    Interactions of arbuscular mycorrhizal fungi, critical loads of nitrogen deposition, and shifts from native to invasive species in a southern California shrubland

    Anthropogenic nitrogen (N) deposition and invasive species are causing declines in global biodiversity, and both factors impact the diversity and functioning of arbuscular mycorrhizal (AM) fungi. Shifts in arbuscular mycorrhizal fungal (AMF) communities can generate feedback to native plants and affect their success, as was observed in California’s coastal sage scrub, which is a Mediterranean-type shrubland threatened by invasive grasses. As vegetation-type conversion from native shrubland to exotic annual grassland increased along a gradient of increasing N deposition, the richness of native plant species and of spore morphotypes decreased. Rapid declines in all plant and fungal values occurred at the critical load (CL) of 10–11 kg N·ha−1·year−1, indicating that AM fungi respond to the same environmental signals as the plants, and can be used to assess CL. Shrub root colonization also decreased along the N gradient, but colonization of the invasive grass was dominated by a fine AMF endophyte that was unresponsive to elevated N. A greenhouse experiment to assess AMF functioning showed that the native shrub Artemisia californica Less. had a negative growth response to an inoculum from high-N but not low-N soils, whereas the invasive grass Bromus rubens L. had a positive response to both inocula. Differential functioning of AM fungi under N deposition may in part explain vegetation-type conversion and the decline of this native shrubland

    The 1924 Workers' Incident at Ruimveldt British Guiana and the development of working people's organisation

    In 1924 stevedores and other categories of dock workers in Georgetown, British Guiana, went on strike in response to a call by the British Guiana Labour Union (BGLU). In a demonstration of solidarity, estate workers from several sugar plantations along the East Bank of Demerara embarked upon a march to Georgetown. At Plantation Ruimveldt, the workers confronted a detachment of police officers and members of the military forces. As a result of an order to shoot into the crowd thirteen workers were killed and twenty-four wounded. The dissertation has been able to establish the '1924 Workers' Incident at Ruimveldt' as a watershed in Guianese working people's struggle by highlighting its prominence among other moments of overt resistance through its impact upon workers' organisation. The significance of the Incident is also brought out in the new relationship which developed between the British government, the Colonial Office and the British TUC, on the one hand, and the Guianese labour leaders on the other. In pursuit of this task, the dissertation addresses the following: the organisational structures of the working people at the formal and non-formal or 'street corner' levels; the leadership which emerged from the ranks of the working people and the middle class; the impact of 'grassroots' organisation in fostering working peoples' consciousness and co-operation among members of the major racial sections in the country; and their attempts to establish links with progressive individuals and organisations within Guiana and throughout the international community

    Phytoplankton and Nutrient Dynamics in a Tidally Dominated Eutrophic Estuary: Daily Variability and Controls on Bloom Formation

    To better understand nutrient dynamics and factors that promote the initiation of algal blooms, the Lafayette River, a tidal subestuary of Chesapeake Bay that experiences seasonal algal blooms, was sampled daily for a period of 54 d in the fall of 2005. Three phytoplankton blooms (chl a concentrations exceeding twice the average of monthly measurements from 2000 to 2009) occurred during this period: a mixed bloom of Akashiwo sanguinea and Gymnodinium sp., a monospecific Skeletonema costatum bloom, and a monospecific Gymnodinium sp. bloom. Over the sampling period, nutrient concentrations increased following precipitation events and were elevated between bloom periods but low during blooms. All measured forms of nitrogen (N) were positively correlated with dinoflagellate abundance with a lag time of 3 to 5 d, suggesting a possible triggering effect, although not by any single form of N. Concentrations of NO2--reached 10 µM between September and October, indicative of incomplete nitrification. Over a 24 h period, nutrient concentrations and chl a biomass varied by an order of magnitude (0.1 to 1 µM N and 4.5 to 45 µg chl a l-1, respectively) and were strongly linked to the tidal phase. In the highly eutrophic Lafayette River, when nutrient concentrations are high, phytoplankton blooms appear to be controlled by spring-neap tidal modulation and wind-driven mixing; however, picoplankton abundance does not appear to be linked to the spring-neap tidal cycle

