Metaforat suomalaispoliitikkojen koronaa käsittelevissä tviiteissä

Tiivistelmä. Olen tutkinut kandidaatin työssäni suomalaispoliitikkojen tviiteissään käyttämiä metaforia. Aineistonani on ollut yhdentoista poliitikon tviitit aikaväliltä 1.3.–30.4.2020, ja aineistoni koostui yhteensä 104 tviitistä. Analysointivaiheessa olen hyödyntänyt Lakoffin ja Johnsonin (1980) metaforateoriaa, jonka pohjalta olen määritellyt metaforat ja muodostanut aineistosta luokat: personifikaatio, kilpailu- ja sotametaforat, spatiaaliset metaforat, säiliömetaforat, matkametaforat sekä ruumiillisuuteen ja aisteihin perustuvat metaforat. Aineistoni yleisimpiä metaforia olivat personifikaatioon liittyvät metaforat (41 %), joista keskeisimpiä olivat hallitusta elollistavat ilmaisut. Toisiksi yleisimpiä aineistoni metaforia olivat kilpailu- ja sotametaforat (18 %), joiden kautta koronasta muodostui kuva vihollisena ja vastustajana. Kilpailu- ja sotametaforien perusteella muodostin metaforan KORONA ON VIHOLLINEN. Kolmanneksi eniten aineistossani oli säiliömetaforia (17 %). Koronakriisistä rakentui kuva säiliönä, jonka sisässä ollaan ja josta on mahdollista kulkea ulos. Olen muodostanut säiliömetaforista metaforan KORONAKRIISI ON SÄILIÖ. Spatiaalisia metaforia aineistossani oli 15 %. Niiden kautta rakentui kuva aikajanasta ja koronasta, joka hahmottui janan tarkasteltavaan osaan x, johon mennyt ja tuleva aika suhteutettiin. Vähemmistönä aineistossani olivat matkametaforat (7 %) sekä ruumiillisuuteen ja aisteihin perustuvat metaforat (5 %). Matkametaforiin liittyvien ilmausten perusteella muodostin metaforan KORONASTA SELVIYTYMINEN ON MATKA. Yhteenvetona saamistani tuloksista voi sanoa, että poliitikkojen tviiteistä muodostuva kuva koronasta on, että korona on epätoivottava vihollinen, joka jättää jälkeensä tuhoa. Korona on nykyhetken keskiössä, ja olemme kriisin sisässä, josta ulospääsy ja selviytyminen tapahtuu tien kautta matkaamalla. Konventionaalisuuden periaatteen mukaisesti ajattelen, että saamani tulokset välittävät koronaan liittyviä ajattelutapoja. Poliitikkojen käyttämät ilmaisut ovat pitkälti metaforateoriassakin esiteltyjä, eli tuloksista ei tullut ilmi täysin uudentyyppisiä metaforia, joilla koronaa olisi kielennetty. Koronasta puhutaan siis tutuin ilmauksin, mikä on ymmärrettävää — onhan kyseessä uusi tilanne, jota pyritään ymmärtämään ja hallitsemaan

RAB24 facilitates clearance of autophagic compartments during basal conditions

Officially accepted for publication August 21, 2015.RAB24 belongs to a family of membrane traffic controlling RAB proteins and has been implicated to function in autophagy. Here we confirm the intracellular localization of RAB24 to autophagic vacuoles with immuno electron microscopy and cell fractionation, and show that prenylation and guanine nucleotide binding are necessary for the targeting of RAB24 to autophagic compartments. Further, we show that RAB24 plays a role in the maturation and/or clearance of autophagic compartments under nutrient-rich conditions, but not during short amino acid starvation. Quantitative electron microscopy showed an increase in the numbers of late autophagic compartments in cells silenced for RAB24, and mRFP-GFP-LC3 probe and autophagy flux experiments indicated that this was due to a hindrance in their clearance. Formation of autophagosomes was shown to be unaffected by RAB24 silencing with siRNA. A defect in aggregate clearance in the absence of RAB24 was also shown in cells forming polyglutamine aggregates. This study places RAB24 function in the termination of the autophagic process under nutrient-rich conditions.Peer reviewe

Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)

In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide.
