Kelengkapan Pengisian Rekam Medis Rawat Inap Pada Pasien JKN di Rumah Sakit Universitas Andalas Tahun 2022
Tujuan Penelitian
Kelengkapan pengisian rekam medis di Rumah Sakit Universitas Andalas belum memenuhi SPM. Tujuan penelitian ini untuk mengetahui gambaran terkait kelengkapan pengisian rekam medis rawat inap pada pasien JKN di Rumah Sakit Universitas Andalas Padang tahun 2022.
Metode
Penelitian ini adalah mix-method secara Sequential Explanotary Design. Sampel penelitian terdiri dari 95 status rekam medis rawat inap Eboni yang dipilih menggunakan sistem Random Sampling dan dianalisis dengan metode analisis univariat. Sedangkan informan dipilih secara purposive sampling, dan dianalisis dengan metode analisis isi.
Hasil
Kelengkapan rekam medis rawat inap pada pasien JKN sebesar 94,6% dengan persentase kelengkapan terendah pada komponen laporan penting yaitu sebesar 88,3%. Hasil wawancara mendalam pada komponen input: tenaga kerja belum cukup, dana sudah disediakan, metode sudah sesuai alur yang ditetapkan, material sudah cukup, namun sarana dan prasana masih kurang. Komponen process: pendaftaran pasien sudah berjalan sesuai alur, pengisian rekam medis masih belum lengkap, analisis rekam medis belum berjalan rutin sehingga pelaporan juga belum rutin. Komponen output: kelengkapan pengisian rekam medis rawat inap pasien JKN masih tidak lengkap.
Kesimpulan
Pengisian rekam medis rawat inap pada pasien JKN belum mencapai SPM baik dari rumah sakit Universitas Andalas maupun Nasional yaitu 100%. Diharapkan tenaga profesional patuh dalam pengisian rekam medis berdasarkan aturan yang telah ditetapkan
Functional Redistribution of Hippocampal Cannabinoid Cb1 Receptors in the Rat Pilocarpine Model of Acquired Epilepsy
Cannabinoids, such as the marijuana derivative Î9-THC, are known to have CBl receptor-mediated anticonvulsant effects in several animal models of seizures and epilepsy, including the rat pilocarpine model of acquired epilepsy. However, the distribution of CBl receptor expression and function in brains of epileptic rats has not been characterized. Therefore, this dissertation was initiated to evaluate the effect of epileptogenesis on the distribution and function of the endogenous CBI receptor system in the rat pilocarpine model, a well-established model of acquired temporal lobe epilepsy. Using immunohistochemistry, we demonstrated that chronically epileptic rats exhibit a unique, long-term, and specific redistribution of hippocampal CBl receptors when compared to controls, with concurrent layer-specific increases and decreases in CBl receptor expression within the hippocampus. In addition, studies in this dissertation demonstrated using [3H] WIN55,212-2 autoradiography and agonist-stimulated [35S]GTPÎłS autoradiography that this CBl receptor-specific reorganization results in corresponding functional changes manifested by alterations in CBl receptor binding and G-protein activation. These regionally selective changes were dependent on NMDA receptor activation during the initial insult of pilocarpine-induced status epilepticus (SE), and were independent of seizure suppression produced with phenobarbital administration in epileptic rats. Furthermore, time-course studies utilizing these techniques demonstrate that within a week following SE, a widespread loss of CBl receptor expression and function occurs throughout the hippocampus. The subsequent redistribution of CBl receptors that occurs temporally correlates with the emergence of spontaneous recurrent seizures, and is still observed up to 1 year following SE. Overall, the reorganization of cannabinoid receptors in epilepsy implicates the endocannabinoid system in modulating neuroexcitability in the epileptic state. This CBl receptor redistribution represents an essentially permanent neuronal plasticity change associated with epileptogenesis, and could account for the anticonvulsant effect of cannabinoids observed in this model
Knowledge, Attitudes, and Practices Associated with Brucellosis in Livestock Owners in Jordan
We evaluated livestock owners' knowledge, attitudes, and practices regarding brucellosis in Jordan. A questionnaire was administered and biological samples were examined to verify the serological status of animals. Seroprevalence estimates indicated that 18.1% (95% CI: 11â25.3) of cattle herds and 34.3% (95% CI: 28.4â40.4) of small ruminant flocks were seropositive. The results showed that 100% of the interviewed livestock keepers were aware of brucellosis: 87% indicated a high risk of infection if unpasteurized milk is consumed and 75% indicated a high risk if unpasteurized dairy products are consumed. Awareness of the risk of infection through direct contact with fetal membranes or via physical contact with infected livestock is considerably lower, 19% and 13%, respectively. These knowledge gaps manifest in a high frequency of high-risk practices such as assisting in animal parturition (62%), disposing aborted fetuses without protective gloves (71.2%) or masks (65%), and not boiling milk before preparation of dairy products (60%). When brucellosis is suspected, basic hygiene practices are often disregarded and suspect animals are freely traded. Public health education should be enhanced as the disease is likely to remain endemic in the ruminant reservoir as long as a suitable compensation program is not established and trust on available vaccines is regained

