    Visible and near infrared spectroscopy of Hayabusa re-entry using semi-autonomous tracking

    A ground-based tracking camera and co-aligned slit-less spectrograph were used to measure the spectral signature of visible radiation emitted from the Hayabusa capsule as it entered into the Earth's atmosphere in June 2010. Good quality spectra were obtained that showed the presence of radiation from the heat shield of the vehicle and the shock-heated air in front of the vehicle. An analysis of the black body nature of the radiation concluded that the peak average temperature of the surface was about (3100±100) K

    Hayabusa, an unmanned Japanese spacecraft, was launched to study and collect samples from the surface of the asteroid 25143 Itokawa. In June 2010, the Hayabusa spacecraft completed it’s seven year voyage. The spacecraft and the sample return capsule (SRC) re-entered the Earth’s atmosphere over the central Australian desert at speeds on the order of 12 km/s. This provided a rare opportunity to experimentally investigate the radiative heat transfer from the shock-compressed gases in front of the sample return capsule at true-flight conditions. This paper reports on the results of observations from a tracking camera situated on the ground about 100 km from where the capsule experienced peak heating during re-entry

    Nucleon axial and pseudoscalar form factors from the covariant Faddeev equation

    We compute the axial and pseudoscalar form factors of the nucleon in the Dyson-Schwinger approach. To this end, we solve a covariant three-body Faddeev equation for the nucleon wave function and determine the matrix elements of the axialvector and pseudoscalar isotriplet currents. Our only input is a well-established and phenomenologically successful ansatz for the nonperturbative quark-gluon interaction. As a consequence of the axial Ward-Takahashi identity that is respected at the quark level, the Goldberger-Treiman relation is reproduced for all current-quark masses. We discuss the timelike pole structure of the quark-antiquark vertices that enters the nucleon matrix elements and determines the momentum dependence of the form factors. Our result for the axial charge underestimates the experimental value by 20-25% which might be a signal of missing pion-cloud contributions. The axial and pseudoscalar form factors agree with phenomenological and lattice data in the momentum range above Q^2 ~ 1...2 GeV^2.Comment: 17 pages, 7 figures, 1 tabl

The experimental results demonstrated the possibility of simultaneously observing both particle-like and wave-like behavior of a quantum via quantum entanglement.     Complementarity and Young's interference fringes from two atoms

    The interference pattern of the resonance fluorescence from a J=1/2 to J=1/2 transition of two identical atoms confined in a three-dimensional harmonic potential is calculated. Thermal motion of the atoms is included. Agreement is obtained with experiments [Eichmann et al., Phys. Rev. Lett. 70, 2359 (1993)]. Contrary to some theoretical predictions, but in agreement with the present calculations, a fringe visibility greater than 50% can be observed with polarization-selective detection. The dependence of the fringe visibility on polarization has a simple interpretation, based on whether or not it is possible in principle to determine which atom emitted the photon.Comment: 12 pages, including 7 EPS figures, RevTex.     Binding of an antimicrobial peptide to bacterial cells: interaction with different species, strains and cellular components

    Antimicrobial peptides (AMPs) selectively kill bacteria by disrupting their cell membranes, and are promising compounds to fight drug-resistant microbes. Biophysical studies on model membranes have characterized AMP/membrane interactions and the mechanism of bilayer perturbation, showing that accumulation of cationic peptide molecules in the external leaflet leads to the formation of pores ("carpet" mechanism). However, similar quantitative studies on real cells are extremely limited. Here, we investigated the interaction of the dansylated PMAP23 peptide (DNS-PMAP23) with a Gram-positive bacterium, showing that 10(7) bound peptide molecules per cell are needed to kill it. This result is consistent with our previous finding for Gram-negative strains, where a similar high threshold for killing was determined, demonstrating the general relevance of the carpet model for real bacteria. However, in the case of the Gram-positive strain, this number of molecules even exceeds the total surface available on the bacterial membrane. The high affinity of DNS-PMAP23 for the anionic teichoic acids of the Gram-positive cell wall, but not for the lipopolysaccharides of Gram-negative bacteria, provides a rationale for this finding. To better define the role of anionic lipids in peptide/cell association, we studied DNS-PMAP23 interaction with E. coli mutant strains lacking phosphatidylglycerol and/or cardiolipin. Surprisingly, these strains showed a peptide affinity similar to that of the wild type. This finding was rationalized by observing that these bacteria have an increased content of other anionic lipids, thus maintaining the total membrane charge essentially constant. Finally, studies of DNS-PMAP23 association to dead bacteria showed an affinity an order of magnitude higher compared to that of live cells, suggesting strong peptide binding to intracellular components that become accessible after membrane perturbation. This effect could play a role in population resistance to AMP action, since dead bacteria could protect the surviving cells by sequestering significant amounts of peptide molecules. Overall, our data indicate that quantitative studies of peptide association to bacteria can lead to a better understanding of the mechanism of action of AMPs

