    Youth Media Still Matters: Elevating the Voices and Identities of Young People in Disruptive Times

    When schools, colleges and youth organizations shut down en masse in March 2020 in response to the emerging pandemic, none of us had any idea what lay ahead. Amid all the uncertainty, one thing quickly became clear: With opportunities to meet in person sharply curtailed, teens and young adults needed new outlets to connect, collaborate and find their creative voice more than ever.Using Zoom and other digital platforms, Spy Hop mentors delivered instruction remotely and students collaborated virtually on projects and performances. While the experience wasn't always the same, Spy Hop programming was able to reach a larger group of students who were no longer limited from participating by their geography.Through surveys, interviews and focus groups with Spy Hop mentors and students, we have assembled a comprehensive picture of the outcomes from Spy Hop programming in 2020 and 2021. In the course of our evaluation, we have found that Spy Hop continues to play an invaluable role in youth development and media skills.While Spy Hop programming continues to have indelible and far-reaching effects on youth participants in Utah and beyond, we believe the organization can deepen its impact even further by following a few suggestions. These include supporting youth employment opportunities beyond high school, broadening access to Spy Hop for students with fewer resources and being more intentional about mentor development

This thesis is a study of new technologies and their impact on political and community involvement. I-Neighbors.org is a technology allowing individuals register a free website and email list for their neighborhood. Through a particular feature of I-Neighbors called GovLink users can contact their elected officials for free. This thesis is based on a study of the behavior of I-Neighbors and GovLink users, looking for evidence that these technologies affect community involvement and political participation. Data gathered through surveys, the United States Census, and a short case study shows that new media can have a profound impact on community dialogue and political involvement.by Jacob William Faber.S.M

    Gender Differences in Coping with Racism: African American Experience and Empowerment

    Black men and women encounter multiple forms of racism in American society and require numerous strategies to manage the stress associated with these experiences. This chapter reviews the current state of the literature regarding Black people and how they cope with racism. Findings demonstrate that Black people tend to cope with racism through social support, religion, avoidance, and problem-focused coping, with some gender differences in coping approaches. We also contrast functional versus dysfunctional coping approaches and underscore the importance of empowerment to promote well-being and social change. Limitations of this review include the predominance of American-based samples used in the literature, which often excludes other Black ethnic and national groups. Further, the experiences of other Black intersectional identities are not well represented in the literature and require more study as their experiences of coping with racism may differ

    A Macroprudential Perspective on the Regulatory Boundaries of US Financial Assets

    This paper uses data from the Financial Accounts of the United States to map out the regulatory boundaries of assets held by US financial institutions from a macroprudential perspective. We provide a quantitative measure of the macroprudential regulatory boundary—the perimeter between the part of the financial sector that is subject to some form of macroprudential regulatory oversight and that which is not—and show how it has evolved over the past 40 years. Additionally, we measure the boundaries between different regulatory agencies and financial institutions that operate within the regulatory perimeter and illustrate how these boundaries potentially become blurred in the face of regulatory overlap. Quantifying the macroprudential regulatory boundary and the boundaries for different regulators within the perimeter is informative for assessing financial stability risks over the credit cycle

    Assay and heterologous expression in Pichia pastoris of plant cell wall type-II membrane anchored glycosyltransferases

    Two Arabidopsis xylosyltransferases, designated RGXT1 and RGXT2, were recently expressed in Baculovirus transfected insect cells and by use of the free sugar assay shown to catalyse transfer of D-xylose from UDP-α-D-xylose to L-fucose and derivatives hereof. We have now examined expression of RGXT1 and RGXT2 in Pichia pastoris and compared the two expression systems. Pichia transformants, expressing soluble, secreted forms of RGXT1 and RGXT2 with an N- or C-terminal Flag-tag, accumulated recombinant, hyper-glycosylated proteins at levels between 6 and 16 mg protein • L-1 in the media fractions. When incubated with 0.5 M L-fucose and UDP-D-xylose all four RGXT1 and RGXT2 variants catalyzed transfer of D-xylose onto L-fucose with estimated turnover numbers between 0.15 and 0.3 sec-1, thus demonstrating that a free C-terminus is not required for activity. N- and O-glycanase treatment resulted in deglycosylation of all four proteins, and this caused a loss of xylosyltransferase activity for the C-terminally but not the N-terminally Flag-tagged proteins. The RGXT1 and RGXT2 proteins displayed an absolute requirement for Mn2+ and were active over a broad pH range. Simple dialysis of media fractions or purification on phenyl Sepharose columns increased enzyme activities 2-8 fold enabling direct verification of the product formed in crude assay mixtures using electrospray ionization mass spectrometry. Pichia expressed and dialysed RGXT variants yielded activities within the range 0.011 to 0.013 U (1 U = 1 nmol conversion of substrate • min-1 • µl medium-1) similar to those of RGXT1 and RGXT2 expressed in Baculovirus transfected insect Sf9 cells. In summary, the data presented suggest that Pichia is an attractive host candidate for expression of plant glycosyltransferases

