Identification of negative regulators of the Cutibacterium acnes-induced innate immune activation in human epidermal keratinocytes

Vizsgálatainkkal arra kĂvántunk választ adni, hogy - a BelsĹ‘ Kárpáti TĂ©rsĂ©g Ă©rintett eocĂ©n-oligocĂ©n flĂłra-spektruma alapján kimutathatĂł-e florisztikai Ă©s klĂmaváltozás, Ă©s milyen minĹ‘sĂ©gűek ezek a változások. Fosszilis flĂłrák taxonĂłmiai feldolgozása alapján az eocĂ©n - kora oligocĂ©n flĂłrák jelentĹ‘s florisztikai hasonlĂłságot mutatnak, a termofil elemek dominanciája mellett jellemzĹ‘ a mĂ©rsĂ©kelt klĂmaigĂ©nyű, lombhullatĂł elemek hiánya. EltĂ©rĂ©st jelent a nyitvatermĹ‘k hiánya, a szárnyas termĂ©sek ritka elĹ‘fordulása, valamint számos a kora oligocĂ©nre jellemzĹ‘ nemzetsĂ©g alárendelt szerepe a vizsgált eocĂ©n flĂłrákban. Ezzel szemben a kora oligocĂ©n/kĂ©sĹ‘ oligocĂ©n flĂłrák alapján gyökeres florisztikai változás tapasztalhatĂł a termofil elemek nagy rĂ©szĂ©nek eltűnĂ©sĂ©vel, Ă©s a mĂ©rsĂ©kelt elemek növekvĹ‘ arányával a kĂ©sĹ‘ oligocĂ©n flĂłrákban. LevĂ©lmorfolĂłgiai vizsgálatok, a szárnyas termĂ©sek elĹ‘fordulásbeli kĂĽlönbsĂ©gei egyĂ©rtelműen a vizsgált eocĂ©n vegetáciĂł zártabb jellegĂ©re, a kora oligocĂ©n flĂłrák esetĂ©ben csapadĂ©kbeli szezonalitásra utalnak. | The project was focused on the Eocene-Oligocene floral spektra of the Inner Carpathian Region in order to trace spatial and temporal pattern of floristic and climatic changes. Based on systematic studies of Eocene-Early Oligocene floras significant floristic similarity with the dominance of thermophilous taxa and the absence of temperate elements is indicated. Disparate characters of Eocene floras are the absence of gymnosperms, the rare occurrence of winged fruits and the subordinate role of genera characteristic in the Early Oligocene. In contrast, a definite floristic change is observable with the disappearance of most thermophilous elements and increasing ratio of temperate elements in the Late Oligocene assemblages. In order to trace the climate pattern of the studied area and time scale numerous methods were adopted ? coexistence approach, stomatal density, d13C measurements, leaf morphology. Quantitative estimates of climate variables indicate the slight decrease of mean annual temperature during the Early/Late Oligocene which is mainly attributable to the decrease of the mean temperature of the coldest month. Estimates of stomatal density suggest palaeoecological difference between the Slovenian and Hungarian localities of the Early Oligocene Palaeogene Basin. Leaf morphology, the disparate occurrence of winged fruits in the Eocene and Early Oligocene assemblages indicates a more open vegetation and seasonality in precipitation in the latter

A kutatás a magyar falusi, mezĹ‘városi parasztság öltözködĂ©sĂ©nek a 19. Ă©s 20. százada közepe közötti átalakulásával foglalkozott. Nem magukat az öltözeteket, hanem a parasztság Ă©letmĂłdjában Ă©s ennek következtĂ©ben a szemlĂ©letĂ©ben bekövetkezett változásokat vizsgáltuk, amelyek vĂ©geredmĂ©nyekĂ©nt számukra presztĂzskĂ©rdĂ©ssĂ© vált a városias öltözködĂ©s. Vizsgáltuk az Ă©letmĂłdot jelentĹ‘sen megváltoztatĂł innováciĂłk, Ăgy a vasĂşt, a kĂ©zművesipart felváltĂł textilipar, a varrĂłgĂ©p megjelenĂ©sĂ©nek következmĂ©nyeit, valamint a közművelĹ‘dĂ©s, ezen belĂĽl a nĂ©piskolai kĂ©zimunka-oktatás hatását. Tanulmányoztuk, hogy ellentĂ©tes törekvĂ©sek, divatáramlatok mikĂ©nt