    An Analysis of Finite Difference and Galerkin Techniques Applied to the Simulation of Advection and Diffusion of Air Pollutants from a Line Source

    A finite difference and a Galerkin type scheme are compared with reference to a very accurate solution describing time dependent advection and diffusion of air pollutants from a line source in an atmosphere vertically stratified and limited by an inversion layer. The accurate solution was achieved by applying the finite difference scheme on a very refined grid with a very small time step. Grid size and time step were defined according to stability and accuracy criteria discussed in the text. It is found that for the problem considered, the two methods can be considered equally accurate. However, the Galerkin method gives larger areas of small errors close to the source, This was assumed to be partly due to the different way the source term is taken into account by the two methods. An improvement of the accuracy of the finite difference scheme was achieved by approximating, at every step, the contribution of the source term by a Gaussian puff moving and diffusing with velocity and diffusivity of the source location, instead of utilizing a stepwise function for the numerical approximation of the delta function representing the source term

    Hollow-Core Fiber-Based Biosensor: A Platform for Lab-in-Fiber Optical Biosensors for DNA Detection

    In this paper, a novel platform for lab-in-fiber-based biosensors is studied. Hollow-core tube lattice fibers (HC-TLFs) are proposed as a label-free biosensor for the detection of DNA molecules. The particular light-guiding mechanism makes them a highly sensitive tool. Their transmission spectrum is featured by alternations of high and low transmittance at wavelength regions whose values depend on the thickness of the microstructured web composing the cladding around the hollow core. In order to achieve DNA detection by using these fibers, an internal chemical functionalization process of the fiber has been performed in five steps in order to link specific peptide nucleic acid (PNA) probes, then the functionalized fiber was used for a three-step assay. When a solution containing a particular DNA sequence is made to flow through the HC of the TLF in an ‘optofluidic’ format, a bio-layer is formed on the cladding surfaces causing a red-shift of the fiber transmission spectrum. By comparing the fiber transmission spectra before and after the flowing it is possible to identify the eventual formation of the layer and, therefore, the presence or not of a particular DNA sequence in the solution

    Physically effective NDF (peNDF) is defined as the fraction of fibrethat stimulates chewing and contributes to the floating mat oflarge particles in the rumen, and consequently to its regular activity.PeNDF is calculated from a physical effectiveness factor (pef),varying from 0 (NDF stimulates no chewing) to 1 (max chewing),which may be obtained by laboratory-based particle sizing techniques,such as Penn State Particle Separator, Mertens Separator,Z-Box, Cut Accuracy Test, based on the proportion of DM retainedon sieves (by horizontal or vertical shaking). We chose Z-Boxmethod, thanks to its easy use and applicability to as-is feed andtotal mixed rations (TMR), and we are trying to obtain an estimatingequation which may predict milk fat content and/or other productivedata from peNDF and other variables measured on TMR.To this aim, samples of TMR collected from several farms aresieved (3 sub samples each), and undergo proximate analysis,NDF, ADF, ADL and starch. Milk yield, milk fat, water addiction toTMR are collected on farm; qualitative data such as type of forage,breed, season, geographical origin and altitude (plain/hill/mountain)are also taken into account, to estimate their possible effect.As a first step, in order to investigate the complex relationshipsexisting among this wide set of variables, Principal ComponentAnalysis (PCA) is used as a data exploration tool. Two PCA models(presence of silage or not in TMR) are calculated separately. Foreach PCA model, the overall correlations among all the consideredvariables and their relative importance are investigated by meansof the loadings plots, posing particular attention to the correlationswith peNDF and with milk fat. Moreover, it is also possible to identifyhow the different groups of samples depend on specific variables

    Effectiveness of expressive writing protocol in palliative care healthworkers: A quantitative study

    Background and aim of the work: Palliative Care professionals are exposed to intense emotional envi-ronment. This puts them at risk for Compassion Fatigue and Burnout. The protective factors that can counter their onset are Compassion Satisfaction, Organizational Commitment and Resilience. Expressive Writing is a valid tool for adapting to traumatic events and enhancing psychological well-being. Aim of this study is to evaluate the effect of the Expressive Writing in Palliative Care professionals on Compassion Satisfaction, Organizational Commitment, Resilience, Compassion Fatigue and perceived distress. Methods: Prospective experimental study with experimental/control groups and pre/post measurements. 50 Palliative Care professionals were recruited in Northern and Central Italy. Participants filled: Organizational Commitment Questionnaire; ProQol-revision III; Resilience Scale for Adults; Impact of Event-Scale Revised; Emotion Thermometer; ad hoc questionnaire for the evaluation of protocol usefulness. Results: Wilcoxon test demon-strated change in Continuative Commitment (Z =-3.357, p = .001), anger (Z =-2.214, p = .027), sleep (Z =-2.268, p = .023), help (Z =-2.184, p = .029), intrusiveness (Z =-2.469, p = .014), hyperarousal (Z =-2.717, p = .007), and total IES (Z =-2.456, p =, 014). Mann Whitney test showed a significantly lower score on post-test Intrusiveness in the experimental group (U = 202, p = .038). Conclusions: The Expressive Writing intervention was effective in improving organizational and emotional variables. Expressive Writing supports healthcare professionals in relieving the burden of traumatic episodes, ordering associated thoughts and emo-tions, and implementing a process of deep comprehension

    Development and Validation of a Novel Diagnostic Test for Human Brucellosis Using a Glyco-engineered Antigen Coupled to Magnetic Beads.