    Inhibition of diacylglycerol lipase beta modulates lipid and endocannabinoid levels in the ex vivo human placenta

    IntroductionLipids and fatty acids are key components in metabolic processes of the human placenta, thereby contributing to the development of the fetus. Placental dyslipidemia and aberrant activity of lipases have been linked to diverse pregnancy associated complications, such as preeclampsia and preterm birth. The serine hydrolases, diacylglycerol lipase alpha and beta (DAGL alpha, DAGL beta) catalyze the degradation of diacylglycerols, leading to the formation of monoacylglycerols (MAG), including one main endocannabinoid 2-arachidonoylglycerol (2-AG). The major role of DAGL in the biosynthesis of 2-AG is evident from various studies in mice but has not been investigated in the human placenta. Here, we report the use of the small molecule inhibitor DH376, in combination with the ex vivo placental perfusion system, activity-based protein profiling (ABPP) and lipidomics, to determine the impact of acute DAGL inhibition on placental lipid networks. MethodsDAGL alpha and DAGL beta mRNA expression was detected by RT-qPCR and in situ hybridization in term placentas. Immunohistochemistry staining for CK7, CD163 and VWF was applied to localize DAGL beta transcripts to different cell types of the placenta. DAGL beta activity was determined by in- gel and MS-based activity-based protein profiling (ABPP) and validated by addition of the enzyme inhibitors LEI-105 and DH376. Enzyme kinetics were measured by EnzChek (TM) lipase substrate assay. Ex vivo placental perfusion experiments were performed +/- DH376 [1 mu M] and changes in tissue lipid and fatty acid profiles were measured by LC-MS. Additionally, free fatty acid levels of the maternal and fetal circulations were determined. ResultsWe demonstrate that mRNA expression of DAGL beta prevails in placental tissue, compared to DAGL alpha (p <= 0.0001) and that DAGL beta is mainly located to CK7 positive trophoblasts (p <= 0.0001). Although few DAGL alpha transcripts were identified, no active enzyme was detected applying in-gel or MS-based ABPP, which underlined that DAGL beta is the principal DAGL in the placenta. DAGL beta dependent substrate hydrolysis in placental membrane lysates was determined by the application of LEI-105 and DH376. Ex vivo pharmacological inhibition of DAGL beta by DH376 led to reduced MAG tissue levels (p <= 0.01), including 2-AG (p <= 0.0001). We further provide an activity landscape of serine hydrolases, showing a broad spectrum of metabolically active enzymes in the human placenta. DiscussionOur results emphasize the role of DAGL beta activity in the human placenta by determining the biosynthesis of 2-AG. Thus, this study highlights the special importance of intra-cellular lipases in lipid network regulation. Together, the activity of these specific enzymes may contribute to the lipid signaling at the maternal-fetal interface, with implications for function of the placenta in normal and compromised pregnancies.Molecular Physiolog

    Quantitative conditional quantum erasure in two-atom resonance fluorescence

    We present a conditional quantum eraser which erases the a priori knowledge or the predictability of the path a photon takes in a Young-type double-slit experiment with two fluorescent four-level atoms. This erasure violates a recently derived erasure relation which must be satisfied for a conventional, unconditional quantum eraser that aims to find an optimal sorting of the system into subensembles with particularly large fringe visibilities. The conditional quantum eraser employs an interaction-free, partial which-way measurement which not only sorts the system into optimal subsystems with large visibility but also selects the appropriate subsystem with the maximum possible visibility.