AAV Vector-Mediated Overexpression of CB1 Cannabinoid Receptor in Pyramidal Neurons of the Hippocampus Protects against Seizure-Induced Excitoxicity
The CB1 cannabinoid receptor is the most abundant G-protein coupled receptor in the brain and a key regulator of neuronal excitability. There is strong evidence that CB1 receptor on glutamatergic hippocampal neurons is beneficial to alleviate epileptiform seizures in mouse and man. Therefore, we hypothesized that experimentally increased CB1 gene dosage in principal neurons would have therapeutic effects in kainic acid (KA)-induced hippocampal pathogenesis. Here, we show that virus-mediated conditional overexpression of CB1 receptor in pyramidal and mossy cells of the hippocampus is neuroprotective and moderates convulsions in the acute KA seizure model in mice. We introduce a recombinant adeno-associated virus (AAV) genome with a short stop element flanked by loxP sites, for highly efficient attenuation of transgene expression on the transcriptional level. The presence of Cre-recombinase is strictly necessary for expression of reporter proteins or CB1 receptor in vitro and in vivo. Transgenic CB1 receptor immunoreactivity is targeted to glutamatergic neurons after stereotaxic delivery of AAV to the dorsal hippocampus of the driver mice NEX-cre. Increased CB1 receptor protein levels in hippocampal lysates of AAV-treated Cre-mice is paralleled by enhanced cannabinoid-induced G-protein activation. KA-induced seizure severity and mortality is reduced in CB1 receptor overexpressors compared with AAV-treated control animals. Neuronal damage in the hippocampal CA3 field is specifically absent from AAV-treated Cre-transgenics, but evident throughout cortical areas of both treatment groups. Our data provide further evidence for a role of increased CB1 signaling in pyramidal hippocampal neurons as a safeguard against the adverse effects of excessive excitatory network activity
Coordinating Environmental Genomics and Geochemistry Reveals Metabolic Transitions in a Hot Spring Ecosystem
We have constructed a conceptual model of biogeochemical cycles and metabolic and microbial community shifts within a hot spring ecosystem via coordinated analysis of the âBison Poolâ (BP) Environmental Genome and a complementary contextual geochemical dataset of âŒ75 geochemical parameters. 2,321 16S rRNA clones and 470 megabases of environmental sequence data were produced from biofilms at five sites along the outflow of BP, an alkaline hot spring in Sentinel Meadow (Lower Geyser Basin) of Yellowstone National Park. This channel acts as a >22 m gradient of decreasing temperature, increasing dissolved oxygen, and changing availability of biologically important chemical species, such as those containing nitrogen and sulfur. Microbial life at BP transitions from a 92°C chemotrophic streamer biofilm community in the BP source pool to a 56°C phototrophic mat community. We improved automated annotation of the BP environmental genomes using BLAST-based Markov clustering. We have also assigned environmental genome sequences to individual microbial community members by complementing traditional homology-based assignment with nucleotide word-usage algorithms, allowing more than 70% of all reads to be assigned to source organisms. This assignment yields high genome coverage in dominant community members, facilitating reconstruction of nearly complete metabolic profiles and in-depth analysis of the relation between geochemical and metabolic changes along the outflow. We show that changes in environmental conditions and energy availability are associated with dramatic shifts in microbial communities and metabolic function. We have also identified an organism constituting a novel phylum in a metabolic âtransitionâ community, located physically between the chemotroph- and phototroph-dominated sites. The complementary analysis of biogeochemical and environmental genomic data from BP has allowed us to build ecosystem-based conceptual models for this hot spring, reconstructing whole metabolic networks in order to illuminate community roles in shaping and responding to geochemical variability

Tenacity of highly pathogenic agents in foods and rapid detection methods for Brucella
Zur Bewertung des Risikos im Fall einer absichtlichen Ausbringung von
Brucellen in die Lebensmittelkette sind Kenntnisse zur TenazitÀt des Erregers
in relevanten Lebensmittelmatrizes erforderlich. Daher wurde das Ăberleben von
Brucella abortus 1119-3 in Rohmilch sowie in kommerziell erhÀltlichen
Lebensmitteln wie H-Milch, Vorzugsmilch, Joghurt und stillem Mineralwasser bei
handelsĂŒblichen Lagerungstemperaturen untersucht. Besonders lange ĂŒberlebten
B. abortus 1119-3 und B. melitensis 16M in H-Milch, in der die Zellzahl
anstieg und bis zum Ende des Versuchs nach 88 Tagen eine Konzentration von
ĂŒber 10^8 KbE/ml aufwies. In stillem Mineralwasser war B. abortus 1119-3 ĂŒber
einen Zeitraum von 60 Tagen kulturell nachweisbar, wobei die Zellzahl
kontinuierlich abnahm. Bis zum Ende der Mindesthaltbarkeit des Produktes
(jeweils vier Tage) ĂŒberlebte der genannte Stamm in Vorzugsmilch und Rohmilch.
