Cannabinoid type 2 receptors (CB2), a key member of the endocannabinoid system have recently emerged as a crucial therapeutic target for cardiovascular diseases (CVDs). Downregulation of CB2 receptors has been witnessed in various cancers, neurological and cardiovascular disorders. Thus, the activation of CB2 receptors may protect against ISO-induced myocardial infarction (MI) in rats. The present study investigates the cardioprotective effect of a selective cannabinoid type 2 receptor agonist β-caryophyllene (BCP), a dietary phytocannabinoid, and a natural bicyclic sesquiterpene against isoproterenol (ISO)-induced MI in rats. Male albino Wistar rats were pre-and co-treated with β-caryophyllene (50 mg/kg, orally) twice daily for 10 days along with the subcutaneous injection of ISO (85 mg/kg) at an interval of 24 h for two days (9th and 10th day). AM630 (1 mg/kg), a CB2 receptor antagonist was injected intraperitoneally as a pharmacological challenge prior to BCP treatment to demonstrate CB2 receptor-mediated cardioprotective mechanisms of BCP. ISO-induced MI showed a significant decline in cardiac function, elevated levels of serum cardiac marker enzymes, and enhanced oxidative stress markers with increased lipid peroxidation. Isoproterenol also induced pro-inflammatory cytokines release following activation of the nuclear factor kappa-B. Furthermore, a significant rise was also observed in the levels of inflammatory mediators, namely cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) in ISO-challenged rats. Additionally, Isoproterenol also increased the expression of pro-apoptotic (Bax, and active caspase-3) proteins along with the decreased expression of the anti- apoptotic protein, Bcl2, and Bcl-xL in the myocardium. β-caryophyllene treatment resulted in significant protective effects on all biochemical and molecular parameters analyzed. Histopathological and ultrastructural evidence was found in line with these findings. Treatment with AM630, a potent CB2 receptor antagonist abrogates protective effects\ of BCP on all the biochemical and molecular parameters analyzed in ISO induced MI in rats. Thus, this study revealed that BCP protects the myocardium against ISO-induced MI by attenuating oxidative stress, apoptosis, and inflammation and the underlying mechanism of this protection is the activation of CB2 receptors. CB2 receptor-selective compounds may provide a new potential class of cardioprotective drugs. The pharmacophore of these compounds could be used for synthesizing leads in drug discovery and development

Protocol for a systematic review of the effects of schools and school-environment interventions on health: evidence mapping and syntheses

Background: Schools may have important effects on students’ and staff’s health. Rather than treating schools merely as sites for health education, ‘school-environment’ interventions treat schools as settings which influence health. Evidence concerning the effects of such interventions has not been recently synthesised. Methods/design: Systematic review aiming to map and synthesise evidence on what theories and conceptual frameworks are most commonly used to inform school-environment interventions or explain school-level influences on health; what effects school-environment interventions have on health/health inequalities; how feasible and acceptable are school-environment interventions; what effects other school-level factors have on health; and through what processes school-level influences affect health. We will examine interventions aiming to promote health by modifying schools’ physical, social or cultural environment via actions focused on school policies and practices relating to education, pastoral care and other aspects of schools beyond merely providing health education. Participants are staff and students age 4-18 years. We will review published research unrestricted by language, year or source. Searching will involve electronic databases including Embase, ERIC, PubMed, PsycInfo and Social Science Citation Index using natural-language phrases plus reference/citation checking. Stage 1 will map studies descriptively by focus and methods. Stage 2 will involve additional inclusion criteria, quality assessment and data extraction undertaken by two reviewers in parallel. Evidence will be synthesised narratively and statistically where appropriate (undertaking subgroup analyses and meta-regression and where no significant heterogeneity of effect sizes is found, pooling these to calculate a final effect size). Discussion: We anticipate: finding a large number of studies missed by previous reviews; that non-intervention studies of school effects examine a greater breadth of determinants than are addressed by intervention studies; and that intervention effect estimates are greater than for school-based health curriculum interventions without school-environment components

Intergenerational patterns of violence against children and perceptions of childhood abuse have not been studied a lot among general population or fathers. The objective of the present study is to determine the link between frequencies of childhood severe physical violence and its perceived legitimacy (first), and minor physical and psychological violence against children in actual family (second) in a sample of 204 fathers from Quebec. Results indicate a significant link between frequencies of childhood severe physical violence and its perceived legitimacy, and violence against children in actual family. Fathers who have often or very often lived childhood severe physical violence have children who suffer more psychological violence than fathers who have lived it sometimes. The more fathers think that childhood violence was deserved, the more their children suffer psychological violence. Finally, children of fathers who think that childhood severe physical violence was sometimes deserved suffer less minor physical violence than those of fathers who think that it was often or very often deserved. Implications for research and practice are discussed