    Galectin-3 and fibulin-1 in systolic heart failure:relation to glucose metabolism and left ventricular contractile reserve

    Abstract Background Heart failure (HF) patients with diabetes (DM) have an adverse prognosis and reduced functional capacity, which could be associated with cardiac fibrosis, increased chamber stiffness and reduced left ventricular (LV) contractile reserve. Galectin-3 (Gal-3) and fibulin-1 are circulating biomarkers potentially reflecting cardiac fibrosis. We hypothesize that plasma levels of Gal-3 and fibulin-1 are elevated in HF patients with DM and are associated with reduced LV contractile reserve in these patients. Methods A total of 155 patients with HF with reduced ejection fraction underwent a low-dose dobutamine echocardiography and blood sampling for biomarker measurements. Patients were classified according to history of DM and an oral glucose tolerance test (OGTT) as: normal glucose tolerance (NGT) ( n \u2009=\u200970), impaired glucose tolerance (IGT) ( n \u2009=\u200925) and DM ( n \u2009=\u200960). Results Galectin-3 levels were elevated in DM patients as compared to non-diabetic patients ( P \u2009=\u20090.02), while higher fibulin-1 levels were observed in HF patients with IGF and DM ( P \u2009=\u20090.07). Reduced LV contractile reserve was associated with increasing Gal-3 levels (\u3b2\u2009=\u2009\u22120.19, P \u2009=\u20090.03) although, this association was attenuated after adjustment for estimated glomerular filtration rate ( P \u2009=\u20090.66). Fibulin-1 was not associated with LV contractile reserve ( P \u2009=\u20090.71). Conclusions Galectin-3 and fibulin-1 levels were elevated in HF patients with impaired glucose metabolism. However, reduced LV contractile reserve among HF patients with DM does not to have an independent impact on plasma Gal-3 and fibulin-1 levels

    HST/ACS Emission Line Imaging of Low Redshift 3CR Radio Galaxies I: The Data

    We present 19 nearby (z<0.3) 3CR radio galaxies imaged at low- and high-excitation as part of a Cycle 15 Hubble Space Telescope snapshot survey with the Advanced Camera for Surveys. These images consist of exposures of the H-alpha (6563 \AA, plus [NII] contamination) and [OIII] 5007 \AA emission lines using narrow-band linear ramp filters adjusted according to the redshift of the target. To facilitate continuum subtraction, a single-pointing 60 s line-free exposure was taken with a medium-band filter appropriate for the target's redshift. We discuss the steps taken to reduce these images independently of the automated recalibration pipeline so as to use more recent ACS flat-field data as well as to better reject cosmic rays. We describe the method used to produce continuum-free (pure line-emission) images, and present these images along with qualitative descriptions of the narrow-line region morphologies we observe. We present H-alpha+[NII] and [OIII] line fluxes from aperture photometry, finding the values to fall expectedly on the redshift-luminosity trend from a past HST/WFPC2 emission line study of a larger, generally higher redshift subset of the 3CR. We also find expected trends between emission line luminosity and total radio power, as well as a positive correlation between the size of the emission line region and redshift. We discuss the associated interpretation of these results, and conclude with a summary of future work enabled by this dataset.Comment: 18 pages, 12 figures, accepted for publication in ApJ

    The SLUGGS Survey: stellar kinematics, kinemetry and trends at large radii in 25 early-type galaxies

    Due to longer dynamical time-scales, the outskirts of early-type galaxies retain the footprint of their formation and assembly. Under the popular two-phase galaxy formation scenario, an initial in situ phase of star formation is followed by minor merging and accretion of ex situ stars leading to the expectation of observable transitions in the kinematics and stellar populations on large scales. However, observing the faint galactic outskirts is challenging, often leaving the transition unexplored. The large-scale, spatially resolved stellar kinematic data from the SAGES Legacy Unifying Galaxies and GlobularS (SLUGGS) survey are ideal for detecting kinematic transitions. We present kinematic maps out to 2.6 effective radii on average, kinemetry profiles, measurement of kinematic twists and misalignments, and the average outer intrinsic shape of 25 SLUGGS galaxies. We find good overall agreement in the kinematic maps and kinemetry radial profiles with literature. We are able to confirm significant radial modulations in rotational versus pressure support of galaxies with radius so that the central and outer rotational properties may be quite different. We also test the suggestion that galaxies may be more triaxial in their outskirts and find that while fast rotating galaxies were already shown to be axisymmetric in their inner regions, we are unable to rule out triaxiality in their outskirts.We compare our derived outer kinematic information to model predictions from a two-phase galaxy formation scenario. We find that the theoretical range of local outer angular momentum agrees well with our observations, but that radial modulations are much smaller than predicted