In Joghurt konnten hingegen lebende Zellen nur zwei (1,5 % Fett), vier (3,5 %
Fett) bzw. einen Tag (10 % Fett) nachgewiesen werden, wobei der schnelle
RĂŒckgang der Zellkonzentration wahrscheinlich durch den niedrigen pH-Wert von
4,2 verursacht wurde. Bei einer absichtlichen Kontamination wĂŒrden Brucellen
fĂŒr zwei Wochen (stilles Mineralwasser) bzw. ĂŒber Monate (H Milch) in einer
hohen Konzentration in den Lebensmitteln ĂŒberleben und beim Verzehr mit hoher
Wahrscheinlichkeit zu einer Brucellose fĂŒhren. FĂŒr die Erkennung und
Typisierung von Brucellen im Fall der absichtlichen Kontamination von
Lebensmitteln mĂŒssen geeignete Schnellmethoden zur VerfĂŒgung stehen. Deshalb
wurden sechs verschiedene Methoden in Kombination mit der PCR auf ihre
Effizienz zur Extraktion und zum Nachweis von Brucella-DNA aus relevanten
Lebensmittelmatrizes untersucht. FĂŒr stilles Mineralwasser und physiologische
Kochsalzlösung erwies sich das QIAamp DNA Mini Kit (Qiagen) mit einer
Nachweisgrenze von 5 x 10^1 KbE/ml am sensitivsten. FĂŒr Rohmilch eigneten sich
sowohl der Kit von Qiagen als auch die Phenol-Chloroform-Extraktion, wobei
letztere mit höherer ZuverlÀssigkeit bis zu 5 x 10^1 KbE/ml nachwies. Die
Nachweisgrenze fĂŒr Brucellen in Joghurt (10 % Fett) lag bei Anwendung der
Phenol-Chloroform-Extraktion mit 1 x 10^2 KbE/ml niedriger als mit dem QIAamp
DNA Mini Kit (1 x 10^4 KbE/ml). Mit der fĂŒr Milchprodukte tauglichen Phenol-
Chloroform-Extraktion wurden bis zu 5 x 10^2 KbE/g in Camembert und in
ZiegenfrischkÀse nachgewiesen. Die DNA-Extraktion aus WeichkÀse erfordert eine
Homogenisation nach VerdĂŒnnung mit FlĂŒssigkeit und fĂŒhrt deshalb zu einer
Abnahme der SensitivitÀt. Im Gegensatz zur mehrere Tage dauernden Kultivierung
von Brucellen auf NĂ€hrmedien, fĂŒr die ein Labor der Sicherheitsstufe 3
erforderlich ist, kann durch die Kombination aus DNA-Extraktion und Real-Time
PCR ein Nachweis innerhalb eines halben (QIAamp DNA Mini Kit) bzw. innerhalb
eines Arbeitstages (Phenol-Chloroform-Extraktion) in einem Labor der
Sicherheitsstufe 2 erfolgen. Eine Erhöhung der SensitivitÀt zum Nachweis der
Brucella-DNA war allerdings mittels Real-Time PCR im Vergleich zur
herkömmlichen PCR nicht möglich. Mit dem Ziel, die Nachweisgrenzen fĂŒr
Brucella-DNA insbesondere in Milchprodukten zu optimieren, wurde die
immunomagnetische Separation (IMS) als spezielle Methode fĂŒr die
Voranreicherung und Aufreinigung der DNA untersucht. Zu diesem Zweck wurden
tosylaktivierte immunomagnetische Partikel eingesetzt, die mit einem
Brucellose-Vollserum bzw. einem monoklonalen anti-Brucella-Antikörper
beschichtet waren. Trotz Optimierung der IMS in AbhÀngigkeit verschiedener
Parameter, wie Probenvolumen, Inkubationszeit und temperatur lag die Grenze
zum Nachweis der Brucella-DNA bei 5 x 10^3 KbE/g fĂŒr ZiegenfrischkĂ€se, bei 1 x
10^3 KbE/ml fĂŒr Rohmilch und stilles Mineralwasser, bei 8 x 10^4 KbE/ml fĂŒr
Joghurt und bei 5 x 10^4 KbE/g fĂŒr Camembert und war deshalb weniger sensitiv
als die vorgenannten Extraktionsmethoden.In case of a deliberate contamination of the food chain with brucellae it is
important to know the survival of the bacteria in relevant food matrices.
