Negara Vs Masyarakat: Konflik Tanah Di Kabupaten Nagekeo, NTT
This paper describes the land conflicts that occurred in the district of Nagekeo between the state VS society. We know that the state represented by the government, if in the area sometimes referred to as local governments in the process of infrastructure development for the public interest often face the issue of compensation in the form of land acquisition and land tenure issues. Issues such as this led to the government and people often face to face in a conflict that sometimes led to violence and anarchy. Although the resolution adopted sometimes has completed but the conflict will not stop there in some land conflicts in Indonesia. Finally, the authors conclude that the land conflict between the State VS Nagekeo Society is land ownership arrangement error and deviation of the Act and Regulations presidential About Land Acquisition for Public Interest, besides there are personal and group interests that triggered the conflict in Nagekeo
RANCANG BANGUN ALAT PENGUKUR JARAK AMAN MOBIL PADA AREA TEMPAT PARKIR UMUM MENGGUNAKAN SENSOR ULTRASONIC HC-SR04 DAN ARDUINO UNO
Penelitian ini bertujuan untuk mengetahui bagaimana tahapan membuat prototype alat pengukurjarak aman mobil menggunakan sensor ultrasonic HC-SR04 berbasis mikrokontroler arduino uno.Penelitian ini menggunakan metode R&D yaitu metode penelitian yang digunakan untuk menghasilkanproduk tertentu dan menguji keefektifan produk tersebut. Hasil penelitian menunjukan terdapat tiga tahapdalam membuat sistem pengukur jarak aman mobil yaitu tahapan persiapan yang berkaitan denganperancangan yang harus dipersiapkan demi menunjang proses pengerjaan, tahapan pelaksanaan yaitumemulai proses Alat Pengukur Jarak Aman Mobil Pada Area Tempat Parkir Umum Menggunakan SensorUltrasonic HC-SR04 Dan Arduino Uno serta tahap akhir menganalisa keberhasilan dari proses perakitansistem pengukur jarak aman mobil
Improved genome editing in human cell lines using the CRISPR method
The Cas9/CRISPR system has become a popular choice for genome editing. In this system, binding of a single guide (sg) RNA to a cognate genomic sequence enables the Cas9 nuclease to induce a double-strand break at that locus. This break is next repaired by an error-prone mechanism, leading to mutation and gene disruption. In this study we describe a range of refinements of the method, including stable cell lines expressing Cas9, and a PCR based protocol for the generation of the sgRNA. We also describe a simple methodology that allows both elimination of Cas9 from cells after gene disruption and re-introduction of the disrupted gene. This advance enables easy assessment of the off target effects associated with gene disruption, as well as phenotype-based structure-function analysis. In our study, we used the Fan1 DNA repair gene as control in these experiments. Cas9/CRISPR-mediated Fan1 disruption occurred at frequencies of around 29%, and resulted in the anticipated spectrum of genotoxin hypersensitivity, which was rescued by re-introduction of Fan1
Development of biodegradable flexible films of starch and poly(lactic acid) plasticized with adipate or citrate esters
AbstractBiodegradable films were produced from blends contained a high amount of thermoplastic starch (TPS) and poly(lactic acid) (PLA) plasticized with different adipate or citrate esters. It was not possible to obtain pellets for the production of films using only glycerol as a plasticizer. The plasticization of the PLA with the esters and mixture stages added through extrusion was critical to achieve a blend capable of producing films by blow extrusion. Adipate esters were the most effective plasticizers because they interacted best with the PLA and yielded films with appropriate mechanical properties
Experimental evidence for fast cluster formation of chain oxygen vacancies in YBa2Cu3O7-d being at the origin of the fishtail anomaly
We report on three different and complementary measurements, namely
magnetisation measurements, positron annihilation spectroscopy and NMR
measurements, which give evidence that the formation of oxygen vacancy clusters
is on the origin of the fishtail anomaly in YBa2Cu3O7-d. While in the case of
YBa2Cu3O7.0 the anomaly is intrinsically absent, it can be suppressed in the
optimally doped state where vacancies are present. We therefore conclude that
the single vacancies or point defects can not be responsible for this anomaly

Combinatorial CRISPR-Cas9 screens for de novo mapping of genetic interactions.
