    Pharmacy students' experience of technology-enhanced learning during the COVID-19 pandemic

    With the advent of the COVID-19 pandemic, pharmacy students and educators experienced an abrupt shift as programmes that were previously taught exclusively in-person were then predominantly taught online. This sudden change provided little time for students to prepare for the new learning environment. The study objective was to explore pharmacy students' experiences of technology-enhanced learning during the COVID-19 pandemic. A cross-sectional survey was developed and distributed by email to all 3rd year (N = 76) and 4th year (N = 68) pharmacy students undertaking an MPharm programme in an Irish university. A total of 32 responses were collected, including 20 third year and 12 fourth year pharmacy students (response rates of 26.3% and 17.6%, respectively). The majority of respondents reported good or very good internet speed (71%) and stability (59%). Almost all were confident or very confident using Canvas (97%) prior to the onset of online learning. Respondents preferred engaging with other students in-person rather than online for coursework (68.8%) and learning new material (56.3%). Students favoured face-to-face delivery, with a recording of the session available online afterwards, for lectures (68.8%), workshops (50%) and tutorials (56.3%). Analysis of free-text comments indicates that respondents used recorded content to support exam revision and that a key drawback of online learning was social isolation. Pharmacy students favoured a blended learning approach, with in-person learning being recorded to support study and revision. Students' experience of TEL during the pandemic should be considered in the development and ongoing review of pharmacy programmes

    Burnt mounds, or fulachtaí fiadh as they are known in Ireland, are probably the most common prehistoric site type in Ireland and Britain. Typically Middle–Late Bronze Age in age (although both earlier and later examples are known), they are artefact-poor and rarely associated with settlements. The function of these sites has been much debated with the most commonly cited uses being for cooking, as steam baths or saunas, for brewing, tanning, or textile processing. A number of major infrastructural development schemes in Ireland in the years 2002–2007 revealed remarkable numbers of these mounds often associated with wood-lined troughs, many of which were extremely well-preserved. This afforded an opportunity to investigate them as landscape features using environmental techniques – specifically plant macrofossils and charcoal, pollen, beetles, and multi-element analyses. This paper summarises the results from eight sites from Ireland and compares them with burnt mound sites in Great Britain. The fulachtaí fiadh which are generally in clusters, are all groundwater-fed by springs, along floodplains and at the bases of slopes. The sites are associated with the clearance of wet woodland for fuel; most had evidence of nearby agriculture and all revealed low levels of grazing. Multi-element analysis at two sites revealed elevated heavy metal concentrations suggesting that off-site soil, ash or urine had been used in the trough. Overall the evidence suggests that the most likely function for these sites is textile production involving both cleaning and/or dyeing of wool and/or natural plant fibres and as a functionally related activity to hide cleaning and tanning. Whilst further research is clearly needed to confirm if fulachtaí fiadh are part of the ‘textile revolution’ we should also recognise their important role in the rapid deforestation of the wetter parts of primary woodland and the expansion of agriculture into marginal areas during the Irish and British Bronze Ages

    Epigenome-wide association study of alcohol consumption in N = 8161 individuals and relevance to alcohol use disorder pathophysiology:identification of the cystine/glutamate transporter SLC7A11 as a top target

    Alcohol misuse is common in many societies worldwide and is associated with extensive morbidity and mortality, often leading to alcohol use disorders (AUD) and alcohol-related end-organ damage. The underlying mechanisms contributing to the development of AUD are largely unknown; however, growing evidence suggests that alcohol consumption is strongly associated with alterations in DNA methylation. Identification of alcohol-associated methylomic variation might provide novel insights into pathophysiology and novel treatment targets for AUD. Here we performed the largest single-cohort epigenome-wide association study (EWAS) of alcohol consumption to date (N = 8161) and cross-validated findings in AUD populations with relevant endophenotypes, as well as alcohol-related animal models. Results showed 2504 CpGs significantly associated with alcohol consumption (Bonferroni p value < 6.8 × 10(−8)) with the five leading probes located in SLC7A11 (p = 7.75 × 10(−108)), JDP2 (p = 1.44 × 10(−56)), GAS5 (p = 2.71 × 10(−47)), TRA2B (p = 3.54 × 10(−42)), and SLC43A1 (p = 1.18 × 10(−40)). Genes annotated to associated CpG sites are implicated in liver and brain function, the cellular response to alcohol and alcohol-associated diseases, including hypertension and Alzheimer’s disease. Two-sample Mendelian randomization confirmed the causal relationship of consumption on AUD risk (inverse variance weighted (IVW) p = 5.37 × 10(−09)). A methylation-based predictor of alcohol consumption was able to discriminate AUD cases in two independent cohorts (p = 6.32 × 10(−38) and p = 5.41 × 10(−14)). The top EWAS probe cg06690548, located in the cystine/glutamate transporter SLC7A11, was replicated in an independent cohort of AUD and control participants (N = 615) and showed strong hypomethylation in AUD (p < 10(−17)). Decreased CpG methylation at this probe was consistently associated with clinical measures including increased heavy drinking days (p < 10(−4)), increased liver function enzymes (GGT (p = 1.03 × 10(−21)), ALT (p = 1.29 × 10(−6)), and AST (p = 1.97 × 10(−8))) in individuals with AUD. Postmortem brain analyses documented increased SLC7A11 expression in the frontal cortex of individuals with AUD and animal models showed marked increased expression in liver, suggesting a mechanism by which alcohol leads to hypomethylation-induced overexpression of SLC7A11. Taken together, our EWAS discovery sample and subsequent validation of the top probe in AUD suggest a strong role of abnormal glutamate signaling mediated by methylomic variation in SLC7A11. Our data are intriguing given the prominent role of glutamate signaling in brain and liver and might provide an important target for therapeutic intervention