    Evidence against Wolbachia symbiosis in Loa loa

    BACKGROUND: The majority of filarial nematode species are host to Wolbachia bacterial endosymbionts, although a few including Acanthocheilonema viteae, Onchocerca flexuosa and Setaria equina have been shown to be free of infection. Comparisons of species with and without symbionts can provide important information on the role of Wolbachia symbiosis in the biology of the nematode hosts and the contribution of the bacteria to the development of disease. Previous studies by electron microscopy and PCR have failed to detect intracellular bacterial infection in Loa loa. Here we use molecular and immunohistological techniques to confirm this finding. METHODS: We have used a combination of PCR amplification of bacterial genes (16S ribosomal DNA [rDNA], ftsZ and Wolbachia surface protein [WSP]) on samples of L. loa adults, third-stage larvae (L3) and microfilariae (mf) and immunohistology on L. loa adults and mf derived from human volunteers to determine the presence or absence of Wolbachia endosymbionts. Samples used in the PCR analysis included 5 adult female worms, 4 adult male worms, 5 mf samples and 2 samples of L3. The quality and purity of nematode DNA was tested by PCR amplification of nematode 5S rDNA and with diagnostic primers from the target species and used to confirm the absence of contamination from Onchocerca sp., Mansonella perstans, M. streptocerca and Wuchereria bancrofti. Immunohistology was carried out by light and electron microscopy on L. loa adults and mf and sections were probed with rabbit antibodies raised to recombinant Brugia malayi Wolbachia WSP. Samples from nematodes known to be infected with Wolbachia (O. volvulus, O. ochengi, Litomosoides sigmodontis and B. malayi) were used as positive controls and A. viteae as a negative control. RESULTS: Single PCR analysis using primer sets for the bacterial genes 16S rDNA, ftsZ, and WSP were negative for all DNA samples from L. loa. Positive PCR reactions were obtained from DNA samples derived from species known to be infected with Wolbachia, which confirmed the suitability of the primers and PCR conditions. The quality and purity of nematode DNA samples was verified by PCR amplification of 5S rDNA and with nematode diagnostic primers. Additional analysis by 'long PCR' failed to produce any further evidence for Wolbachia symbiosis. Immunohistology of L. loa adults and mf confirmed the results of the PCR with no evidence for Wolbachia symbiosis. CONCLUSION: DNA analysis and immunohistology provided no evidence for Wolbachia symbiosis in L. loa

    Human immune responses to infective stage larval-specific chitinase of filarial parasite, Onchocerca volvulus, Ov-CHI-1.

    BACKGROUND: Ov-CHI-1 is a chitinase specifically expressed in the infective stage larvae of the human filarial parasite Onchocerca volvulus. Evidence has show that it could be a vaccine candidate, however, there is no data available regarding the immunological status of people naturally exposed to infective stage larvae and thus provoked by this antigen. METHOD: We analysed the Ov-CHI-1-specific immune response present in four endemic foci of human onchocerciasis (Ecuador, Nigeria, Togo and Cameroon) by enzyme-linked immunosorbent assays and T-cell proliferation assays. RESULTS: In these foci of infection, antibodies to Ov-CHI-1 were found to be present in only 22% of individuals from Ecuador, but were detected in 42–62% of infected individuals in the three foci from West Africa (Nigeria, Togo and Cameroon). There was found to be no relationship between antibody level and age, gender, or infection intensity as indicated by microfilarial density and numbers of skin nodules. The isotype response to Ov-CHI-1 was dominated by the presence of IgG3, IgG1 was present to a lesser extent. Our results show a positive correlation between N- and C-termini of Ov-CHI-1 in their ability to provoke humoral and cellular immune responses in the human. Peripheral blood mononuclear cell (PBMC) proliferative responses to Ov-CHI-1 when assayed, were found to be significantly higher in the individuals from endemic areas and there was a statistically elevated response to Ov-CHI-1 in the infected individuals when compared to putative immune individuals. CONCLUSION: Ov-CHI-1 is an antigen that we have found strongly induces both humoral and cellular immune responses in humans

    Helium irradiation effects in polycrystalline Si, silica, and single crystal Si

    Transmission electron microscopy (TEM) has been used to investigate the effects of room temperature 6 keV helium ion irradiation of a thin (≈55 nm thick) tri-layer consisting of polycrystalline Si, silica, and single-crystal Si. The ion irradiation was carried out in situ within the TEM under conditions where approximately 24% of the incident ions came to rest in the specimen. This paper reports on the comparative development of irradiation-induced defects (primarily helium bubbles) in the polycrystalline Si and single-crystal Si under ion irradiation and provides direct measurement of a radiation-induced increase in the width of the polycrystalline layer and shrinkage of the silica layer. Analysis using TEM and electron energy-loss spectroscopy has led to the hypothesis that these result from helium-bubble-induced swelling of the silicon and radiation-induced viscoelastic flow processes in the silica under the influence of stresses applied by the swollen Si layers. The silicon and silica layers are sputtered as a result of the helium ion irradiation; however, this is estimated to be a relatively minor effect with swelling and stress-related viscoelastic flow being the dominant mechanisms of dimensional change

    Residual disorder and diffusion in thin Heusler alloy films

    Co2FeSi/GaAs(110) and Co2FeSi/GaAs(111)B hybrid structures were grown by molecular-beam epitaxy and characterized by transmission electron microscopy (TEM) and X-ray diffraction. The films contained inhomogeneous distributions of ordered L2_1 and B2 phases. The average stoichiometry was controlled by lattice parameter measurements, however diffusion processes lead to inhomogeneities of the atomic concentrations and the degradation of the interface, influencing long-range order. An average long-range order of 30-60% was measured by grazing-incidence X-ray diffraction, i.e. the as-grown Co2FeSi films were highly but not fully ordered. Lateral inhomogeneities of the spatial distribution of long-range order in Co2FeSi were found using dark-field TEM images taken with superlattice reflections