Therefore, the survival of Brucella abortus 1119-3 was investigated in raw
milk as well as in commercially available foods like UHT-milk, certified raw
milk, yogurt and still mineral water at general storage conditions. B. abortus
1119-3 and B. melitensis 16M survived longest in UHT-milk in which the total
cell count multiplied to more than 10^8 cfu/ml at the end of the productâs
shelf life after 88 days. In still mineral water viable cells of B. abortus
1119-3 could be detected for 60 days in culture. Until this day the total cell
count declined continuously. In raw milk and certified raw milk this strain
survived until the end of the productâs shelf life (four days, respectively).
In yogurt viable cells could be detected for two days (1.5 % fat), four days
(3.5 % fat) or one day (10.0 % fat), most probably due to the low pH of 4.2.
In case of a deliberate contamination brucellae would survive at a high
concentration for two weeks (still mineral water) or for months (UHT-milk) so
that ingestion of the food would most probably lead to brucellosis. Suitable
methods for the fast detection and typing of brucellae should be available in
case of deliberate contamination of foods. Therefore, six different methods
used in combination with PCR were examined regarding their efficiency to
extract and detect Brucella-DNA from relevant food matrices. For extraction
from still mineral water and physiological saline, the QIAamp DNA Mini Kit
(Qiagen) revealed the best detection limit of 5 x 10^1 cfu/ml. For raw milk
both the kit and the phenol chloroform extraction were best suited of which
the latter was more reliable at a detection limit of 5 x 10^1 cfu/ml. The
detection limit for brucellae in yogurt (10 % fat) was better with phenol
chloroform extraction (1 x 10^2 cfu/ml) than with QIAamp DNA Mini Kit (1 x
10^4 cfu/ml). By means of phenol chloroform extraction which is most suitable
for milk products, the detection limit for Brucella DNA in camembert and goat
cheese corresponded with 5 x 10^2 cfu/g. The extraction of DNA from soft
cheese requires a homogenisation after dilution in a fluid which diminishes
sensitivity. In contrast to cultivation of brucellae on nutrient agar for
several days under biosafety level 3 conditions, the detection by using DNA
extraction followed by real-time PCR can be achieved within half a working day
(QIAamp DNA Mini Kit) or alternatively within one day (phenol chloroform
extraction ) under biosafety level 2 conditions. However, the limit for
detection of Brucella DNA could not be improved by use of real-time PCR
compared to the classical PCR. In order to optimize the detection limits for
Brucella DNA especially in milk products, the immunomagnetic separation (IMS)
was tested as a pre-enrichment and purification procedure. For this purpose,
tosyl activated immunomagnetic particles coated with brucellosis serum or with
a monoclonal anti-Brucella antibody were used. Despite the optimization of IMS
considering different parameters such as sample volume, incubation time and
temperature, the limit for the detection of Brucella DNA was 5 x 10^3 cfu/g
for goat cheese, 1 x 10^3 cfu/ml for raw milk and still mineral water, 8 x
10^4 cfu/ml for yogurt and 5 x 10^4 cfu/g for camembert and was thus less
sensitive than DNA extraction methods mentioned above
Induktion von CD4+ CD25+ FoxP3+ regulatorischen T-Zellen durch retrovirale transmembrane HĂŒllproteine