    Brucellosis is a highly contagious zoonosis and still a major human health problem in endemic areas of the world. Although several diagnostic tools are available, most of them are difficult to implement especially in developing countries where complex health facilities are limited. Taking advantage of the identical structure and composition of the Brucella spp. and Yersinia enterocolitica O:9 O-polysaccharide, we explored the application of a recombinant Y. enterocolitica O:9-polysaccharide-protein conjugate (OAg-AcrA) as a novel antigen for diagnosis of human brucellosis. We have developed and validated an indirect immunoassay using OAg-AcrA coupled to magnetic beads. OAg-AcrA was produced and purified with high yields in Y. enterocolitica O:9 cells co-expressing the oligosaccharyltransferase PglB and the protein acceptor AcrA of Campylobacter jejuni without the need for culturing Brucella. Expression of PglB and AcrA in Y. enterocolitica resulted in the transfer of the host O-polysaccharide from its lipid carrier to AcrA. To validate the assay and determine the cutoff values a receiver-operating characteristic analysis was performed using a panel of characterized serum samples obtained from healthy individuals and patients of different clinical groups. Our results indicate that, using this assay, it is possible to detect infection caused by the three main human brucellosis agents (B. abortus, B. melitensis and B. suis) and select different cutoff points to adjust sensitivity and specificity levels as needed. A cutoff value of 13.20% gave a sensitivity of 100% and a specificity of 98.57%, and a cutoff value of 16.15% resulted in a test sensitivity and specificity of 93.48% and 100%, respectively. The high diagnostic accuracy, low cost, reduced assay time and simplicity of this new glycoconjugate-magnetic beads assay makes it an attractive diagnostic tool for using not only in clinics and brucellosis reference laboratories but also in locations with limited laboratory infrastructure and/or minimally trained community health workers.Fil: Ciocchini, Andres Eduardo.     The (decision) tree of fertility: an innovative decision-making algorithm in assisted reproduction technique

    Purpose: Several mathematical models have been developed to estimate individualized chances of assisted reproduction techniques (ART) success, although with limited clinical application. Our study aimed to develop a decisional algorithm able to predict pregnancy and live birth rates after controlled ovarian stimulation (COS) phase, helping the physician to decide whether to perform oocytes pick-up continuing the ongoing ART path. Methods: A single-center retrospective analysis of real-world data was carried out including all fresh ART cycles performed in 1998–2020. Baseline characteristics, ART parameters and biochemical/clinical pregnancies and live birth rates were collected. A seven-steps systematic approach for model development, combining linear regression analyses and decision trees (DT), was applied for biochemical, clinical pregnancy, and live birth rates. Results: Of fresh ART cycles, 12,275 were included. Linear regression analyses highlighted a relationship between number of ovarian follicles > 17 mm detected at ultrasound before pick-up (OF17), embryos number and fertilization rate, and biochemical and clinical pregnancy rates (p < 0.001), but not live birth rate. DT were created for biochemical pregnancy (statistical power–SP:80.8%), clinical pregnancy (SP:85.4%), and live birth (SP:87.2%). Thresholds for OF17 entered in all DT, while sperm motility entered the biochemical pregnancy’s model, and female age entered the clinical pregnancy and live birth DT. In case of OF17 < 3, the chance of conceiving was < 6% for all DT. Conclusion: A systematic approach allows to identify OF17, female age, and sperm motility as pre-retrieval predictors of ART outcome, possibly reducing the socio-economic burden of ART failure, allowing the clinician to perform or not the oocytes pick-up

    Combining Litter Observations with a Regional Ocean Model to Identify Sources and Sinks of Floating Debris in a Semi-enclosed Basin: The Adriatic Sea

    Visual ship transect surveys provide crucial information about the density, and spatial distribution of floating anthropogenic litter in a basin. However, such observations provide a ‘snapshot’ of local conditions at a given time and cannot be used to deduce the provenance of the litter or to predict its fate, crucial information for management and mitigation policies. Particle tracking techniques have seen extensive use in these roles, however, most previous studies have used simplistic initial conditions based on bulk average inputs of debris to the system. Here, observations of floating anthropogenic macro debris in the Adriatic Sea are used to define initial conditions (number of particles, location, and time) in a Lagrangian particle tracking model. Particles are advected backward and forward in time for 60 days (120 days total) using surface velocities from an operational regional ocean model. Sources and sinks for debris observed in the central and southern Adriatic in May 2013 and March 2015 included the Italian coastline from Pescara to Brindisi, the Croatian island of Mljet, and the coastline from Dubrovnik through Montenegro to Albania. Debris observed in the northern Adriatic originated from the Istrian peninsula to the Italian city of Termoli, as well as the Croatian island of Cres and the Kornati archipelago. Particles spent a total of roughly 47 days afloat. Coastal currents, notably the eastern and western Adriatic currents, resulted in large alongshore displacements. Our results indicate that anthropogenic macro debris originates largely from coastal sources near population centers and is advected by the cyclonic surface circulation until it strands on the southwest (Italian) coast, exits the Adriatic, or recirculates in the southern gyreVersión del edito