    Salve Regina University Act on Climate: Strategic Plan for the University to Reach State Carbon Neutrality Goals

    In order to become more sustainable and meet the mandate set by the 2021 Rhode Island Act on Climate law (RI General Law §42-6.2), Salve Regina University must work to reach net-zero greenhouse gas emissions by the year 2050. Action to meet these standards begins now and must be continually built upon to ensure that Salve Regina University, as leader in Rhode Island, is always working for a more sustainable future. Throughout the Spring 2022 semester, students of the BIO-140: Humans and Their Environment course instructed by Dr. Jameson Chace have researched ways in which Salve Regina can begin on the path to zero greenhouse gas emissions today. By focusing on change in the areas of energy, transportation, food, financial investments, and sequestration, Salve Regina can reduce the greenhouse gas emissions of today for a more sustainable tomorrow. Recommendations are broken into three time periods. Action for today to achieve by 2030 include improving energy efficiency, installing the first electric vehicle (EV) parking/charging stations, increasing carbon sequestration, reducing beef in the campus diet, and assessing the carbon impact of university financial holdings. Actions to be initiated soon and to be achieved by 2040 include shifting away from natural gas heating when system renewals take place, increasing EV parking to meet rising demand, during turnover replace current university vehicles with electric or hybrid, continuing with sequestration efforts on campus, begin phasing out high carbon diet items, and by 2040 the university investment portfolio should be carbon neutral. If carbon neutrality can be reached by 2050 the most challenging aspects of campus life that need to change will require planning now and thoughtful implementation. The class in 2022 envisions a campus in 2050 where solar lights illuminate campus and buildings through the night, all university vehicles and most faculty and staff vehicles are electric and are found charging during the day at solar powered charging stations, dining services in Miley supports community agriculture and includes incentives for meatless and low carbon meal plans, the university has become a leader in low carbon/green market investing demonstrating how careful planning can reap high returns, and carbon sequestration on campus grounds has maximized such that off campus carbon offsets are established with local land trusts to complete the carbon neutrality goals.     A Genome Wide Association Scan of Bovine Tuberculosis Susceptibility in Holstein-Friesian Dairy Cattle

    peer-reviewedBackground: Bovine tuberculosis is a significant veterinary and financial problem in many parts of the world. Although many factors influence infection and progression of the disease, there is a host genetic component and dissection of this may enlighten on the wider biology of host response to tuberculosis. However, a binary phenotype of presence/absence of infection presents a noisy signal for genomewide association study. Methodology/Principal Findings: We calculated a composite phenotype of genetic merit for TB susceptibility based on disease incidence in daughters of elite sires used for artificial insemination in the Irish dairy herd. This robust measure was compared with 44,426 SNP genotypes in the most informative 307 subjects in a genome wide association analysis. Three SNPs in a 65 kb genomic region on BTA 22 were associated (i.e. p,1025, peaking at position 59588069, p = 4.0261026) with tuberculosis susceptibility. Conclusions/Significance: A genomic region on BTA 22 was suggestively associated with tuberculosis susceptibility; it contains the taurine transporter gene SLC6A6, or TauT, which is known to function in the immune system but has not previously been investigated for its role in tuberculosis infection

    Pathogenic mtDNA mutations causing mitochondrial myopathy: The need for muscle biopsy.

