    Polylactic acid scaffolds obtained by 3D printing and modified by oxygen plasma

    The purpose of tissue engineering is to repair, replace, and regenerate tissues and organs. For this aim, materials supports, as polylactic acid (PLA) are used. PLA is a thermoplastic polymer that presents biodegradability, biocompatibility and good processability. PLA scaffolds can accurately constructed by 3D printing. Then, the objectives of this work were to modify the hydrophobic surface of PLA scaffolds using oxygen plasma and to study the cell viability and proliferation. The characterization was done by AFM, contact angle, FTIR and studies of proliferation and cell viability. Results showed that the material acquired hydrophilic properties by the presence of oxygen reactive species and by contact angle decrease. It was also observed an increase in the surface roughness. We can conclude that although the surface modifications were effective and the PLA scaffolds were not cytotoxic, there were no improvements in the proliferation process with the studied osteo-1 lineage cells.

    Synthesis and biocompatibility of an experimental glass ionomer cement prepared by a non-hydrolytic sol-gel method

    The aims of this study were to demonstrate the synthesis of an experimental glass ionomer cement (GIC) by the non-hydrolytic sol-gel method and to evaluate its biocompatibility in comparison to a conventional glass ionomer cement (Vidrion R). Four polyethylene tubes containing the tested cements were implanted in the dorsal region of 15 rats, as follows: GI - experimental GIC and GII - conventional GIC. The external tube walls was considered the control group (CG). The rats were sacrificed 7, 21 and 42 days after implant placement for histopathological analysis. A four-point (I-IV) scoring system was used to graduate the inflammatory reaction. Regarding the experimental GIC sintherization, thermogravimetric and x-ray diffraction analysis demonstrated vitreous material formation at 110oC by the sol-gel method. For biocompatibility test, results showed a moderate chronic inflammatory reaction for GI (III), severe for GII (IV) and mild for CG (II) at 7 days. After 21 days, GI presented a mild reaction (II); GII, moderate (III) and CG, mild (II). At 42 days, GI showed a mild/absent inflammatory reaction (II to I), similar to GII (II to I). CG presented absence of chronic inflammatory reaction (I). It was concluded that the experimental GIC presented mild/absent tissue reaction after 42 days, being biocompatible when tested in the connective tissue of rats.O objetivo deste estudo foi demonstrar a sinterização pelo método sol-gel não-hidrolítico de um cimento de ionômero de vidro experimental (CIV) e avaliar sua biocompatibilidade em relação a um cimento de ionômero de vidro convencional (Vidrion R). Quatro tubos de polietileno contendo os cimentos testados foram implantados no dorso de 15 ratos, da seguinte maneira: GI - CIV Experimental e GII - CIV Convencional. A lateral do tubo foi considerada Grupo Controle. Os ratos foram sacrificados em 7, 21 e 42 dias pós-implantação para análise histopatológica. Uma escala de I a IV foi utilizada como sistema de score para graduar a reação inflamatória. Em relação à sinterização do CIV experimental, as análises termogravométrica e por difração de raio-x demonstraram a formação de material vítreo aos 110oC pelo método sol-gel. Para o teste de biocompatibilidade, os resultados mostraram uma reação inflamatória moderada para o GI (III), severa para o GII (IV) e branda para o Grupo Controle (II) aos 7 dias. Após 21 dias, GI apresentou uma reação branda (II); GII, moderada (III) e Grupo Controle, branda (II). Aos 42 dias, GI apresentou uma reação inflamatória branda/ausente (II a I), similar ao GII (II a I). O Grupo Controle demonstrou ausência de reação inflamatória (I). Concluiu-se que o CIV Experimental apresentou reação tecidual branda/ausente após 42 dias, sendo biocompatível quando testado em tecido conjuntivo de ratos

    Primeiro isolamento no Brasil de herpesvirus eqüino tipo 1 em um cavalo com doença neurológica

    O presente relato refere-se ao primeiro isolamento no Brasil do herpesvírus eqüino tipo 1 (HVE-1) proveniente de um caso clínico de mieloncefalopatia herpética em uma égua, que evoluiu para o óbito. O isolado do HVE-1, denominado 07/05, foi obtido a partir de uma amostra de líquor na primeira passagem em células Vero, confirmando-se sua identidade pela PCR e pela prova de neutralização viral. Como o isolamento viral a partir do líquor geralmente não é bem sucedido, conforme demonstrado em vários relatos de casos, o presente achado sugere que a doença neurológica foi causada por uma amostra particularmente neurovirulenta de HVE-1This report describes the first Brazilian equine herpesvirus type 1 (EHV-1) isolation from a single fatal equine herpes myeloencephalopathy case in a mare. The isolation of EHV-1 was confirmed from the first passage of cerebrospinal fluid (CSF) sample in Vero cells by PCR and virus neutralization assay. As virus isolation from CSF is unlikely to be successful, as has been shown in several case reports, this circumstantial evidence suggests that the neurological disease was caused by particularly neurovirulent strain of EHV-1FAPES

