    On the precision loss in approximate homomorphic encryption

    Since its introduction at Asiacrypt 2017, the CKKS approximate homomorphic encryption scheme has become one of the most widely used and implemented homomorphic encryption schemes. Due to the approximate nature of the scheme, application developers using CKKS must ensure that the evaluation output is within a tolerable error of the corresponding plaintext computation. Choosing appropriate parameters requires a good understanding of how the noise will grow through the computation. A strong understanding of the noise growth is also necessary to limit the performance impact of mitigations to the attacks on CKKS presented by Li and Micciancio (Eurocrypt 2021). In this work we present a comprehensive noise analysis of CKKS, that considers noise coming both from the encoding and homomorphic operations. Our main contribution is the first average-case analysis for CKKS noise, and we also introduce refinements to prior worst-case noise analyses. We develop noise heuristics both for the original CKKS scheme and the RNS variant presented at SAC 2018. We then evaluate these heuristics by comparing the predicted noise growth with experiments in the HEAAN and FullRNS-HEAAN libraries, and by comparing with a worst-case noise analysis as done in prior work. Our findings show mixed results: while our new analyses lead to heuristic estimates that more closely model the observed noise growth than prior approaches, the new heuristics sometimes slightly underestimate the observed noise growth. This evidences the need for implementation-specific noise analyses for CKKS, which recent work has shown to be effective for implementations of similar schemes

    Protein Arginine Deiminases (PADs) catalyze the post-translational conversion of peptidyl-Arginine to peptidyl-Citrulline in a calcium-dependent, irreversible reaction. Evidence is emerging that PADs play a role in carcinogenesis. To determine the cancer-associated functional implications of PADs, we designed a small molecule PAD inhibitor (called Chor-amidine or Cl-amidine), and tested the impact of this drug on the cell cycle. Data derived from experiments in colon cancer cells indicate that Cl-amidine causes a G1 arrest, and that this was p53-dependent. In a separate set of experiments, we found that Cl-amidine caused a significant increase in microRNA-16 (miRNA-16), and that this increase was also p53-dependent. Because miRNA-16 is a putative tumor suppressor miRNA, and others have found that miRNA-16 suppresses proliferation, we hypothesized that the p53-dependent G1 arrest associated with PAD inhibition was, in turn, dependent on miRNA-16 expression. Results are consistent with this hypothesis. As well, we found the G1 arrest is at least in part due to the ability of Cl-amidine-mediated expression of miRNA-16 to suppress its\u27 G1-associated targets: cyclins D1, D2, D3, E1, and cdk6. Our study sheds light into the mechanisms by which PAD inhibition can protect against or treat colon cancer

    The Ambulatory Pediatric Association Fellowship in Pediatric Environmental Health: A 5-Year Assessment

    Background: Evidence is mounting that environmental exposures contribute to causation of disease in children. Yet few pediatricians are trained to diagnose, treat, or prevent disease of environmental origin. Objectives: To develop a cadre of future leaders in pediatric environmental health (PEH), the Ambulatory Pediatric Association (APA) launched a new 3-year fellowship in 2001—the world’s first formal training program in PEH. Sites were established at Boston Children’s Hospital, Mount Sinai School of Medicine, George Washington University, University of Cincinnati, and University of Washington. Fellows are trained in epidemiology, biostatistics, toxicology, risk assessment, and preventive medicine. They gain clinical experience in environmental pediatrics and mentored training in clinical research, policy development, and evidence-based advocacy. Thirteen fellows have graduated. Two sites have secured follow-on federal funding to enable them to continue PEH training. Discussion: To assess objectively the program’s success in preparing fellows for leadership careers in PEH, we conducted a mailed survey in 2006 with follow-up in 2007. Conclusions: Fifteen (88%) of 17 fellows and graduates participated; program directors provided information on the remaining two. Nine graduates are pursuing full-time academic careers, and two have leadership positions in governmental and environmental organizations. Ten have published one or more first-authored papers. Seven graduates are principal investigators on federal or foundation grants. The strongest predictors of academic success are remaining affiliated with the fellowship training site and devoting <20% of fellowship time to clinical practice. Conclusion: The APA fellowship program is proving successful in preparing pediatricians for leadership careers in PEH

