    Effectiveness, inequality and ethos in three English schools

    Many school effectiveness studies have focussed on schools serving disadvantaged communities. ‘Schools matter most for underprivileged and/or initially low achieving students. Effective or ineffective schools are especially effective or ineffective for these students’ (Scheerens & Bosker 1997: 96). Analysing how schools can improve in socially disadvantaged communities is essential. The nature of a school cannot, of course, remove social inequality or social exclusion, but it can have a significant positive impact (Sammons 2007). In a study of improving schools in disadvantaged settings, Muijs et al. (2004) found they focus on a number of factors: teaching and learning; enhancing leadership capacity; creating an information rich environment; creating a positive school culture; building a learning community; promoting continuous professional development; involving parents; and engaging external support. This study of three English schools supports Muijs et al.’s findings, but points also to the importance of an ‘ethos of inclusion’.     Effects of nitrate and phosphate supply on chromophoric and fluorescent dissolved organic matter in the Eastern Tropical North Atlantic: a mesocosm study

    In open-ocean regions, as is the Eastern Tropical North Atlantic (ETNA), pelagic production is the main source of dissolved organic matter (DOM) and is affected by dissolved inorganic nitrogen (DIN) and phosphorus (DIP) concentrations. Changes in pelagic production under nutrient amendments were shown to also modify DOM quantity and quality. However, little information is available about the effects of nutrient variability on chromophoric (CDOM) and fluorescent (FDOM) DOM dynamics. Here we present results from two mesocosm experiments ("Varied P" and "Varied N") conducted with a natural plankton community from the ETNA, where the effects of DIP and DIN supply on DOM optical properties were studied. CDOM accumulated proportionally to phytoplankton biomass during the experiments. Spectral slope (S) decreased over time indicating accumulation of high molecular weight DOM. In Varied N, an additional CDOM portion, as a result of bacterial DOM reworking, was determined. It increased the CDOM fraction in DOC proportionally to the supplied DIN. The humic-like FDOM component (Comp.1) was produced by bacteria proportionally to DIN supply. The protein-like FDOM component (Comp.2) was released irrespectively to phytoplankton or bacterial biomass, but depended on DIP and DIN concentrations. Under high DIN supply, Comp.2 was removed by bacterial reworking, leading to an accumulation of humic-like Comp.1. No influence of nutrient availability on amino acid-like FDOM component in peptide form (Comp.3) was observed. Comp.3 potentially acted as an intermediate product during formation or degradation of Comp.2. Our findings suggest that changes in nutrient concentrations may lead to substantial responses in the quantity and quality of optically active DOM and, therefore, might bias results of the applied in situ optical techniques for an estimation of DOC concentrations in open-ocean regions

    Effects of varied nitrate and phosphate supply on polysaccharidic and proteinaceous gel particles production during tropical phytoplankton bloom experiments

    Gel particles such as the polysaccharidic transparent exopolymer particles (TEP) and the proteinaceous Coomassie stainable particles (CSP) play an important role in marine biogeochemical and ecological processes like particle aggregation and export, or microbial nutrition and growth. So far, effects of nutrient availability or of changes in nutrient ratios on gel particle production and fate are not well understood. The tropical ocean includes large oxygen minimum zones, where nitrogen losses due to anaerobic microbial activity result in a lower supply of nitrate relative to phosphate to the euphotic zone. Here, we report of two series of mesocosm experiments that were conducted with natural plankton communities collected from the eastern tropical North Atlantic (ETNA) close to Cape Verde in October 2012. The experiments were performed to investigate how different phosphate (experiment 1, Varied P: 0.15–1.58 μmol L−1) or nitrate (experiment 2, Varied N: 1.9–21.9 μmol L−1) concentrations affect the abundance and size distribution of TEP and CSP. In the days until the bloom peak was reached, a positive correlation between gel particle abundance and Chl a concentration was determined, linking the release of dissolved gel precursors and the subsequent formation of gel particles to autotrophic production. After the bloom peak, gel particle abundance remained stable or even increased, implying a continued partitioning of dissolved into particulate organic matter after biomass production itself ceased. During both experiments, differences between TEP and CSP dynamics were observed; TEP were generally more abundant than CSP. Changes in size distribution indicated aggregation of TEP after the bloom, while newly formed CSP decomposed. Abundance of gel particles clearly increased with nitrate concentration during the second experiment, suggesting that changes in [DIN] : [DIP] ratios can affect gel particle formation with potential consequences for carbon and nitrogen cycling as well as food web dynamics in tropical ecosystems

    Bax regulates neuronal Ca2+ homeostasis

    Excessive Ca(2+) entry during glutamate receptor overactivation (\u22excitotoxicity\u22) induces acute or delayed neuronal death. We report here that deficiency in bax exerted broad neuroprotection against excitotoxic injury and oxygen/glucose deprivation in mouse neocortical neuron cultures and reduced infarct size, necrotic injury, and cerebral edema formation after middle cerebral artery occlusion in mice. Neuronal Ca(2+) and mitochondrial membrane potential (Δψm) analysis during excitotoxic injury revealed that bax-deficient neurons showed significantly reduced Ca(2+) transients during the NMDA excitation period and did not exhibit the deregulation of Δψm that was observed in their wild-type (WT) counterparts. Reintroduction of bax or a bax mutant incapable of proapoptotic oligomerization equally restored neuronal Ca(2+) dynamics during NMDA excitation, suggesting that Bax controlled Ca(2+) signaling independently of its role in apoptosis execution. Quantitative confocal imaging of intracellular ATP or mitochondrial Ca(2+) levels using FRET-based sensors indicated that the effects of bax deficiency on Ca(2+) handling were not due to enhanced cellular bioenergetics or increased Ca(2+) uptake into mitochondria. We also observed that mitochondria isolated from WT or bax-deficient cells similarly underwent Ca(2+)-induced permeability transition. However, when Ca(2+) uptake into the sarco/endoplasmic reticulum was blocked with the Ca(2+)-ATPase inhibitor thapsigargin, bax-deficient neurons showed strongly elevated cytosolic Ca(2+) levels during NMDA excitation, suggesting that the ability of Bax to support dynamic ER Ca(2+) handling is critical for cell death signaling during periods of neuronal overexcitation