    Get PDF
Objective: The aim of this study was to evaluate the clinical presentation of aplastic anemia as well as to assess the efficacy of Cyclosporin -A in patients with aplastic anemia. Methodology: This is a hospital based interventional study. During the three year study period, 44 children were enrolled. Mean age was 9.3 years and there was a male predominance. Results: Most common clinical presentation was anemia and bleeding. Four children died before therapy was started. Cyclosporin-A was started in 40 patients. Eleven patients died before completion of therapy and three patients were lost to follow-up. Out of 26 patients who completed therapy, 11 were cured and 9 were responders while 6 were non responders according to the selected criteria. Conclusion: In developing third world countries like Pakistan majority of the patients with aplastic anemia cannot afford BMT. Alternative modalities of treatment must therefore be looked into. Cyclosporin-A seems to be a reasonable therapeutic option in such cases

Mysticism in the poetry of Emily Dickinson: a theological interpretation

    Get PDF
This paper is an attempt to analyze the poetry of Miss Emily Dickinson (1830-1886) in order to reveal the extent of mysticism in it, and it focuses on the concept of “theology.” Mysticism involves a deep, almost obsessive, concern with such problems as death, the existence of the soul, immortality, the existence of God and heaven, salvation or redemption, etc. The critical approach was used to analyze some of Dickinson’s major poems. A glance at her poetry reveals that it shows an extreme preoccupation with the effect of death and explores various themes such as the nature of the soul, the problem of immortality, the possibility of faith and the reality of God. The researchers also tried to reveal the internal and external influences that shaped Dickinson’s poetry. The paper concluded that the theme of death was inexhaustible for her. If her poetry seldom became “lyrical,” seldom departed from the colorless sobriety of its bare iambics and toneless assonance, it did so most of all when the subject was death. Although Dickinson’s poetry contains some mystical elements, mystical poetry, in the traditional sense, at least, is not her special poetic gift

Tributyltin stimulates synthesis of interferon gamma and tumor necrosis factor alpha in human lymphocytes

Tributyltin (TBT) is found in human blood and other tissues and thus is of considerable concern as to its effects on human health. Previous studies have demonstrated that TBT has detrimental effects on immune function. Recently, we found that exposures to TBT caused increased secretion of two important proinflammatory cytokines, tumor necrosis factor alpha (TNFα) and interferon gamma (IFNγ). Elevation of either of these cytokines has the potential to cause chronic inflammation, which is an important factor in a number of diseases including cancer. The current study examined the mechanism of TBT-induced elevations of TNFα and IFNγ secretion and found that the p38 mitogen-activated protein kinase pathway was essential to the ability of TBT to stimulate secretion. Additionally, this study demonstrated that increased secretion of these cytokines was due to TBT-induced increases in their overall synthesis, rather than simply being due to an increase in the release of already formed proteins. The TBT-induced increases in synthesis were evident within 6 hours of exposure. The p38 mitogen-activated protein kinase pathway is also necessary for the TBT-induced increases in both TNFα and IFNγ synthesis. The role of increased transcription of TNFα and IFNγ mRNA in response to TBT exposures as a possible explanation for the increased synthesis of these cytokines was also examined. It was found that increased mRNA levels did not appear to explain fully the increases in either TNFα or IFNγ synthesis. Thus, TBT is able to increase secretion of two important proinflammatory cytokines by increasing their synthesis

Dendritic cell and co-stimulatory molecule targeted therapy for autoimmune diseases: a review of the newly implemented strategies

    Get PDF
Dendritic cells (DCs) play an important role in the formation of the immune response, and they are involved in the pathogenesis of autoimmune diseases. Targeting DCs has thus emerged as a new therapeutic modality in the management of inflammatory and autoimmune diseases. DCs can be manipulated ex vivo and then injected back into humans to suppress the immune response. They can also be manipulated in vivo by delivering specific molecules into the DCs. Co-stimulatory molecules that shape DCs interaction with T cells can also be targeted to suppress immunity. This review tackles the latest advances in each of the 3 presented approaches
