    The project that I spent my time working on was the White River Pacific Right Bank Flood Protection Project. In January of 2009 the White River significantly flooded neighborhoods and a county park in the City of Pacific. The overall goal of this Capital Improvement Project is to reduce the potential for White River flooding and to improve environmental conditions for ESA-listed Chinook salmon, other wildlife and more. Design alternatives (that have yet to be decided on) include many components that will happen over multiple years including property acquisition, levee construction and floodplain restoration along the right bank (River and Floodplain Management Section). Currently, the project is in the feasibility and environmental review stage, and a draft Environmental Impact Statement (EIS) is being prepared in order to evaluate project alternatives

    Promoter Binding by the Adr1 Transcriptional Activator May Be Regulated by Phosphorylation in the DNA-Binding Region

    Background: Post-translational modification regulates promoter-binding by Adr1, a Zn-finger transcriptional activator of glucose-regulated genes. Support for this model includes the activation of an Adr1-dependent gene in the absence of Adr1 protein synthesis, and a requirement for the kinase Snf1 for Adr1 DNA-binding. A fusion protein with the Adr1 DNA-binding domain and a heterologous activation domain is glucose-regulated, suggesting that the DNA binding region is the target of regulation. Methodology/Principal Findings: Peptide mapping identified serine 98 adjacent to the Zn-fingers as a phosphorylation site. An antibody specific for phosphorylated serine 98 on Adr1 showed that the level of phosphorylated Adr1 relative to the level of total Adr1 decreased with glucose derepression, in a Snf1-dependent manner. Relative phosphorylation decreased in a PHO85 mutant, and this mutant constitutively expressed an Adr1-dependent reporter. Pho85 did not phosphorylate Adr1 in vitro, suggesting that it affects Adr1 indirectly. Mutation of serine 98 to the phosphomimetic amino acid aspartate reduced in vitro DNA-binding of the recombinant Adr1 DNA-binding domain. Mutation to aspartate or alanine affected activation of a reporter by full-length Adr1, and in vivo promoter binding. Conclusions/Significance: Mutation of Adr1 serine 98 affects in vitro and in vivo DNA binding, and phosphorylation of serine 98 in vivo correlates with glucose availability, suggesting that Adr1 promoter-binding is regulated in part by serine 98 phosphorylation

    Consanguinity and susceptibility to infectious diseases in humans.

    Studies of animal populations suggest that low genetic heterozygosity is an important risk factor for infection by a diverse range of pathogens, but relatively little research has looked to see whether similar patterns exist in humans. We have used microsatellite genome screen data for tuberculosis (TB), hepatitis and leprosy to test the hypothesis that inbreeding depression increases risk of infection. Our results indicate that inbred individuals are more common among our infected cases for TB and hepatitis, but only in populations where consanguineous marriages are common. No effect was found either for leprosy, which is thought to be oligogenic, or for hepatitis in Italy where consanguineous marriages are rare. Our results suggest that consanguinity is an important risk factor in susceptibility to infectious diseases in humans

    Blood pressure in primary school children in Uganda: a cross-sectional survey.

    BACKGROUND: Non-communicable diseases are an emerging concern in sub-Saharan Africa, and risks for these conditions are often based on exposures in early life, with premonitory signs developing during childhood. The prevalence of hypertension has been reported to be high in African adults, but little is known about blood pressure in African children. We studied prevalence and risk factors for high blood pressure (HBP) among school children in central Uganda. METHODS: Two urban and five rural schools were randomly selected from government schools in Wakiso district, Uganda. Questionnaires were administered and anthropometric measures taken. Blood pressure (BP) was measured three times in one sitting (on day 1) and the average compared to internationally-used normograms. Children with BP >95th percentile were re-tested at two additional sittings (day 2 and day 3) within one week, and at two further follow up visits over a period of six months. Those with sustained HBP were referred for further investigation. RESULTS: Of 552 students included, 539 completed the initial assessments (days 1-3) of whom 92 (17.1%) had HBP at the initial sitting. Age (adjusted odds ratio (aOR) 1.29 (95% confidence interval 1.14, 1.47), p< 0.001), body mass index (1.70 (1.25-2.31) p = 0.001) and soil-transmitted helminths (2.52 (1.04-6.11), 0.04) were associated with increased prevalence of HBP at the initial sitting. After further investigation, sustained HBP was seen in 14 children, yielding an estimated prevalence of 3.8% allowing for losses to follow up. Four children required treatment. CONCLUSION: It is feasible to measure blood pressure accurately in the school setting. A high HBP prevalence on initial readings gave cause for concern, but follow up suggested a true HBP prevalence commensurate with international normograms. Extended follow up is important for accurate assessment of blood pressure among African children

    Analysis of public health risks from consumption of informally marketed milk in sub-Saharan African countries

    Despite policies to discourage them, informal milk markets account for over 80% of milk sales in most sub-Saharan African (SSA) countries. Informal milk market agents include farmer dairy co-operatives, small traders using bicycles and public or private transport and small retail outlets, such as dairy kiosks, and shops. Studies conducted by the International Livestock Research Institute (ILRI) and national collaborators (e.g., in Kenya1) show that convenient delivery and lower prices (reflecting lower handling and processing costs) are the principal benefits for consumers. Current milk handling and safety regulations in most SSA countries are derived from models in industrialised countries. These may not be appropriate for local market conditions where such regulations may unnecessarily inhibit efficient milk marketing. An important step in developing targeted policies more supportive of market participation of the majority is to collect quantitative and qualitative information about milk-borne health risks under different production and marketing situations. This paper gives an over-view of on-going activities in central Kenya aimed at assessing public health risks from informally marketed milk and presents preliminary results of milk quality and handling practices of informal milk market agents and consumers

    Feature Improvement and Cost Reduction of Baitcasting Fishing Reels for Emerging Markets

    Baitcasting fishing reels are a challenging product to sell to new users in emerging markets. Their complex and less-than-intuitive design make them poor candidates for a novice fisherman selecting his or her first fishing reel. Based upon manufacturer constraints and design requirements, our team lowered the price point and improved the usability of the Okuma Cerros baitcasting fishing reel to make it more appealing to a wider range of consumers, especially in emerging markets. This project resulted in a three-phase redesign: reducing cost via alternative materials and replacing bearings with bushings; prototyping a simplified cast control system; and proposing an improved user interface
