    The Effect of Vincristine Sulphate on the Axoplasmic Flow of Proteins in Cultured Sympathetic Neurons

    The effect of vincristine sulphate on the axoplasmic flow of labelled proteins in neurites of chick embryo sympathetic neurons growing in tissue culture was studied by autoradiography. In control neurons most of the 3H-proteins synthesized during a 90-min pulse with a 3H-amino acid were localized in cell bodies. There was a diminishing gradient of labelled proteins in the neurites which was highest in portions adjacent to the cell bodies and lowest at the periphery. During a physiological chase there was a gradual increase in the amount of label in the neurites, so that after a 15-h chase even the most peripheral portions were well labelled. This indicates that a portion of the labelled proteins synthesized in the cell bodies are transported peripherally into the neurites. The centrifugal movement of labelled proteins in neurites was markedly decreased when cells were grown in medium containing 10 µg/ml vincristine sulphate. After a 15-h chase in the presence of drug only a small amount of label was in the peripheral portion of the neurites. Treatment with vincristine did not decrease the rate of amino acid incorporation or alter the rate of protein turnover during the course of the experiment. Thus an explanation of the results based on an altered rate of total cell protein synthesis or degradation is unlikely. The capacity of sympathetic neurons to take up and concentrate exogenous [3H]norepinephrine in their neurites was only slightly reduced by vincristine. This indicates that at least some cellular activities requiring metabolic energy are relatively unaffected by the interruption in axoplasmic flow caused by vincristine and that the mechanism by which vincristine interferes with axoplasmic flow does not involve general cellular toxicity. The major morphological differences between control and vincristine-treated neurons were the absence of microtubules and the presence of crystal-like structures within the cells.     Truth Table Invariant Cylindrical Algebraic Decomposition by Regular Chains

    A new algorithm to compute cylindrical algebraic decompositions (CADs) is presented, building on two recent advances. Firstly, the output is truth table invariant (a TTICAD) meaning given formulae have constant truth value on each cell of the decomposition. Secondly, the computation uses regular chains theory to first build a cylindrical decomposition of complex space (CCD) incrementally by polynomial. Significant modification of the regular chains technology was used to achieve the more sophisticated invariance criteria. Experimental results on an implementation in the RegularChains Library for Maple verify that combining these advances gives an algorithm superior to its individual components and competitive with the state of the art

    Design, development and test of shuttle/Centaur G-prime cryogenic tankage thermal protection systems

    The thermal protection systems for the shuttle/Centaur would have had to provide fail-safe thermal protection during prelaunch, launch ascent, and on-orbit operations as well as during potential abort. The thermal protection systems selected used a helium-purged polyimide foam beneath three rediation shields for the liquid-hydrogen tank and radiation shields only for the liquid-oxygen tank (three shields on the tank sidewall and four on the aft bulkhead). A double-walled vacuum bulkhead separated the two tanks. The liquid-hydrogen tank had one 0.75-in-thick layer of foam on the forward bulkhead and two layers on the larger area sidewall. Full scale tests of the flight vehicle in a simulated shuttle cargo bay that was purged with gaseous nitrogen gave total prelaunch heating rates of 88,500 Btu/hr and 44,000 Btu/hr for the liquid-hydrogen and -oxygen tanks, respectively. Calorimeter tests on a representative sample of the liquid-hydrogen tank sidewall thermal protection system indicated that the measured unit heating rate would rapidly decrease from the prelaunch rate of approx 100 Btu/hr/sq ft to a desired rate of less than 1.3 Btu/hr/sq ft once on orbit

    CMB anisotropy from spatial correlations of clusters of galaxies

    The SZ effect from clusters of galaxies is a dominant source of secondary CMB anisotropy in the low-redshift universe. We present analytic predictions for the CMB power spectrum from massive halos arising from the SZ effect. Since halos are discrete, the power spectrum consists of a Poisson and a correlation term. The latter is always smaller than the former, which is dominated by nearby bright rich clusters. In practice however, those bright clusters are easy to indentify and can thus be subtracted from the map. After this subtraction, the correlation term dominates degree-scale fluctuations over the Poisson term, as the main contribution to the correlation term comes from distant clusters. We find that the correlation term is detectable by Planck experiment. Since the degree scale spectrum is quite insensitive to the highly uncertain core structures of halos, our predictions are robust on these scales. Measuring the correlation term on degree scales thus cleanly probes the clustering of distant halos. This has not been measured yet, mainly because optical and X-ray surveys are not sufficiently sensitive to include such distant clusters and groups. Our analytic predictions are also compared to adiabatic hydrodynamic simulations. The agreement is remarkably good, down to ten arcminutes scales, indicating that our predictions are robust for the Planck experiment. Below ten arcminute scales, where the details of the core structure dominates the power spectrum, our analytic and simulated predictions might fail. In the near future, interferometer and bolometer array experiments will measure the SZ power spectrum down to arcminutes scales, and yield new insight into the physics of the intrahalo medium.Comment: 9 pages, 4 figures. submitted to Proceedings of the 9th Marcel Grossmann meetin

    Reviewing research evidence and the case of participation in sport and physical recreation by black and minority ethnic communities

    The paper addresses the implications of using the process of systematic review in the many areas of leisure where there is a dearth of material that would be admitted into conventional Cochrane Reviews. This raises important questions about what constitutes legitimate knowledge, questions that are of critical import not just to leisure scholars, but to the formulation of policy. The search for certainty in an area that lacks conceptual consensus results in an epistemological imperialism that takes a geocentric form. While clearly, there is a need for good research design whatever the style of research, we contend that the wholesale rejection of insightful research is profligate and foolhardy. A mechanism has to be found to capitalise on good quality research of whatever form. In that search, we draw upon our experience of conducting a review of the material available on participation in sport and physical recreation by people from Black and minority ethnic groups. The paper concludes with a proposal for a more productive review process that makes better use of the full panoply of good quality research available. © 2012 © 2012 Taylor & Francis

    A model study of the effects of climatic precipitation changes on ground temperatures

    Temperature changes at the Earth surface propagate into the subsurface and leave a thermal signature in the underlying soil and rock. Inversions of subsurface temperature measurements yield reconstructions of ground surface temperature (GST) histories that provide estimates of climatic changes. A question remaining in the interpretation of reconstructed GST histories is the extent to which GST changes reflect changes principally in surface air temperature (SAT), or whether other factors may be significant. Here we use a Land Surface Processes (LSP) model to examine the influence of precipitation changes on GST and subsurface temperature and moisture fields on annual to decadal timescales. We model soil and vegetation conditions representative of a prairie region in the southern Great Plains of North America and force the model with meteorological data synthesized from a typical year in the region. Model responses are observed after changes in the amount of daily precipitation, the intensity and frequency of daily precipitation, and the diurnal and seasonal timing of precipitation. We show that: (1) increasing daily precipitation cools mean annual GST, (2) increasing the intensity and reducing the frequency of daily precipitation, while holding the annual amount of precipitation constant, cools mean annual GST, and (3) shifting maximum precipitation to occur in the warmest months cools mean annual GST. We compare modeled results to observed precipitation changes during the 20th century and conclude that the observed precipitation changes would cause only small changes to GST within the modeled region, on the order of 0.1 K or less