    Ocorrência de enfermidades virais em asininos (Equus asinus) no estado de São Paulo, Brasil

    Among the diseases that affect equines, viral diseases play an important role from a health and economic point of view, especially influenza, viral arteritis, herpes infections and vesicular stomatitis. In the Brazilian literature, there is little or no account of the occurrence of infectious diseases in donkeys. Given the importance of donkeys in different activities and the lack of information on infections that may occur in these animals, the aim of this study was to determine the frequency of anti-equine herpesvirus (EHV), anti-equine arteritis virus (EAV), anti-vesicular stomatitis, and anti-equine influenza (H3N8) antibodies in the serum of 85 donkeys bred in some regions of the state of Sao Paulo. We found the following antibody frequencies: 50.6% (43/85) antibodies against influenza virus subtype H3N8, 47% (40/85) anti-EHV, and 20% (17/85) anti-EAV. The donkeys were not seropositive for vesicular stomatitis. The results suggested that the agents EHV, EAV, and equine influenza subtype H3N8 circulate among donkeys in some regions of the state of Sao Paulo, Brazil, reinforcing the importance of establishing a routine diagnosis and epidemiological study of this species.Dentre as doenças que acometem os equídeos, as enfermidades virais assumem um papel importante do ponto de vista sanitário e econômico, especialmente a influenza, arterite viral, as infecções herpéticas e a estomatite vesicular. Na literatura nacional, existe pouco ou nenhum relato sobre a ocorrência de enfermidades infecciosas nos asininos. Tendo em vista a importância dos asininos para diferentes atividades e a falta de informações sobre as doenças que acometem esses animais, este trabalho teve como objetivo estudar a frequência de anticorpos anti-EHV, antivírus da arterite equina, anti-estomatite vesicular e anti-influenza equina (H3N8) em 85 soros de jumentos criados no estado de São Paulo. Estimou-se que 50,6% apresentavam anticorpos contra o subtipo H3N8 do vírus da influenza; 47% (40/85) apresentavam anticorpos contra o EHV e 20% apresentavam anticorpos contra o vírus da arterite. Os jumentos não foram soro reagentes contra a estomatite vesicular. Os resultados obtidos sugerem que os agentes EHV, vírus da arterite equina e influenza equina subtipo H3N8, circulam entre os jumentos do estado de São Paulo, caracterizando a importância do estabelecimento de uma rotina diagnostica e estudos epidemiológicos na espécie

    CÂNCER DE COLO DO ÚTERO, GENOTIPAGEM DO PAPILOMA VÍRUS HUMANO (HPV) EM MULHERES QUILOMBOLAS DE UM MUNICÍPIO BRASILEIRO: ACEITABILIDADE DA VACINA

