    Umumnya olahraga yang paling sering dan paling banyak di minati oleh masyarakat di Indonesia yaitu olahraga permainan, seperti salah satunya cabang olahraga bola basket. teknik dasar dalam permainan bola basket seperti dribbling, passing, dan shooting agar dapat berhasil dalam bermain basket. Di antara beberapa jenis shooting di atas saya sebagai peneliti lebih tertarik dengan jenis shooting Lay up Shoot karna persentase bola masuk dengan teknik lay up shoot ini besar dibanding dengan teknik shooting lainnya. Adapun tujuan penelitian ini yaitu untuk mengetahui tingkat Keterampilan Lay Up Siswa Pada Permainan Bola Basket Di SMP Negeri 2 Seluma. Tempat penelitian ini dilakukan di lapangan bola basket SMP Negeri 2 Seluma, Waktu penelitian ini dilakukan 1 bulan yaitu pada bulan Juni-Juli 2021 sesuai dengan Surat Izin Penelitian. penelitian ini adalah penelitian deskriptif kuantitatif dengan menggunakan metode survei. Berdasarkan hasil penelitian dapat disimpulkan Setelah dilakukan pengukuran tentang survei tingkat keterampilan lay up siswa pada permainan bola basket di SMP Negeri 2 Seluma dari 30 siswa sebanyak 16 orang  dalam kategori sangat baik (53,33%). Sebanyak 3 orang dalam kategori sedang (10%). Sebanyak 5 orang dalam kategori rendah (16,67%). Sebanyak 6 orang dalam kategori sangat rendah (20%).     Characterization of an Insoluble and Soluble Form of Melanin Produced by Streptomyces cavourensis SV 21, a Sea Cucumber Associated Bacterium

    Melanin is a widely distributed and striking dark-colored pigment produced by countless living organisms. Although a wide range of bioactivities have been recognized, there are still major constraints in using melanin for biotechnological applications such as its fragmentary known chemical structure and its insolubility in inorganic and organic solvents. In this study, a bacterial culture of Streptomyces cavourensis SV 21 produced two distinct forms of melanin: (1) a particulate, insoluble form as well as (2) a rarely observed water-soluble form. The here presented novel, acid-free purification protocol of purified particulate melanin (PPM) and purified dissolved melanin (PDM) represents the basis for an in-depth comparison of their physicochemical and biological properties, which were compared to the traditional acid-based precipitation of melanin (AM) and to a synthetic melanin standard (SM). Our data show that the differences in solubility between PDM and PPM in aqueous solutions may be a result of different adjoining cation species, since the soluble PDM polymer is largely composed of Mg2+ ions and the insoluble PPM is dominated by Ca2+ ions. Furthermore, AM shared most properties with SM, which is likely attributed to a similar, acid-based production protocol. The here presented gentler approach of purifying melanin facilitates a new perspective of an intact form of soluble and insoluble melanin that is less chemical altered and thus closer to its original biological form

    Evaluating Patterns and Factors Related to Sleep Disturbances in Prostate Cancer Patients

    Prostate cancer patients may experience disturbed sleep as a result of their diagnosis or treatment. This study sought to evaluate disturbed sleep and excessive daytime sleepiness in newly diagnosed patients and those receiving androgen deprivation therapy (ADT). This study was conducted with 74 patients. Subjective data using the Pittsburgh Sleep Quality Index (PSQI) and Epworth Sleepiness Scale (ESS) and actigraphy data on ADT/ADT-naïve patients were collected. The prevalence of poor sleep quality, determined from PSQI and ESS scores, was 50% and 16.7% respectively. Those on ADT (n = 20) had poorer sleep quality as determined by significantly higher PSQI scores (70 vs. 40% scoring > 5) and were more likely to have poor sleep quality, sleep latency, and sleep efficiency than ADT-naïve patients (n = 40). Actigraphy data showed that ADT patients slept significantly longer (7.7 vs. 6.8 h), experienced a higher Fragmentation Index (48.3 vs. 37.4%), and had longer daytime nap duration (64.1 vs. 45.2 min) than ADT-naïve patients. The use of objective measures such as actigraphy in the clinical arena is recommended and may be used as a valuable tool for research into sleep assessment in prostate cancer patients