    Lactobacilli and bifidobacteria in the feces of schoolchildren of two different socioeconomic groups: children from a favela and children from a private school

    OBJECTIVE: To determine the number of lactobacillus and bifidobacterium colonies in the feces of schoolchildren from two different socioeconomic levels. METHODS: We analyzed fecal samples of children aged 6 to 10 years without gastrointestinal symptoms or recent use of antimicrobials. The first group included 86 children living in a favela in the city of Osasco, state of São Paulo, southeastern Brazil. The second group included 36 children attending a private school in the same city. Body mass index (BMI) was used to assess nutritional status according to the reference values of the National Center for Health Statistics (NCHS). Specific anaerobic culture media were used for isolation of colonies for 48 and 72 hours at 37 °C. The number of colonies was determined using the plate-counting method. RESULTS: The mean lactobacillus (1.125 x 10(9) colony-forming units, CFU/g) and bifidobacterium (1.675 x 10(9) CFU/g) counts in the private school group were higher (p -1.0 SD (n = 57): 0.350 x 10(9) and 0.420 x 10(9) CFU/g, respectively. CONCLUSION: The microbiota of schoolchildren living in unfavorable environmental conditions shows lower numbers of fecal lactobacillus and bifidobacterium colonies, especially in children with lower BMI values.OBJETIVO: Determinar o número de colônias de lactobacilos e bifidobactérias nas fezes de crianças escolares, pertencentes a dois estratos socioeconômicos. MÉTODOS: Foram analisadas amostras de fezes de crianças com idade entre 6 e 10 anos sem sintomas gastrointestinais ou uso recente de antimicrobianos. O primeiro grupo foi constituído por 86 crianças, moradoras em uma favela localizada no município de Osasco (SP). O segundo grupo foi constituído por 36 crianças matriculadas em uma escola particular da mesma cidade. O estado nutricional foi avaliado usando o índice de massa corporal (IMC) de acordo com os valores de referência do National Center for Health Statistics (NCHS). O isolamento das colônias foi realizado em meios de cultura específicos em anaerobiose, durante 48 e 72 horas a 37 °C. A determinação do número foi feita pelo método da contagem em placa. RESULTADOS: A mediana de lactobacilos (1,125 x 10(9) unidades formadoras de colônia, UFC/g) e bifidobactérias (1,675 x 10(9) UFC/g) na escola particular foi superior (p -1,0 desvio padrão (n = 57): 0,350 x 10(9) e 0,420 x 10(9) UFC/g, respectivamente.     Mapping Local Charge Recombination Heterogeneity by Multidimensional Nanospectroscopic Imaging

    As materials functionality becomes more dependent on local physical and electronic properties, the importance of optically probing matter with true nanoscale spatial resolution has increased. In this work, we mapped the influence of local trap states within individual nanowires on carrier recombination with deeply subwavelength resolution. This is achieved using multidimensional nanospectroscopic imaging based on a nano-optical device. Placed at the end of a scan probe, the device delivers optimal near-field properties, including highly efficient far-field to near-field coupling, ultralarge field enhancement, nearly background-free imaging, independence from sample requirements, and broadband operation. We performed ~40-nanometer–resolution hyperspectral imaging of indium phosphide nanowires via excitation and collection through the probes, revealing optoelectronic structure along individual nanowires that is not accessible with other methods

    Cutaneous Involvement in Diseases with Plasma Cell Differentiation: Diagnostic Approach

    Neoplasms with plasma cell differentiation may occasionally involve the skin. Cutaneous lesions may represent the first sign of an underlying systemic plasma cell malignancy, such as multiple myeloma, or the skin itself may be the primary site of occurrence of a hematological tumor with plasma cell differentiation. Starting from examples encountered in our daily practice, we discussed the diagnostic approach pathologists and clinicians should use when faced with cutaneous lesions with plasma cell differentiation. Cases of primary cutaneous marginal zone lymphoma, localized primary amyloidosis/amyloidoma, and cutaneous manifestations (secondary either to multiple myeloma or to plasmablastic lymphoma) are discussed, focusing on the importance of the adequate patient’s work-up and precise clinicopathological correlation to get to the correct diagnosis and appropriate treatment. The pertinent literature has been reviewed, and the clinical presentation, pathological findings, main differential diagnoses, treatment, and outcome of neoplasms with plasma cell differentiation involving the skin are discussed