    Pathogenic mitochondrial tRNA (mt-tRNA) gene mutations represent a prominent cause of primary mitochondrial DNA (mtDNA)-related disease despite accounting for only 5%-10% of the mitochondrial genome.(1,2) Although some common mt-tRNA mutations, such as the m.3243A>G mutation, exist, the majority are rare and have been reported in only a small number of cases.(3) The MT-TP gene, encoding mt-tRNA(Pro), is one of the less polymorphic mt-tRNA genes, and only 5 MT-TP mutations have been reported as a cause of mitochondrial muscle disease to date (table e-1 at Neurology.org/ng, P6-10). We report 5 patients with myopathic phenotypes, each harboring different pathogenic mutations in the MT-TP gene, highlighting the importance of MT-TP mutations as a cause of mitochondrial muscle disease and the requirement to study clinically relevant tissue

    There is mounting evidence that parasitic infections change nutrient utilisation within the tissues, and that this is responsible for the reduction in animal performance that has been observed. Feeding forages that contain condensed tannins (CT) are thought to alleviate the impact of parasite infection on amino acid (AA) and protein metabolism by improving protein supply post-ruminally. However, there has been no quantification of how nutrients are partitioned in the lamb fed fresh forages during a parasitic infection. Therefore, the objective of this study was to quantify the partitioning of AA between the gastrointestinal tract (GIT), liver and the hind limb tissues (muscle, skin, fat) in lambs during an established parasite infection. It was hypothesised that the feeding of CT would alter the partitioning of AA between the GIT, liver and hind limbs in lambs with an established parasite burden due to the increased availability of dietary AA to the small intestine. This hypothesis was tested in two separate experiments, which had a similar experimental design. In the first experiment (Experiment One; 1999) lambs were fed fresh Lucerne (Medicago sativa; contains no CT). In Experiment Two, which was conducted in 2000, the lambs were fed fresh Sulla (Hedysarum coronarium; 2.2% CT; Experiment Two). One week prior to infection, permanent indwelling catheters were placed in the mesenteric artery, and the mesenteric, portal and hepatic veins and vena cava for blood sampling. Additional permanent catheters were placed in the mesenteric vein (upstream from the sampling catheter) and abdominal aorta for infusion of para-aminohippuric acid (PAH) and indocyanin green (ICG) respectively, to measure plasma flow across the splanchnic tissues (PAH) and the hind limbs (ICG). A permanent Teflon cannula was fitted in the abomasum for the infusion of [1-13C]-valine and [35S]-cysteine (Chapters Five, Six and Seven only) on day 48 post infection to measure valine and cysteine kinetics across the mesenteric-drained viscera (MDV), portal-drained viscera (PDV), liver, total splanchnic tissues (TSP; PDV + liver) and hind limbs. A temporary catheter was inserted into the jugular vein two days before the start of blood sampling for the infusion of deuterium oxide (D2O), and [13C]-sodium bicarbonate and [35S]-sulfate (Chapters Five, Six and Seven only) on day 45 post infection, and [3,4-3H]-valine on day 48 post infection. Lambs were dosed with 6 000 L3 T. colubriformis larvae for 6 d (n=5) or kept as parasite free controls (n=6). Faecal egg production was monitored every second day from day 22 to day 48 post infection and total intestinal worm burdens were determined at slaughter. Blood was continuously collected from the mesenteric, portal and hepatic veins, the mesenteric artery and the vena cava in 2-hour aliquots. Plasma was harvested and AA and metabolite concentrations measured and the specific radioactivity (SRA) and isotopic enrichment (IE) of valine and cysteine were determined. After the completion of blood sampling, but while the [3, 4-3H]-valine infusate was still being administered, the sheep were euthanased by an intravenous overdose of sodium pentobarbitone. Tissue samples were rapidly collected from the sheep in the following order: skin, muscle (biceps femoris), liver, duodenum, ileum, spleen, mesenteric lymph nodes and thymus. Digesta was also sampled from the abomasum and ileum after slaughter in order for the apparent absorption of AA to be determined. The results from Experiment One (Lucerne-fed lambs) suggest that there is no re-partitioning of AA from the posterior hind limbs to the GIT and liver during an established infection- The changes that occurred within the PDV suggests that an established parasitic infection may trigger a localised alteration in AA metabolism and/or protein turnover without significantly changing the metabolism of AA and proteins in tissues peripheral to the TSP tissues and impacting negatively on the growth of the parasitised lambs. In Experiment Two (Sulla-fed lambs) a reduction in feed intake was likely to be the reason for the alterations in the first pass metabolism of AA in the TSP tissues due to the decreased apparent AA absorption by the MDV observed in the parasitised lambs. However, the results from this experiment are in agreement with those from Experiment One confirming that there is no increase in partitioning of AA from the hind limbs to the GIT or liver during an established parasite infection. Although a statistical comparison cannot be made between the data in Experiment One (Lucerne-fed) and Experiment Two (Sulla-fed), it appears that the beneficial effects of feeding CT during a parasitic infection is due to the reduction in larval establishment in the GIT of the lamb, rather than increased AA availability. In conclusion, an established infection imposes no measurable metabolic cost on the lamb, when feed intake is not reduced. When feed intake is reduced, there is no detectable mobilisation of protein from the hind limb. Therefore, localised or other sources of AA and/or energy substrates may be utilised
