Morphological heterogeneity of HeLa cell mitochondria visualized by a modified diaminobenzidine staining technique

The diaminobenzidine (DAB) technique for the ultrastructural localization of sites of cytochrome c oxidase activity in animal tissues has been adapted to the visualization of mitochondria in animal cells growing in culture. The modified technique allows the staining of mitochondria in all cells in coverslip preparatins for light microscopy. Electron microscopy of thin sections of material treated by this method has revealed that all mitochondrial profiles within a cell (and only these) are stained and they exhibit a well preserved size and internal structure. Coverslip cultures of synchronized and unsynchronized HeLa (F-315) cells stained with the DAB reaction were examined under oil immersion. In the majority of the cells, mitochondria were recognized as discrete bodies in the thinner peripheral portion of the cytoplasm. This observation indicates that in a large proportion of HeLa F-315 cells, at least under the growth conditions used here, the mitochondrial complement is dividied into distinct organelles. This examination also revealed a considerable morphological heterogeneity of mitochondria, which exhibited an ovoid or short rod-like or, less frequently, long filamentous shape, with some evidence of branching. The variability in mitochondrial morphology appeared to be far more prounced between different cells than within individual cells; this cellular heterogeneity was not related in any obvious way to cell-cycle-dependent changes

Toward quantum processing in molecules: A THz-bandwidth coherent memory for light

The unusual features of quantum mechanics are enabling the development of technologies not possible with classical physics. These devices utilize nonclassical phenomena in the states of atoms, ions, and solid-state media as the basis for many prototypes. Here we investigate molecular states as a distinct alternative. We demonstrate a memory for light based on storing photons in the vibrations of hydrogen molecules. The THz-bandwidth molecular memory is used to store 100-fs pulses for durations up to 1ns, enabling 10,000 operational time bins. A Descriptive Case Study of Staff Development In Two Seventh-day Adventist Districts as They Relate to Potential Change

Problem. Leadership at the superintendent level is paramount in the process of change in Seventh-day Adventist schools. The superintendent/education director of state-sized districts, known as conferences, is responsible for staff development programs. How a superintendent performs his or her duties in the area of staff development is the focus of this study. Purpose. The purpose of this study is to describe staff development programs of two Seventh-day Adventist districts who have a reputation for being active in promoting staff development among their teachers. I wanted to know what the superintendents of these districts were doing in this active mode of staff development. This study is intended to provide data for other districts to follow similar paths of staff development. Method. Two conferences/districts in North America were identified as being exemplary in terms of their staff development programs. Using a case study approach, the district programs were analyzed utilizing interviews, observations, surveys, documents, and records. A quantitative analysis of existing survey data (Profile \u2795) was compared with findings from the qualitative portions of the study. Results. Several recurring motifs emerged in the area of staff development within the two exemplary Seventh-day Adventist school districts: (1) the superintendents of these districts have a clear sense of organizational vision and values, (2) the superintendents actively promote innovative practices to be used in staff development, and (3) the superintendents challenge their teachers to assess and revise their own actions in order to improve the likelihood of success for their students. I discovered that these two exemplary school districts incorporate the following characteristics in their staff-development program: (1) a supportive, cooperative climate in the schools and district, (2) adequate resources provided by the superintendents, (3) implementation of the Joyce and Showers Training Model, (4) time during the school day for collaboration, and (5) realistic goals set by the district

Extending electron orbital precession to the molecular case: Can orbital alignment be used to observe wavepacket dynamics?

The complexity of ultrafast molecular photoionization presents an obstacle to the modelling of pump-probe experiments. Here, a simple optimized model of atomic rubidium is combined with a molecular dynamics model to predict quantitatively the results of a pump-probe experiment in which long range rubidium dimers are first excited, then ionized after a variable delay. The method is illustrated by the outline of two proposed feasible experiments and the calculation of their outcomes. Both of these proposals use Feshbach 87Rb2 molecules. We show that long-range molecular pump-probe experiments should observe spin-orbit precession given a suitable pump-pulse, and that the associated high-frequency beat signal in the ionization probability decays after a few tens of picoseconds. If the molecule was to be excited to only a single fine structure state state, then a low-frequency oscillation in the internuclear separation would be detectable through the timedependent ionization cross section, giving a mechanism that would enable observation of coherent vibrational motion in this molecule.Comment: 9 pages, 10 figures, PRA submissio

Factors which Cause Families to Home School Their Children in Northeast Tennessee

The problem which this researcher investigated centered around the motivational factors which caused parents to educate their children at home. The purpose of this study was to assess the factors as perceived to cause families to home school their children. This study of home school education offered the opportunity to examine the dynamics within home schooling families as they conducted a non-traditional educational alternative. The study may provide information and insight to assist public school policy makers in considering inclusion of program components presently perceived as missing. Two home school associations in upper East Tennessee responded to a home school questionnaire. The data from the questionnaire presented a demographic overview of home school families and indicated the motivational factors for home schooling as opposed to enrollment in public schools. Six research questions were answered, and twelve hypotheses were tested using the Chi Square Test of Independence. The Chi Square Analysis revealed a statistically significant relationship between the academic achievement in math of home school children and the types of curriculums used by parents. The Chi Square Analysis revealed no statistically significant relationship between the academic achievement of home school children and the types of group activities used by parents, and the hindrance of current state home school laws The results indicate that parents chose to home school their children because they felt it was the responsibility of parents to provide a child\u27s education, and to control the moral environment of the child. Public school system personnel should make an effort to acquire information from registered home school families as to the causes and reasons families are opting to leave public education