Retrovirale Infektionen sind oft mit opportunistischen Infektionen auf der Basis eines supprimierten Immunsystems assoziiert. Es stellt sich die Frage, ob der Mechanismus der Induktion dieser Immunsuppression bei allen Retroviren identisch ist. Wegen ihrer konservierten AminosĂ€uresequenz â insbesondere in der sogenannten immunsuppressiven DomĂ€ne â wurde fĂŒr die transmembranen HĂŒllproteine eine mögliche Beteiligung daran vermutet. Bei Versuchen in vitro und in vivo konnten bereits einige immunsuppressive Eigenschaften beschrieben werden. Als ErgĂ€nzung wurde in dieser Arbeit die Zytokinmodulation und die Induktion regulatorischer T-Zellen (Tregs) durch retrovirale Proteine untersucht. Hierzu wurde zunĂ€chst das transmembrane HĂŒllprotein des humanen endogenen Retrovirus K (HERV-K TM) rekombinant hergestellt. Zur Vermeidung von Endotoxinen, die die Versuchsergebnisse verfĂ€lschen könnten, wurde zur Expression der Hefestamm Hansenula polymorpha RB11 verwendet. Die Aufreinigung von HERV-K TM aus dem Hefezellpellet wurde durch die Verwendung entsprechender Puffer und Glasperlen bei der Lyse optimiert. Die Produktion des transmembranen HĂŒllproteins p15E des porcinen endogenen Retrovirus in Hansenula polymorpha RB11 muss noch weiter optimiert werden. Weiterhin wurden die immunsuppressiven DomĂ€nen von HIV-1 und MuLV in der Form eines Heteropolymers untersucht. Nach der Inkubation humaner PBMCs mit HERV-K TM bzw. mit dem HIV-Isu/MuLV-Isu- Heteropolymer wurde fĂŒr FoxP3, IL-10 und IFNα1 eine Erhöhung des mRNAExpressionslevels gemessen. Diese trat nach einer Inkubationszeit von fĂŒnf Stunden bzw. bei IL-10 auch nach 24 Stunden auf. Parallel dazu wurde eine 500 bis 600fach erhöhte Freisetzung von IL-10 im Ăberstand der behandelten Zellen gemessen. Die Zahl der CD4+ CD25+ FoxP3+ Tregs wurde nach Inkubation humaner PBMCs mit den genannten retroviralen Peptiden bzw. Proteinen mittels Durchflusszytometrie bestimmt. Dabei stellte sich heraus, dass insbesondere HERV-K TM eine geringfĂŒgige Steigerung der Zahl an regulatorischen T-Zellen bewirkte. Dieser Anstieg bestĂ€tigt die Daten der Expressionsanalyse von FoxP3. Die hier gezeigte Induktion regulatorischer T-Zellen in vitro lĂ€sst vermuten, dass dies auch bei einer Infektion mit einem Retrovirus in vivo geschieht. Somit konnten weitere Erkenntnisse ĂŒber den Mechanismus der Immunsuppression gewonnen werden
Aspects of alpha-helical coiled coil folding
Die alpha-helikale coiled coil-Struktur ist eines der am hÀufigsten in der
Natur vorkommenden Faltungsmotive, dass an einer Vielzahl von wichtigen
Prozessen wie der viralen Membranfusion, der DNA Reparatur, der
Muskelkontraktion und der Transkription beteiligt ist. De novo entworfene
coiled coil-Peptide eignen sich aufgrund ihrer amphiphilen Natur und der
bekannten Designprinzipien hervorragend als Modellsysteme fĂŒr gröĂere und
komplexere biologisch relevante Proteine. Im Rahmen dieser Arbeit wurden de
novo entwickelte coiled coil-Peptide verwendet, um verschiedene Aspekte der
Proteinfaltung zu untersuchen. In Teil 1 wurde die kontextabhÀngige Bedeutung
von WasserstoffbrĂŒckenbindungen im PeptidrĂŒckgrat auf die Strukturbildung und
StabilitĂ€t von Proteinen untersucht. HierfĂŒr wurden Amidbindungen durch
Esterbindungen ersetzt, um systematisch einzelne WasserstoffbrĂŒcken zu
eliminieren. Die thermodynamische Analyse der Depsipeptide, sowie die
MolekĂŒldynamik-Simulationen, zeigten, dass der Einfluss der WasserstoffbrĂŒcken
auf die StabilitĂ€t der QuartĂ€rstruktur in groĂem MaĂe von der PolaritĂ€t der
jeweiligen Umgebung abhÀngt. Im hydrophoben Milieu der Proteinumgebung sind
die WasserstoffbrĂŒcken wesentlich stĂ€rker als in solvatisierten Regionen des
ProteinrĂŒckgrates. In Teil 2 wurde der Einfluss der multiplen O-Glykosylierung
auf die Faltung und StabilitĂ€t der coiled coil-Struktur untersucht. HierfĂŒr
wurden in eine 26 AminosÀuren lange Peptidsequenz systematisch ein bis sechs
ÎČ-D-Galactosereste ĂŒber die Serinseitenketten in den Positionen b, c und f des