befolyásolták a polgárosodás folyamatát. Utaltunk azokra a jogszabályokra is, amelyek lehetĹ‘vĂ© tettĂ©k, hogy a feudális kor kötöttsĂ©gei után, a polgári kor törvĂ©nykezĂ©se rĂ©vĂ©n, a parasztság is rĂ©szese lehessen a gazdasági Ă©let ĂşjĂtásainak. EredmĂ©nyeinket az „ÉletmĂłd, szemlĂ©letmĂłd Ă©s a mĂłdi változása - a parasztság körĂ©ben a 19-20. század fordulĂłján” cĂmű kötetĂĽnkben adtuk közre. | The research dealt with the clothing of the peasants of Hungarian villages and towns between 1850s and 1950s. Its focus was not primarily on the variety of clothing and local diversities, but on the impact of economic and social changes on peasant clothing. The role of lifestyle changing innovations (e.g., the railway, sewing machine and textile manufacturing replacing craftsmen) and of changes in the educational system, particularly handiwork classes in public schools, has been explored, so were the influences of contradictory social aspirations (e.g., preserving local costumes) and fashions on urbanisation. The changes in the legal system have also been reviewed as they enabled peasants to enjoy the economic benefits of the transition from a feudal to a civil society. The results were published in ""The Changes in Lifestyle, Attitude and Fashion - among the Peasantry of turn of the 19-20. Century""
Szárazföldi környezeti rekonstrukciĂł, ökoszisztĂ©ma dinamika Ă©s klĂmaváltozás az oligocĂ©n folyamán a BelsĹ‘ Kárpáti TĂ©rsĂ©gben. = Terrestrial environment reconstruction, ecosystem dynamics and palaeoclimate during the Oligocene at the Inner Carpathian Region.
A BelsĹ‘ Kárpáti TĂ©rsĂ©g Ă©s az ezt körĂĽlvevĹ‘ Alp-Kárpát-Dinári tĂ©rsĂ©g terrĂ©n kollázsa olyan korai oligocĂ©n flĂłramaradványos rĂ©tegeket tartalmaz, amelyekek számos informáciĂłt hordoznak e terĂĽletek Ĺ‘sföldrajzi kapcsolatára Ă©s klĂmájára vonatkozĂłan. Vizsgálataink során számos taxonĂłmiai revĂziĂłt vĂ©geztĂĽnk hazai- Ă©s Ă©rintett kĂĽlföldi lelĹ‘helyek flĂłráin. A Budai- Ă©s BĂĽkki Tektonikai EgysĂ©g flĂłrája csaknem teljes megegyezĂ©st mutat. A flĂłra kizárĂłlag melegigĂ©nyes, Ĺ‘si tĂpusĂş un. paleotrĂłpusi elemet tartalmaz, amelyek egy rĂ©sze subxerofil Ă©s több esetben kimondottan trĂłpusi családok tagjai is megjelennek. Korai oligocĂ©n flĂłráink tehát a mainál dĂ©lebbi szĂ©lessĂ©gen, feltehetĹ‘en a "sclerofil paleogĂ©n öv" rĂ©szekĂ©nt, az eurĂłpai flĂłráktĂłl rĂ©szben izoláltan Ă©ltek. | The terrane collage of the Inner Carpathian Region and the surrounding Alpine-Carpathian-Dinaric region comprise fossiliferous strata with abundant plant remains that provide relevant information on the palaeogeographic context of the area and its climate. We focused on the taxonomical revision of numerous Hungarian and other relevant floras outside Hungary. The floras of the BĂĽkk and Buda Tectonic Units well correspond to each other. The flora comprises exclusively thermophilous, ancient elements a considerable part of which is subxerohytic. Temperate (arctotertiary) elements occurring with various taxa in the northern areas of Europe are completely absent from these floras. Despite high-grade floristic similarity climatological difference of the BĂĽkk and Buda Tectonic Units could be demonstrated. Drier climate was proved with high significancy for the BĂĽkk Tectonic Unit. The definitely uniform floristic pattern of the BĂĽkk and Buda Units and the Slovenian Early Oligocene supports the existence of an integrated Palaeogene Basin. Numerous elements of this special flora appear in coeval strata of the Alpine-Carpathian-Dinaric region. At the same time a flora with similar floristic composition has not been proved yet outside this terrane collage (the stable Europe). Our Early Oligocene floras must have flourished at more southern latitudes, isolated to a certain extent from the European floras, presumably as part of the 'Palaeogene Sclerophyllous Belt'
