    Instantaneous cell migration velocity may be ill-defined

    Cell crawling is critical to biological development, homeostasis and disease. In many cases, cell trajectories are quasi-random-walk. In vitro assays on flat surfaces often described such quasi-random-walk cell trajectories as approximations to a solution of a Langevin process. However, experiments show quasi-diffusive behavior at small timescales, indicating that instantaneous velocity and velocity autocorrelations are not well-defined. We propose to characterize mean-squared cell displacement using a modified F\"urth equation with three temporal and spatial regimes: short- and long-time/range diffusion and intermediate time/range ballistic motion. This analysis collapses mean-squared displacements of previously published experimental data onto a single-parameter family of curves, allowing direct comparison between movement in different cell types, and between experiments and numerical simulations. Our method also show that robust cell-motility quantification requires an experiment with a maximum interval between images of a few percent of the cell-motion persistence time or less, and a duration of a few orders-of-magnitude longer than the cell-motion persistence time or more.Comment: 5 pages, plus Supplemental materia

    A minimal model for congestion phenomena on complex networks

    We study a minimal model of traffic flows in complex networks, simple enough to get analytical results, but with a very rich phenomenology, presenting continuous, discontinuous as well as hybrid phase transitions between a free-flow phase and a congested phase, critical points and different scaling behaviors in the system size. It consists of random walkers on a queueing network with one-range repulsion, where particles can be destroyed only if they can move. We focus on the dependence on the topology as well as on the level of traffic control. We are able to obtain transition curves and phase diagrams at analytical level for the ensemble of uncorrelated networks and numerically for single instances. We find that traffic control improves global performance, enlarging the free-flow region in parameter space only in heterogeneous networks. Traffic control introduces non-linear effects and, beyond a critical strength, may trigger the appearance of a congested phase in a discontinuous manner. The model also reproduces the cross-over in the scaling of traffic fluctuations empirically observed in the Internet, and moreover, a conserved version can reproduce qualitatively some stylized facts of traffic in transportation networks

    Randomized, double-blind, placebo-controlled trial of rapamycin in amyotrophic lateral sclerosis

    In preclinical studies rapamycin was found to target neuroinflammation, by expanding regulatory T cells, and affecting autophagy, two pillars of amyotrophic lateral sclerosis (ALS) pathogenesis. Herein we report a multicenter, randomized, double-blind trial, in 63 ALS patients who were randomly assigned in a 1:1:1 ratio to receive rapamycin 2 mg/m2/day,1 mg/m2/day or placebo (EUDRACT 2016-002399-28; NCT03359538). The primary outcome, the number of patients exhibiting an increase >30% in regulatory T cells from baseline to treatment end, was not attained. Secondary outcomes were changes from baseline of T, B, NK cell subpopulations, inflammasome mRNA expression and activation status, S6-ribosomal protein phosphorylation, neurofilaments; clinical outcome measures of disease progression; survival; safety and quality of life. Of the secondary outcomes, rapamycin decreased mRNA relative expression of the pro-inflammatory cytokine IL-18, reduced plasmatic IL-18 protein, and increased the percentage of classical monocytes and memory switched B cells, although no corrections were applied for multiple tests. In conclusion, we show that rapamycin treatment is well tolerated and provides reassuring safety findings in ALS patients, but further trials are necessary to understand the biological and clinical effects of this drug in ALS

    Project of an advanced ISOL facility for exotic beams at LNL

    Abstract In the framework of the European program to define a second generation Radioactive Ion Beam facility, LNL are proposing the construction in the next five–seven years of a specialized national facility for RIB originated by fission fragments produced by secondary neutrons. It consists on a two-accelerator ISOL-type facility to provide intense neutron-rich radioactive ion beams of highest quality, in the range of masses between 80 and 160. The conceptual design is based on a high intensity 50 MeV (100 kW) proton linac as driver and on the availability of the heavy-ion accelerator ALPI as post accelerator. The estimated neutron yield is 2×1014 n/s at 0°, high enough to satisfy the demand for an advanced RIB facility. An intense R&D program on different items is actually in progress in collaboration with other Laboratories and University groups and is moving in a European context

    'We looked after people better when we were informal' : the 'quasi-formalisation' of Montevideo's waste-pickers

    This article was written thanks to funding from the Economic and Social Research Council (Grant Code: ES/S011048/1).Drawing on participatory research, this article explores the state formalisation of Uruguayan clasificadores (waste‐pickers). It goes beyond the informal/formal binary, instead proposing the concepts of ‘para‐formality’ to describe economic activity that exists in parallel to regulated and taxed spheres, and ‘quasi‐formality’ to describe processes of formalisation that are supported by underlying informal practices. When unregulated, clasificadores enjoyed parallel services in health, finance and social security, implying that benefits of ‘formalisation’ must be explored ethnographically rather than assumed. The persistence of ‘quasi‐formal’ activity within formalised recycling plants complicates simple narratives of informal to formal transitions and suggests that the concept can be useful for the study of labour policies in Latin America and beyondPublisher PDFPeer reviewe

    MEDLEM database, a data collection on large elasmobranchs in the Mediterranean and Black Seas

    The Mediterranean Large Elasmobranchs Monitoring (MEDLEM) database contains more than 3,000 records (with more than 4,000 individuals) of large elasmobranch species from 21 different countries around the Mediterranean and Black seas, observed from 1666 to 2017. The principal species included in the archive are the devil ray (1,868 individuals), the basking shark (935 individuals), the blue shark (622 individuals), and the great white shark (342 individuals). In the last decades, other species such as the thresher shark (187 individuals), the shortfin mako (180 individuals), and the spiny butterfly ray (138) were reported with increasing frequency. This was possibly due to increased public awareness on the conservation status of sharks, and the consequent development of new monitoring programs. MEDLEM does not have homogeneous reporting coverage throughout the Mediterranean and Black seas and it should be considered as a database of observed species presence. Scientific monitoring efforts in the south-eastern Mediterranean and Black seas are generally lower than in the northern sectors and the absence of some species in our database does not imply their actual absence in these regions.