    The core problem in this study is the difficulty of determining classes for mentally retarded children. Mentally retarded children are children with special needs that must be considered in terms of education, to assist in the teaching and learning process for mentally retarded children, a very good class grouping is needed to support the teaching and learning process. The purpose of this study is to build a decision system that can determine and classify classes for mentally retarded children. The method used in this study is the Wight Product method by determining a criterion that exists in this study. The results of this study are a system that can determine classes for special needs that can assist in the learning process with mental retardation needs.Permasalahan inti pada penelitian ini yaitu sulitnya menentukan kelas untuk anak anak tunagrahita. Tunagrahita merupakan anak dalam kebutuha khusus yang harus diperhatikan dalam segi pendidikan, untuk membantuk dalam proses belajar mengajar pada anak tunagrahita diperlukan pengelompokan kelas yang sangat baik untuk menunjang proses blajar mengajar. Tujuan penelitian ini yaitu membangun sistem keputusan yang dapat menentukan dan mengelompokan kelas pada anak tunagrahita. Metode yang digunakan pada penelitian ini yaitu metode Wight Product dengan menentukan sebuah kriteria yang ada dalam penelitian ini. Hasil dari penelitian ini berupa sistem yang dapat menentukan kelas untuk kebutuhan khusus yang dapat membantu dalam proses belajar dengan kebutuhan tunagrahita

    The role of screening questionnaires in the assessment of risk and severity of obstructive sleep apnea — polysomnography versus polygraphy

    Obstructive sleep apnea (OSA) is a disease of significant importance, which may lead to numerous severe clinical consequences. The gold standard in the diagnosis of this sleep-related breathing disorder (SRBD) is polysomnography (PSG). However, due to the need for high expertise of staff who perform this procedure, its complexity, and relatively low availability, some simpler substitutes have been developed; among them is polygraphy (PG), which is most widely used.Also, there is a variety of questionnaires suitable to assess the pre-test probability and severity of OSA. The most frequently used ones are the STOP-BANG questionnaire (SBQ), NoSAS questionnaire, and Berlin questionnaire (BQ). However, they have different sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV) when being used in various populations. The aim of this study is to provide a concise and clinically-oriented review of the most frequently used questionnaires, with special attention to its strengths and limitations. Moreover, we discuss whether PSG or PG would be more preferred for confirming OSA diagnosis with the highest likelihood

    Occupational exposure to gases/fumes and mineral dust affect DNA methylation levels of genes regulating expression

    Many workers are daily exposed to occupational agents like gases/fumes, mineral dust or biological dust, which could induce adverse health effects. Epigenetic mechanisms, such as DNA methylation, have been suggested to play a role. We therefore aimed to identify differentially methylated regions (DMRs) upon occupational exposures in never-smokers and investigated if these DMRs associated with gene expression levels. To determine the effects of occupational exposures independent of smoking, 903 never-smokers of the LifeLines cohort study were included. We performed three genome-wide methylation analyses (Illumina 450 K), one per occupational exposure being gases/fumes, mineral dust and biological dust, using robust linear regression adjusted for appropriate confounders. DMRs were identified using comb-p in Python. Results were validated in the Rotterdam Study (233 never-smokers) and methylation-expression associations were assessed using Biobank-based Integrative Omics Study data (n = 2802). Of the total 21 significant DMRs, 14 DMRs were associated with gases/fumes and 7 with mineral dust. Three of these DMRs were associated with both exposures (RPLP1 and LINC02169 (2x)) and 11 DMRs were located within transcript start sites of gene expression regulating genes. We replicated two DMRs with gases/fumes (VTRNA2-1 and GNAS) and one with mineral dust (CCDC144NL). In addition, nine gases/fumes DMRs and six mineral dust DMRs significantly associated with gene expression levels. Our data suggest that occupational exposures may induce differential methylation of gene expression regulating genes and thereby may induce adverse health effects. Given the millions of workers that are exposed daily to occupational exposures, further studies on this epigenetic mechanism and health outcomes are warranted

    Comprehensive diagnostics of acute myeloid leukemia by whole transcriptome RNA sequencing

    Acute myeloid leukemia (AML) is caused by genetic aberrations that also govern the prognosis of patients and guide risk-adapted and targeted therapy. Genetic aberrations in AML are structurally diverse and currently detected by different diagnostic assays. This study sought to establish whole transcriptome RNA sequencing as single, comprehensive, and flexible platform for AML diagnostics. We developed HAMLET (Human AML Expedited Transcriptomics) as bioinformatics pipeline for simultaneous detection of fusion genes, small variants, tandem duplications, and gene expression with all information assembled in an annotated, user-friendly output file. Whole transcriptome RNA sequencing was performed on 100 AML cases and HAMLET results were validated by reference assays and targeted resequencing. The data showed that HAMLET accurately detected all fusion genes and overexpression of EVI1 irrespective of 3q26 aberrations. In addition, small variants in 13 genes that are often mutated in AML were called with 99.2% sensitivity and 100% specificity, and tandem duplications in FLT3 and KMT2A were detected by a novel algorithm based on soft-clipped reads with 100% sensitivity and 97.1% specificity. In conclusion, HAMLET has the potential to provide accurate comprehensive diagnostic information relevant for AML classification, risk assessment and targeted therapy on a single technology platform