    Security Guidelines for Implementing Homomorphic Encryption

    Fully Homomorphic Encryption (FHE) is a cryptographic primitive that allows performing arbitrary operations on encrypted data. Since the conception of the idea in [RAD78], it was considered a holy grail of cryptography. After the first construction in 2009 [Gen09], it has evolved to become a practical primitive with strong security guarantees. Most modern constructions are based on well-known lattice problems such as Learning with Errors (LWE). Besides its academic appeal, in recent years FHE has also attracted significant attention from industry, thanks to its applicability to a considerable number of real-world use-cases. An upcoming standardization effort by ISO/IEC aims to support the wider adoption of these techniques. However, one of the main challenges that standards bodies, developers, and end users usually encounter is establishing parameters. This is particularly hard in the case of FHE because the parameters are not only related to the security level of the system, but also to the type of operations that the system is able to handle. In this paper, we provide examples of parameter sets for LWE targeting particular security levels that can be used in the context of FHE constructions. We also give examples of complete FHE parameter sets, including the parameters relevant for correctness and performance, alongside those relevant for security. As an additional contribution, we survey the parameter selection support offered in open-source FHE libraries

    Loci influencing blood pressure identified using a cardiovascular gene-centric array

    Blood pressure (BP) is a heritable determinant of risk for cardiovascular disease (CVD). To investigate genetic associations with systolic BP (SBP), diastolic BP (DBP), mean arterial pressure (MAP) and pulse pressure (PP), we genotyped 50 000 single-nucleotide polymorphisms (SNPs) that capture variation in 2100 candidate genes for cardiovascular phenotypes in 61 619 individuals of European ancestry from cohort studies in the USA and Europe. We identified novel associations between rs347591 and SBP (chromosome 3p25.3, in an intron of HRH1) and between rs2169137 and DBP (chromosome1q32.1 in an intron of MDM4) and between rs2014408 and SBP (chromosome 11p15 in an intron of SOX6), previously reported to be associated with MAP. We also confirmed 10 previously known loci associated with SBP, DBP, MAP or PP (ADRB1, ATP2B1, SH2B3/ATXN2, CSK, CYP17A1, FURIN, HFE, LSP1, MTHFR, SOX6) at array-wide significance (P 2.4 10(6)). We then replicated these associations in an independent set of 65 886 individuals of European ancestry. The findings from expression QTL (eQTL) analysis showed associations of SNPs in the MDM4 region with MDM4 expression. We did not find any evidence of association of the two novel SNPs in MDM4 and HRH1 with sequelae of high BP including coronary artery disease (CAD), left ventricular hypertrophy (LVH) or stroke. In summary, we identified two novel loci associated with BP and confirmed multiple previously reported associations. Our findings extend our understanding of genes involved in BP regulation, some of which may eventually provide new targets for therapeutic intervention.</p

    A Multisite Preregistered Paradigmatic Test of the Ego-Depletion Effect

    We conducted a preregistered multilaboratory project (k = 36; N = 3,531) to assess the size and robustness of ego-depletion effects using a novel replication method, termed the paradigmatic replication approach. Each laboratory implemented one of two procedures that was intended to manipulate self-control and tested performance on a subsequent measure of self-control. Confirmatory tests found a nonsignificant result (d = 0.06). Confirmatory Bayesian meta-analyses using an informed-prior hypothesis (δ = 0.30, SD = 0.15) found that the data were 4 times more likely under the null than the alternative hypothesis. Hence, preregistered analyses did not find evidence for a depletion effect. Exploratory analyses on the full sample (i.e., ignoring exclusion criteria) found a statistically significant effect (d = 0.08); Bayesian analyses showed that the data were about equally likely under the null and informed-prior hypotheses. Exploratory moderator tests suggested that the depletion effect was larger for participants who reported more fatigue but was not moderated by trait self-control, willpower beliefs, or action orientation.</p