Heptads eingefĂŒhrt. Aufgrund der geometrischen Vorgabe des Heptad-
Wiederholungsmusters können die Liganden in einem definierten Abstand
zueinander prÀsentiert werden. Die Studie zeigte, dass die O-Glykosylierung
mit ÎČ-D-Galactose ohne signifikante StabilitĂ€tsverluste toleriert wird und das
coiled coil-Faltungsmotiv daher groĂes Potential als multivalente
GerĂŒststruktur hat. Um weite RezeptorabstĂ€nde zu ĂŒberbrĂŒcken und die Valenz
des coiled coil-basierten GerĂŒstes zu erhöhen, wurde ein fasernbildendes
coiled coil-System entwickelt. Dieses besitzt eine einheitliche, aus
Lysinresten bestehende elektrostatische WechselwirkungsdomÀne, die ein
Ăberlappen der Helices ermöglicht. Anhand von coiled coil-basierten
Modellpeptiden wurde in Teil 3 der Einfluss der O-Glykosylierung auf die
Amyloidaggregation studiert. HierfĂŒr wurden ein bis drei ÎČ-D-Galactosereste in
verschiedene Positionen der Sequenz eingefĂŒhrt. Die mehrfache Glykosylierung
konnte die Amyloidbildung inhibieren, wohingegen die einfache Glykosylierung
positionsabhĂ€ngig nur zu einer Verzögerung der Amyloidbildung fĂŒhrte.The alpha-helical coiled coil folding motif is one of the most common in
nature, and is involved in many important biological processes including
virus-host cell membrane fusion, DNA repair, muscle contraction, and
transcription. De novo designed peptides that form coiled coils are suitable
model systems for larger and more complex biologically relevant proteins
because of their amphiphilic nature and well-known design principles. In this
thesis, de novo designed coiled coil-forming peptides were used to investigate
several aspects of protein folding. Part 1 describes the context-dependent
relevance of backbone hydrogen bonds in protein folding and stability. For
this purpose, amide bonds were replaced with ester bonds to systematically
eliminate specific hydrogen bonds. Thermodynamic analysis and molecular
dynamics simulations showed that the contribution of main chain hydrogen bonds
to quaternary structure stability largely depends on the environment. In
accordance with small molecule studies in solution, main chain hydrogen bonds
are significantly stronger in the interior of the hydrophobic interface than
they are in the solvent exposed aqueous regions of the coiled coil. The
influence of multiple O-glycosylation on the folding and stability of coiled
coils was the focus of part 2 of this thesis. One to six ÎČ-D-galactose
residues were introduced as monosaccharides into a 26 amino acid peptide
sequence via the serine side chains in positions b, c, and f of the heptad.
The carbohydrate ligands are presented in a defined relative distance due to
the geometric constrains of the heptad repeat. The study showed that
O-glycosylation with ÎČ-D-galactose is well-tolerated and does not result in
significant destabilization. Therefore, the coiled coil folding motif shows
potential as a multivalent scaffold. Furthermore, to bridge long receptor
distances and to extend the valence of the coiled coil-based scaffold, a
fiber-forming coiled coil system was developed. In this system, the
electrostatic recognition domain is composed exclusively of lysine residues,
which enhances overlap between the helices. Part 3 describes the use of coiled
coil-based model peptides to investigate the influence of O-glycosylation on
amyloid formation. One to three ÎČ-D-galactose residues were introduced at
different sites in the peptide sequence. Multiple glycosylation results in
complete inhibition of amyloid formation. Interestingly, single glycosylation
slows down the aggregation process in a manner that depends on the position in
the peptide sequence