We developed a systematic approach to map human genetic networks by combinatorial CRISPR-Cas9 perturbations coupled to robust analysis of growth kinetics. We targeted all pairs of 73 cancer genes with dual guide RNAs in three cell lines, comprising 141,912 tests of interaction. Numerous therapeutically relevant interactions were identified, and these patterns replicated with combinatorial drugs at 75% precision. From these results, we anticipate that cellular context will be critical to synthetic-lethal therapies
Diagnosis of diffuse idiopathic skeletal hyperostosis with chest computed tomography:inter-observer agreement
To evaluate and improve the interobserver agreement for the CT-based diagnosis of diffuse idiopathic skeletal hyperostosis (DISH). Six hundred participants of the CT arm of a lung cancer screening trial were randomly divided into two groups. The first 300 CTs were scored by five observers for the presence of DISH based on the original Resnick criteria for radiographs. After analysis of the data a consensus meeting was organised and the criteria were slightly modified regarding the definition of 'contiguous', the definition of 'flowing ossifications' and the viewing plane and window level. Subsequently, the second set of 300 CTs was scored by the same observers. kappa >= 0.61 was considered good agreement. The 600 male participants were on average 63.5 (SD 5.3) years old and had smoked on average 38.0 pack-years. In the first round kappa values ranged from 0.32 to 0.74 and 7 out of 10 values were below 0.61. After the consensus meeting the interobserver agreement ranged from 0.51 to 0.86 and 3 out of 10 values were below 0.61. The agreement improved significantly. This is the first study that reports interobserver agreement for the diagnosis of DISH on chest CT, showing mostly good agreement for modified Resnick criteria. . DISH is diagnosed on fluoroscopic and radiographic examinations using Resnick criteria . Evaluation of DISH on chest CT was modestly reproducible with the Resnick criteria . A consensus meeting and Resnick criteria modification improved inter-rater reliability for DISH . Reproducible CT criteria for DISH aids research into this poorly understood entity

Cardiovascular screening may benefit middle-aged sportsmen, as coronary artery disease (CAD) is the main cause of exercise-related sudden cardiac death. Arterial stiffness, as measured by pulse wave velocity (PWV), may help identify sportsmen with subclinical CAD. We examined the additional value of PWV measurements to traditional CAD risk factors for identifying CAD.From the Measuring Athlete's Risk of Cardiovascular events (MARC) cohort of asymptomatic, middle-aged sportsmen who underwent low-dose Cardiac CT (CCT) after routine sports medical examination (SME), 193 consecutive sportsmen (aged 55 ± 6.6 years) were included with additional PWV measurements before CCT. Sensitivity, specificity and predictive values of PWV values (>8.3 and >7.5 m/s) assessed by Arteriograph were used to identify CAD (coronary artery calcium scoring ≥ 100 Agatston Units or coronary CT angiography luminal stenosis ≥ 50%) and to assess the additional diagnostic value of PWV to established cardiovascular risk factors.Forty-seven sportsmen (24%) had CAD on CCT. They were older (58.9 vs. 53.8 years, p<0.001), had more hypertension (17 vs. 4%, p=0.003), higher cholesterol levels (5.7 vs. 5.4 mmol/l) p=0.048), and more often were (ever) smokers (55 vs. 34%, p=0.008). Mean PWV was higher in those with CAD (8.9 vs. 8.0 m/s, p=0.017). For PWV >8.3m/s respectively >7.5 m/s sensitivity to detect CAD on CT was 43% and 74%, specificity 69% and 45%, positive predictive value 31% and 30%, and negative predictive value 79% and 84%. Adding PWV to traditional risk factor models did not change the area under the curve (from 0.78 (95% CI = 0.709-0.848)) to AUC 0.78 (95% CI 0.710-0.848, p = 0.99)) for prediction of CAD on CCT.Limited additional value was found for PWV on top of established risk factors to identify CAD. PWV might still have a role to identify CAD in middle-aged sportsmen if risk factors such as cholesterol are unknown
Charge degree of freedom and single-spin fluid model in YBa_2Cu_4O_8
We present a 17O nuclear magnetic resonance study in the stoichiometric
superconductor YBa_2Cu_4O_8. A double irradiation method enables us to show
that, below around 180 K, the spin-lattice relaxation rate of plane oxygen is
not only driven by magnetic, but also significantly by quadrupolar
fluctuations, i.e. low-frequency charge fluctuations. In the superconducting
state, on lowering the temperature, the quadrupolar relaxation diminishes
faster than the magnetic one. These findings show that, with the opening of the
pseudo spin gap, a charge degree of freedom of mainly oxygen character is