The early Oligocene flora and palaeo-environment of the Tard Clay Formation — latest results
The early Oligocene flora of the Tard Clay Formation is one of the most significant fossil floras of Hungary.
Although, the investigation of the highly diverse flora started as early as the 1940s, the numerous, both exotic and extinct genera and species are still raising many questions. The significant development in the application of microscopic methods for cuticular examinations as well as morphometric analyses helped to resolve many taxonomical and systematic questions in the last decades. Recent research activities have been focussing mainly on revealing the botanical affinities of fossil genera in order to provide more realistic ecological and climate reconstructions. This paper provides a summary of the latest results achieved by studies of the Tard Clay floras highlighting conclusions of systematic, palaeoclimate and palaeoecological analyses
STUDIES ON THE EFFECT OF PROPIONIBACTERIUM ACNES ON THE BARRIER PROPERTIES OF HUMAN IN VITRO CULTURED KERATINOCYTES
The human skin is heavily colonized by a specialized microbial community called the microbiome, which plays a complex role in the protection from the attack of external pathogens. This microbial flora can interact with the cells in the healthy skin and play a role in the maintenance of skin homeostasis, but also known to contribute to the pathogenesis of different diseases.
Our aim was to analyze whether the Propionibacterium acnes (P. acnes) bacterium, a member of the skin microbiome, or the bacterium induced pro-inflammatory mediator, TNF has any effect on the barrier properties of our epidermis.
For that, a confluent monolayer of in vitro cultured human immortalized keratinocytes (HPV-KER cells) were treated with different P.acnes strains and external TNF in different doses, and changes in the barrier properties were analyzed in real time using the xCELLIgence system. We also analyzed the effect of the bacterium on the mRNA expression changes of tight junction proteins claudin 1, 2, 4 (CLDN1, 2, 4), ocludin1 (OCL1) and ZO1 in these cultures using real-time RT-PCR.
Our results suggest that the bacterium induced an elevation, followed by a drop of the measured impedance values in the keratinocyte monolayers, possibly due to dynamic alterations of the barrier properties. The extent of these changes depended on the used P. acnes strain and the applied doses. Addition of TNF (1, 5, 10 ng/ml), a cytokine that is a know mediator of the P. acnes-induced innate immune and inflammatory events in keratinocytes also lead to a marked decrease of the measured impedance of the HPV-KER monolayers.
Real-time RT-PCR analysis of tight junction genes suggested that CLDN2 and 4 mRNAs were not present in these cells. However, the expression of CLDN1 decreased, whereas ZO1 and OCL1 mRNA levels increased in response to the bacterial treatment.
