    Stuttering Min oscillations within E. coli bacteria: A stochastic polymerization model

    We have developed a 3D off-lattice stochastic polymerization model to study subcellular oscillation of Min proteins in the bacteria Escherichia coli, and used it to investigate the experimental phenomenon of Min oscillation stuttering. Stuttering was affected by the rate of immediate rebinding of MinE released from depolymerizing filament tips (processivity), protection of depolymerizing filament tips from MinD binding, and fragmentation of MinD filaments due to MinE. Each of processivity, protection, and fragmentation reduces stuttering, speeds oscillations, and reduces MinD filament lengths. Neither processivity or tip-protection were, on their own, sufficient to produce fast stutter-free oscillations. While filament fragmentation could, on its own, lead to fast oscillations with infrequent stuttering; high levels of fragmentation degraded oscillations. The infrequent stuttering observed in standard Min oscillations are consistent with short filaments of MinD, while we expect that mutants that exhibit higher stuttering frequencies will exhibit longer MinD filaments. Increased stuttering rate may be a useful diagnostic to find observable MinD polymerization in experimental conditions.Comment: 21 pages, 7 figures, missing unit for k_f inserte

    Loss of Arabidopsis matrix metalloproteinase-5 affects root development and root bacterial communities during drought stress

    Matrix metalloproteinases (MMPs) are zinc-dependent endo-peptidases that in mammals are known to be involved in remodeling the extracellular matrix (ECM) in developmental and pathological processes. In this study, we report At5-MMP of Arabidopsis thaliana to be important for root development and root bacterial communities. At5-MMP is mainly localized in the root vasculature and lateral root, an At5-MMP T-DNA insertion mutant (mmp5 KO) showed reduced root growth and a lower number of root apexes, causing reduced water uptake from the soil. Subsequently, mmp5 KO is sensitive to drought stress. Inhibited auxin transport was accompanied with resistance to indole-3-acetic acid (IAA), 2, 4-dichlorophenoxyacetic acid (2, 4-D), and 1-naphthaleneacetic acid (NAA). The content of endogenous abscisic acid (ABA) was lower in roots of mmp5 KO than in wild type. Genes responsive to ABA as well as genes encoding enzymes of the proline biosynthesis were expressed to a lower extent in mmp5 KO than in wild type. Moreover, drought stress modulated root-associated bacterial communities of mmp5 KO: the number of Actinobacteria increased. Therefore, At5-MMP modulates auxin/ABA signaling rendering the plant sensitive to drought stress and recruiting differential root bacterial communities

    Persistence of Overseeded Cool-Season Grasses in Bermudagrass Turf

    Cool-season grass species are commonly overseeded into bermudagrass turf for winter color. When the overseeded grass persists beyond the spring; however, it becomes a weed. The ability of perennial ryegrass, Italian (annual) ryegrass, intermediate ryegrass, and hybrid bluegrass to persist in bermudagrass one year after seeding was determined. Perennial ryegrass, intermediate ryegrass, and Italian ryegrass produced acceptable ground cover in the spring after fall seeding. Hybrid bluegrass did not establish well, resulting in unacceptable cover. Perennial ryegrass generally persisted the most one year after seeding, either because of summer survival of plants or because of new germination the following fall. Plant counts one year after seeding were greater in the higher seeding rate treatment compared to the lower seeding treatment rate of perennial ryegrass, suggesting new germination had occurred. Plant counts one year after seeding plots with intermediate ryegrass or Italian ryegrass were attributed primarily to latent germination and not summer survival. Applications of foramsulfuron generally did not prevent overseeded species stand one year after seeding, supporting the conclusion of new germination. Although quality is less with intermediate ryegrass compared to perennial ryegrass, it transitions out easier than perennial ryegrass, resulting in fewer surviving plants one year later

    Trained laser-patterned carbon as high-performance mechanical sensors

    We describe the mechanical properties of turbostratically graphitized carbon films obtained by carbon laser-patterning (CLaP) and their application as bending or mechanical pressure sensors. Stable conductive carbonized films were imprinted on a flexible polyethylene terephthalate (PET) substrate by laser-induced carbonization. After initial gentle bending, i.e. training, these sponge-like porous films show a quantitative and reversible change in resistance upon bending or application of pressure in normal loading direction. Maximum response values of ΔR/R0 = 388% upon positive bending (tensile stress) and −22.9% upon negative bending (compression) are implicit for their high sensitivity towards mechanical deformation. Normal mechanical loading in a range between 0 and 500 kPa causes a response between ΔR/R0 = 0 and −15%. The reversible increase or decrease in resistance is attributed to compression or tension of the turbostratically graphitized domains, respectively.     Single-molecule-based super-resolution microscopy offers researchers a unique opportunity to quantify protein copy number with nanoscale resolution. However, while fluorescent proteins have been characterized for quantitative imaging using calibration standards, similar calibration tools for immunofluorescence with small organic fluorophores are lacking. Here we show that DNA origami, in combination with GFP antibodies, is a versatile platform for calibrating fluorophore and antibody labeling efficiency to quantify protein copy number in cellular contexts using super-resolution microscopy

    Engineered single- and multi-cell chemotaxis pathways in E. coli

    We have engineered the chemotaxis system of Escherichia coli to respond to molecules that are not attractants for wild-type cells. The system depends on an artificially introduced enzymatic activity that converts the target molecule into a ligand for an E. coli chemoreceptor, thereby enabling the cells to respond to the new attractant. Two systems were designed, and both showed robust chemotactic responses in semisolid and liquid media. The first incorporates an asparaginase enzyme and the native E. coli aspartate receptor to produce a response to asparagine; the second uses penicillin acylase and an engineered chemoreceptor for phenylacetic acid to produce a response to phenylacetyl glycine. In addition, by taking advantage of a ‘hitchhiker' effect in which cells producing the ligand can induce chemotaxis of neighboring cells lacking enzymatic activity, we were able to design a more complex system that functions as a simple microbial consortium. The result effectively introduces a logical ‘AND' into the system so that the population only swims towards the combined gradients of two attractants

    Skutterudite Results Shed Light on Heavy Fermion Physics

    Only few selected examples among the great diversity of anomalous rare earth skutterudite are reviewed. Focus is first given on PrFe4P12 in comparison with URu2Si2. For PrFe4P12, great progress has been made on determining the nature of the order parameter (OP). A non magnetic order parameter with a multipolar component emerges here while for URu2Si2 the nature of the so-called hidden order remains mysterious. The two systems have several similarities in their temperature--pressure (T, P) and magnetic field--temperature (H, T) phase diagrams, in their spin dynamics, in their nesting character and in their high sensitivity to impurities. Advances on one side must stimulate new views on the other. Besides general considerations on the choice of the OP, a simple basic problem is the treatment of the Kondo coupling in a system with low charge carrier number for the cases of uncompensated and compensated semi-metal.     Field Reentrance of the Hidden Order State of URu2Si2 under Pressure

    Combination of neutron scattering and thermal expansion measurements under pressure shows that the so-called hidden order phase of URu2Si2 reenters in magnetic field when antiferromagnetism (AF) collapses at H_AF (T). Macroscopic pressure studies of the HO-AF boundaries were realized at different pressures via thermal expansion measurements under magnetic field using a strain gauge. Microscopic proof at a given pressure is the reappearance of the resonance at Q_0=(1,0,0) under field which is correlated with the collapse of the AF Bragg reflections at Q_0.Comment: 5 pages, 6 figures, accepted for publication in J.
