    Effects of manganese, 2,5-xylidine, veratryl alcohol and tween 80 on the production of ligninolytic enzymes by Ceriporiopsis subvermispora

    The effects of adding manganese, 2,5-xylidine, veratryl alcohol and Tween 80 in a culture medium used for the production of ligninolytic enzymes by polyurethane foam-immobilized Ceriporiopsis subvermispora were studied. While 11 ppm Mn2+ promoted the highest maximum activity of  manganese peroxidase (108.0 ± 43.3 U/L, in the 6th day of cultivation), the medium without manganese led to the highest maximum activity of laccase (15.5 ± 2.1 U/L, in the 12th day of cultivation). By supplementing the medium containing 11 ppm Mn2+ with 1.0 mM 2,5-xylidine, it was possible to improve the maximum activity of laccase to 21.5 ± 4.9 U/L. The supplementation of the medium containing 11 ppm Mn2+ with 1.0mM veratryl alcohol, in turn, led to an apparent second peak of MnP activity (110.0 ± 1.4 U/L, in the 24th day of cultivation; compared to 147.5 ± 60.1 U/L, in the 9th day of cultivation). When the medium containing 11 ppm Mn2+ and 1.0 mM 2,5-xylidine was supplemented with 0.05% (v/v) Tween 80, the maximum activities of Lac and MnP reached 53.3 ± 17.7 U/L (21st day of cultivation) and 174.8 ± 1.4 U/L (9th day of cultivation),  respectively. During the cultivations, the exhaustion of glucose in the medium promoted nutritional stress, which, in turn, led to cell autolysis; reflected by an apparent reduction in the concentration of mycelium, and by an increase in the concentration of ammonium. The concentrations of extracellular proteins increased throughout the cultivations; such  concentrations, however, did not generally exhibit good correlations with the measured enzyme activities.Key words: Ceriporiopsis subvermispora, manganese, 2,5-xylidine, veratryl alcohol, Tween 80; manganese peroxidase, laccase

    Callous-Unemotional Traits Moderate Anticipated Guilt and Wrongness Judgments to Everyday Moral Transgressions in Adolescents.

    Callous-unemotional (CU) traits observed during childhood and adolescence are thought to be precursors of psychopathic traits in adulthood. Adults with high levels of psychopathic traits typically present antisocial behavior. Such behavior can be indicative of atypical moral processing. Evidence suggests that moral dysfunction in these individuals may stem from a disruption of affective components of moral processing rather than from an inability to compute moral judgments per se. No study to date has tested if the dissociation between affective and cognitive dimensions of moral processing linked to psychopathic traits in adulthood is also linked to CU traits during development. Here, 47 typically developing adolescents with varying levels of CU traits completed a novel, animated cartoon task depicting everyday moral transgressions and indicated how they would feel in such situations and how morally wrong the situations were. Adolescents with higher CU traits reported reduced anticipated guilt and wrongness appraisals of the transgressions. However, our key finding was a significant interaction between CU traits and anticipated guilt in predicting wrongness judgments. The strength of the association between anticipated guilt and wrongness judgement was significantly weaker for those with higher levels of CU traits. This evidence extends our knowledge on the cognitive-affective processing deficits that may underlie moral dysfunction in youth who are at heightened risk for antisocial behavior and psychopathy in adulthood. Future longitudinal research is required to elucidate whether there is an increased dissociation between different components of moral processing from adolescence to adulthood for those with high psychopathic traits

    Accidental Jorge Lobo's disease in a worker dealing with Lacazia loboi infected mice: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Jorge Lobo's disease (Lacaziosis) is a subcutaneous infection of humans living in the Amazon region of Latin America, and in dolphins inhabiting the east coastal areas of the United States. The disease mainly affects people from rural areas living or working in close contact with vegetation and aquatic environments. Most patients refer having developed lesions after accidental trauma with plant thorns or insect bites. Inter-human transmission has never been confirmed suggesting that <it>Lacazia loboi </it>is acquired from environmental propagules.</p> <p>Case presentation</p> <p>We report the case of a 41-year-old woman from SĂŁo Paulo, Brazil, a non-endemic area of Jorge Lobo's disease, with <it>L. loboi </it>skin infection most likely accidentally acquired while manipulating experimentally infected mice in the laboratory.</p> <p>Conclusion</p> <p>Because many patients with Jorge Lobo's disease do not recall accidental skin trauma before their infections, the possibility of accidentally acquired Jorge Lobo's disease through unnoticed broken skin should be considered during the clinical investigation of nodular skin diseases in people who have contact with the fungus or who live in endemic areas. This is the second report of animal to human transmission of this disease.</p

    Towards the reconstruction of integrated genome-scale models of metabolism and gene expression

    The reconstruction of integrated genome-scale models of metabolism and gene expression has been a challenge for a while now. In fact, various methods that allow integrating reconstructions of Transcriptional Regulatory Networks, gene expression data or both into Genome-Scale Metabolic Models have been proposed. Several of these methods are surveyed in this article, which allowed identifying their strengths and weaknesses concerning the reconstruction of integrated models for multiple prokaryotic organisms.
