTHFR C677T single nucleotide polymorphism (SNP) rs1801133 using an additive model compared to non-migraineurs adjusting for depression status. A meta-analysis was performed and included 15 studies of MTHFR and migraine. Results: MTHFR C677T polymorphism was associated with migraine with aura (MA) (OR 1.31, 95% CI 1.01-1.70, p = 0.039) that remained significant after adjusting for age, sex and depression status. A meta-analysis of 15 case-control studies showed that T allele homozygosity is significantly associated with MA (OR = 1.42; 95% CI, 1.10-1.82) and total migraine (OR = 1.37; 95% CI, 1.07-1.76), but not migraine without aura (OR = 1.16; 95% CI, 0.36-3.76). In studies of non-Caucasian population, the TT genotype was associated with total migraine (OR= 3.46; 95% CI, 1.22-9.82), whereas in studies of Caucasians this variant was associated with MA only (OR = 1.28; 95% CI, 1.002-1.63). Conclusions: MTHFR C677T is associated with MA in individuals selected for depression study.     This project is funded by the Department of Health and Human Services and seeks to add to existing evidence of an underlying theory of change whereby effective professional development can increase the quality of Head Start classrooms, family support can improve family stability and school engagement, and that the interacting effects of this classroom quality and family influence promote child school readiness. The availability of strong and nuanced coaching data and robust, frequent CLASS observational data will enhance the existing body of knowledge. In total, being able to address the full model and a theory of change that incorporates site level, classroom level quality, family factors, and student outcomes, with improved high-integrity data is a significant contribution to the existing literature

    Lessons from Crisis Recovery in Schools: How Hurricanes Impacted Schools, Families and the Community

    This article examines school and school district-level efforts to reopen schools after significant damage from hurricanes. Through an empirical, qualitative research design, four themes emerged as critical to the hurricane recovery process: the importance of communication, resolving tension, coordinating with other services and learning from the past. School administrators must proactively plan for catastrophic events and balance the need for open communication within and outside the school district; to resolve tensions; and to coordinate with other public and private efforts to restore the community. Finally, they should use each experience as a learning opportunity