Our results suggest that our microbiome can modulate the barrier properties of the epidermis. It is possibly achieved, in one hand, through the direct regulation of genes playing a key role in the formation of cell-to-cell contacts. On the other hand, secreted factors, such as the TNFα pro-inflammatory mediator, may also have a direct effect and can loosen the epidermal barrier, possibly leading to the easier penetration of keratinocyte- as well as bacterial-derived factors to deeper tissue compartments. These findings strengthen the importance of a balanced interaction among the epidermal cells and our microbiome for the maintenance of healthy skin functions

Az emberi bĹ‘r egyik funkciĂłja az, hogy megvĂ©dje a szervezetet a patogĂ©n kĂłrokozĂłktĂłl Ă©s a káros környezeti hatásoktĂłl. Ismert, hogy bĹ‘rĂĽnk felszĂnĂ©n kĂĽlönbözĹ‘ baktĂ©riumok Ă©s mikrobák által kialakĂtott Ăşgynevezett kommenzális flĂłra találhatĂł. Az viszont kevĂ©sbĂ© ismert, hogy ezen mikrobiális flĂłra milyen hatással van a humán epidermális sejtekre Ă©s a bĹ‘r homeosztázisára.
CĂ©lunk annak a megismerĂ©se volt, hogy a kommenzális flĂłra egy tagja a Propionibacterium acnes (P.acnes) baktĂ©rium törzs hatással van-e az epidermális barrier funkciĂłjára. Az eredményeink arra utalnak, hogy baktérium kezelés hatására dózisfüggő változások indulnak, melyek mértéke eltér a különböző baktérium törzsek között.
Azt is vizsgáltuk, hogy baktĂ©rium kezelĂ©s hatására változik-e a konfluens monolayer HPV-KER sejt kultĂşrában a claudin 1,2,4 (CLDN 1,2,4), az occludin (OCL1) Ă©s a zonula occludens (ZO1) tight junction fehĂ©rjĂ©k mRNS szintű kifejezĹ‘dĂ©se. Az általunk használt sejtekben a vizsgált gĂ©nek közĂĽl a CLDN2 Ă©s 4 nem volt kimutathatĂł mRNS szinten. A CLDN1 mRNS szintű kifejezĹ‘dĂ©se csökkent, mĂg az OCL1 Ă©s a ZO1 szintje emelkedett baktĂ©rium kezelĂ©s hatására.
A P.acnes baktérium hatására veleszületett immun és gyulladásos folyamatok indulnak a keratinocita sejtekben. Ezért a sejteket TNF gyulladásos citokinnel (1, 5, 10g/ml) kezeltük és vizsgáltuk a barrier funkcióra gyakorolt hatását. Azt tapasztaltuk, hogy TNF kezelés, ezáltal a gyulladásos folyamatok hatására meglazulhat az epidermális barrier szerkezete, mely lehetővé teszi, hogy a baktérium által szekretált anyagok eljussanak a mélyebb szöveti rétegekbe.
Ezen eredmények alapján feltételezhető, hogy a mikrobiom hatással lehet a bőr barrier funkciójára. Valamint hogy ezen események fontos szerepet játszhatnak számos különböző bőrbetegség patogenezisében

A Propionibacterium acnes (P. acnes) tagja az egĂ©szsĂ©ges bĹ‘r mikroflĂłrájának, azonban bizonyos körĂĽlmĂ©nyek között patogĂ©n faktorkĂ©nt is viselkedhet kĂĽlönbözĹ‘ betegsĂ©gek kialakulásában, mint az acne vulgaris patogenezise során. Ismert, hogy humán epidermális keratinociták a baktĂ©rium hatására veleszĂĽletett immun Ă©s gyulladásos folyamatokat indĂtanak a patogĂ©nfelismerĹ‘ Toll-like receptorok (TLR) aktiválásán keresztĂĽl. KevĂ©sbĂ© ismert ezen folyamatok negatĂv szabályozása, mely a folyamatok lecsendesĂtĂ©se rĂ©vĂ©n vĂ©d a szabályozatlan, gyakran destruktĂv jellegű gyulladás kialakulásátĂłl.