    Avaliação in vitro da atividade antibacteriana da água ozonizada frente ao Staphylococcus aureus

    Staphylococcus aureus belongs to the normal flora of the skin, mucosa and nasopharynx of several animal species, including man, but it is also associated to illnesses such as abscesses, bacteremia, endocarditis and osteomyelitis, besides showing resistance to multiple drugs. The purpose of this paper was to evaluate the disinfecting ability of ozone when dissolved in water. Suspensions of Staphylococcus aureus with concentrations varying from 10(6) to 10(16) microorganisms/ml were prepared. One milliliter of each recently prepared suspension was added to 99 ml of distilled water (with or without previous ozonization) contained in a crystal reactor. Aliquots of 0.1 ml of this new suspension were taken at various time intervals and, then, serially diluted and inoculated on plaques. The data indicated that there was difference in the disinfecting effect when distilled water was used with and without previous ozonization.O Staphylococcus aureus faz parte da microbiota da pele, mucosas e nasofaringe de várias espécies animais, incluindo o homem, mas também está associado a enfermidades como abscessos, bacteremias, endocardites e osteomielites, além de se mostrar resistente a múltiplas drogas. Este trabalho foi realizado com a finalidade de avaliar o efeito do gás ozônio, dissolvido em água, sobre o S. aureus. Foram preparadas suspensões de S. aureus, com concentrações variando de 10(6) a 10(16) microrganismos/ml. De cada suspensão recém-preparada foi retirado 1 ml e adicionado a 99 ml de água destilada estéril (com ou sem prévia ozonização), em um reator de cristal. Foram preparadas diluições seriadas (1/10) das suspensões testadas e inoculado 0,1 ml de cada suspensão em Tryptic Soy Agar, incubadas a 37ºC por 24 h, quando então se procedeu à contagem das UFC. Os resultados obtidos mostraram que o tempo máximo para a inativação total das bactérias tratadas com água previamente ozonizada (0,6 mg/ml) foi de 5’25" e, para a água não previamente ozonizada, foi de 23’45", indicando um efeito antibacteriano mais rápido da água previamente ozonizada, frente ao S. aureus

    Impacts of hurricanes on surface water flow within a wetland

    s u m m a r y Between 2001 and 2005, seven category 3 or higher major hurricanes made landfall within the US. The hydrologic impacts of these distinct climatic phenomena frequently occurring in wetland watersheds, however, are not well understood. The focus of this study was to evaluate the impacts of hurricane wind and rainfall conditions on water velocity and water elevations within the study wetland, the Florida Everglades. Specifically water velocity data was measured near two tree islands (Gumbo Limbo (GL) and Satin Leaf (SL)) and wind speed, water elevation, and rainfall were obtained from nearby wind observation stations. During the direct impacts of the hurricanes (Hurricanes Katrina and Wilma), water speed, flow direction, and hydraulic gradients were altered, and the extent of variation was positively related to wind characteristics, with significant alterations in flow direction at depth during Hurricane Wilma due to higher wind speeds. After the direct impacts, the longer lasting effect of hurricanes (time scale of a few days) resulted in altered flow speeds that changed by 50% or less. These longer lasting changes in flow speeds may be due to the redistribution of emergent vegetation

    Water column biogeochemistry of oxygen minimum zones in the eastern tropical North Atlantic and eastern tropical South Pacific Oceans

    Recent modeling results suggest that oceanic oxygen levels will decrease significantly over the next decades to centuries in response to climate change and altered ocean circulation. Hence the future ocean may experience major shifts in nutrient cycling triggered by the expansion and intensification of tropical oxygen minimum zones (OMZs). There are numerous feedbacks between oxygen concentrations, nutrient cycling and biological productivity; however, existing knowledge is insufficient to understand physical, chemical and biological interactions in order to adequately assess past and potential future changes. We investigated the pelagic biogeochemistry of OMZs in the eastern tropical North Atlantic and eastern tropical South Pacific during a series of cruise expeditions and mesocosm studies. The following summarizes the current state of research on the influence of low environmental oxygen conditions on marine biota, viruses, organic matter formation and remineralization with a particular focus on the nitrogen cycle in OMZ regions. The impact of sulfidic events on water column biogeochemistry, originating from a specific microbial community capable of highly efficient carbon fixation, nitrogen turnover and N2O production is further discussed. Based on our findings, an important role of sinking particulate organic matter in controlling the nutrient stochiometry of the water column is suggested. These particles can enhance degradation processes in OMZ waters by acting as microniches, with sharp gradients enabling different processes to happen in close vicinity, thus altering the interpretation of oxic and anoxic environments