    O câncer de colo uterino é o segundo câncer feminino mais comum no Brasil. O papilomavírus humano (HPV) é causa necessária para seu desenvolvimento. A vacina surge como uma arma contra a infecção por HPV e, consequentemente, contra o câncer. Mas existem mais de 100 genótipos diferentes de HPV, que são divididos em baixo risco e alto risco, de acordo com o potencial oncogênico. Analisam-se os aspectos epidemiológicos e biomoleculares da infecção por HPV na mucosa genital de mulheres qui-lombolas, verificando a associação os achados citológicos e biomoleculares e observando a frequência do papilomavírus nessa população e de seus tipos, para avaliar a aplicabilidade da vacina na população em estudo. A população deste estudo é de 101 mulheres pertencentes à comunidade quilombola de Ju-çatuba, no estado do Maranhão. Foram coletadas amostras de mucosa genital, submetidas à pesquisa para a presença do HPV pela técnica de PCR Nested. As amostras positivas foram sequenciadas para fins de genotipagem viral. Em uma população predominante de mulheres em idade fértil, com parceiro fixo e baixo nível de escolaridade, verificou-se que 10 amostras da cérvice uterina foram positivas paraHPV, sendo 7 positivas para os tipos de alto risco. Mediante a presença dos tipos virais de alta oncoge-nicidade, observa-se a necessidade de implementação de condutas gestoras para redução do risco de aparecimento de câncer do colo do útero.Palavras-chave: Papilomavírus humano. Reação em cadeia da polimerase.CANCER OF THE CERVIX, GENOTYPING OF HUMAN PAPILLOMAVIRUS (HPV) IN QUILOMBOLA WOMEN IN A BRAZILIAN CITY: ACCEPTABILITY OF THE VACCINE.ABSTRACT: Cervical cancer is the second most common female cancer in Brazil. The human papilloma-virus (HPV) is a need cause for its development. The vaccine appears as a weapon against HPV infection and therefore against cancer. But there are more than 100 different HPV genotypes, which are divided into low risk and high risk according to the oncogenic potential. To analyze the epidemiological and biomolecu-lar aspects of HPV infection in the genital mucosa of women Maroons, verifying the association cytological and molecular biological findings and noting the frequency of this population papillomavirus and its types, to evaluate the applicability of the vaccine in the study population. The study population was 101 women belonging to the maroon community of Juçatuba in the state of Maranhão. Genital mucosa samples were collected and submitted to search for the presence of HPV by PCR Nested. The positive samples were sequenced for the purpose of viral genotyping. In a predominant population of women of childbearing age, with a steady partner, and low level of education, it was found that 10 samples of uterine cervix were positive for HPV, with 7 positive for high-risk types. Through the presence of viral types of high tumorige-nicity, there is a need to implement management practices for reducing the risk of developing cancer of the cervix.KEYWORDS: Human Papillomavirus. El virus del papiloma humano (VPH) es la causa necesaria para su desarrollo. La vacuna se presenta como un arma contra la infección por VPH y, por tanto, contra el cáncer. Pero hay más de 100 genotipos diferentes de VPH, que se dividen en bajo riesgo y de alto riesgo de acuerdo con el potencial oncogénico. Analizar los aspectos epidemiológicos y biomoleculares de la infección por el VPH en la mucosa genital de la mujer cimarrones, la verificación de la citología asociación y hallazgos de biología molecular y tomando nota de la frecuen-cia de este virus del papiloma de la población y sus tipos, para evaluar la aplicabilidad de la vacuna en la población de estudio. La populación de estudio fue de 101 mujeres pertenecientes a la comunidad marrón de Juçatuba en el Estado de Maranhão. Se recogieron muestras de mucosa genital presentados para buscar la presencia de VPH por PCR anidada. Las muestras positivas se secuenciaron con el propósito de determinación del genotipo viral . RESULTADOS: En una población predominante de las mujeres en edad fértil, con una pareja estable, y el bajo nivel de la educación, se encontró que 10 muestras fueron positivas para el VPH cervical, 7 siendo positivo para los tipos de alto riesgo. A través de la presencia de tipos virales de alta tumorigenicidad, hay una necesidad de aplicar prácticas de gestión para reducir el riesgo de desarrollar cáncer de la cérvix.PALABRAS CLAVE: Virus del papiloma humano.     Role of age and comorbidities in mortality of patients with infective endocarditis

    [Purpose]: The aim of this study was to analyse the characteristics of patients with IE in three groups of age and to assess the ability of age and the Charlson Comorbidity Index (CCI) to predict mortality. [Methods]: Prospective cohort study of all patients with IE included in the GAMES Spanish database between 2008 and 2015.Patients were stratified into three age groups:<65 years,65 to 80 years,and ≥ 80 years.The area under the receiver-operating characteristic (AUROC) curve was calculated to quantify the diagnostic accuracy of the CCI to predict mortality risk. [Results]: A total of 3120 patients with IE (1327 < 65 years;1291 65-80 years;502 ≥ 80 years) were enrolled.Fever and heart failure were the most common presentations of IE, with no differences among age groups.Patients ≥80 years who underwent surgery were significantly lower compared with other age groups (14.3%,65 years; 20.5%,65-79 years; 31.3%,≥80 years). In-hospital mortality was lower in the <65-year group (20.3%,<65 years;30.1%,65-79 years;34.7%,≥80 years;p < 0.001) as well as 1-year mortality (3.2%, <65 years; 5.5%, 65-80 years;7.6%,≥80 years; p = 0.003).Independent predictors of mortality were age ≥ 80 years (hazard ratio [HR]:2.78;95% confidence interval [CI]:2.32–3.34), CCI ≥ 3 (HR:1.62; 95% CI:1.39–1.88),and non-performed surgery (HR:1.64;95% CI:11.16–1.58).When the three age groups were compared,the AUROC curve for CCI was significantly larger for patients aged <65 years(p < 0.001) for both in-hospital and 1-year mortality. [Conclusion]: There were no differences in the clinical presentation of IE between the groups. Age ≥ 80 years, high comorbidity (measured by CCI),and non-performance of surgery were independent predictors of mortality in patients with IE.CCI could help to identify those patients with IE and surgical indication who present a lower risk of in-hospital and 1-year mortality after surgery, especially in the <65-year group