Munkánk során ezĂ©rt cĂ©lul tűztĂĽk ki már más rendszerekben leĂrt, a TLR szignálfolyamatok negatĂv szabályozĂł elemeinek (SIGIRR, TOLLIP, TNIP1 Ă©s TNFAIP3) azonosĂtását Ă©s jellemzĂ©sĂ©t in vitro tenyĂ©sztett humán keratinocitákban (HPV-KER). Ezen szabályozĂłk mRNS szintű kifejezĹ‘dĂ©sĂ©t Ă©s ezek változását kĂĽlönbözĹ‘ filogenetikai csoportba tartozĂł P. acnes 889 Ă©s 6609 baktĂ©riumtörzs hatására valĂłs idejű RT-PCR mĂłdszerrel detektáltuk, fehĂ©rjeszintű vizsgálatainkat western blot analĂzissel vĂ©geztĂĽk.
Vizsgálva az NF-ÎşB transzkripciĂłs faktor aktiváciĂłját azt tapasztaltuk, hogy baktĂ©riumkezelĂ©s hatására az NF-ÎşB mennyisĂ©ge dĂłzisfĂĽggĹ‘ mĂłdon nĹ‘tt a sejtmagban, ami a P. acnes hatására indulĂł dĂłzisfĂĽggĹ‘ TLR aktiváciĂłra utal. EredmĂ©nyeink azt mutatják, hogy mind a nĂ©gy vizsgált negatĂv szabályozĂł mRNS szinten kifejezĹ‘dik HPV-KER sejtekben. Ezen felĂĽl a TNIP1 Ă©s a TNFAIP3 expressziĂłja gyors, tranziens mĂłdon emelkedik P. acnes kezelĂ©s hatására. A baktĂ©rium dĂłzisának emelĂ©sĂ©vel párhuzamosan dĂłzisfĂĽggĹ‘ mĂłdon fokozĂłdik mindkĂ©t gĂ©n mRNS szintű kifejezĹ‘dĂ©se. Ezen folyamatok hátterĂ©ben az NF-ÎşB dĂłzisfĂĽggĹ‘ aktiváciĂłja Ă©s nukleáris transzlokáciĂłja állhat. MĂg a TOLLIP mennyisĂ©ge csökken, addig a TNFAIP3 nĹ‘ fehĂ©rjeszinten a baktĂ©rium mennyisĂ©gĂ©nek emelĂ©sĂ©vel. A kĂĽlönbözĹ‘ klinikai izolátumĂş P. acnes törzsek hatásában nem találtunk kĂĽlönbsĂ©get a negatĂv szabályozĂłk kifejezĹ‘dĂ©sĂ©re.

Cutibacterium acnes regulates the epidermal barrier properties of HPV-KER human immortalized keratinocyte cultures
Our skin provides a physical barrier to separate the internal part of our body from the environment. Maintenance of complex barrier functions is achieved through anatomical structures in the skin, the stratified squamous epithelium specialized junctional organelles, called tight junctions (TJs). Several members of our microbial communities are known to affect the differentiation state and function of the colonized organ. Whether and how interactions between skin cells and cutaneous microbes, including Cutibacterium acnes (C. acnes), modify the structure and/or function of our skin is currently only partly understood. Thus, in our studies, we investigated whether C. acnes may affect the epidermal barrier using in vitro model systems. Real-time cellular analysis showed that depending on the keratinocyte differentiation state, the applied C. acnes strains and their dose, the measured impedance values change, together with the expression of selected TJ proteins. These may reflect barrier alterations, which can be partially restored upon antibiotic-antimycotic treatment. Our findings suggest that C. acnes can actively modify the barrier properties of cultured keratinocytes, possibly through alteration of tight cell-to-cell contacts. Similar events may play important roles in our skin, in the maintenance of cutaneous homeostasis
