Seh1 targets GATOR2 and Nup153 to mitotic chromosomes
In metazoa the Nup107-160 nucleoporin Y complex plays a major role in formation of the nuclear pore complex in interphase and is localised to kinetochores in mitosis. The Nup107-160 complex shares a single highly conserved subunit, Seh1, with the GATOR2 complex, an essential activator of mTORC1 kinase. mTORC1/GATOR2 has a central role in the coordination of cell growth and proliferation. Here we use chemical genetics and quantitative chromosome proteomics to study the role of the Seh1 protein in mitosis. Surprisingly, Seh1 is not required for the association of the Nup107-160 complex with mitotic chromosomes, but it is essential for the association of both GATOR2 complex and nucleoporin Nup153 with mitotic chromosomes. Our analysis also reveals a role for Seh1 at human centromeres, where it is required for efficient localisation of the chromosomal passenger complex (CPC). Furthermore this analysis detects a functional interaction between the Nup107-160 complex and the small kinetochore protein SKAP.</jats:p

Studies of the lamin proteinase reveal multiple parallel biochemical pathways during apoptotic execution
Although specific proteinases play a critical role in the active phase of apoptosis, their substrates are largely unknown. We previously identified poly(ADP-ribose) polymerase (PARP) as an apoptosis-associated substrate for proteinase(s) related to interleukin 1 beta-converting enzyme (ICE). Now we have used a cell-free system to characterize proteinase(s) that cleave the nuclear lamins during apoptosis. Lamin cleavage during apoptosis requires the action of a second ICE-like enyzme, which exhibits kinetics of cleavage and a profile of sensitivity to specific inhibitors that is distinct from the PARP proteinase. Thus, multiple ICE-like enzymes are required for apoptotic events in these cell-free extracts. Inhibition of the lamin proteinase with tosyllysine "chloromethyl ketone" blocks nuclear apoptosis prior to the packaging of condensed chromatin into apoptotic bodies. Under these conditions, the nuclear DNA is fully cleaved to a nucleosomal ladder. Our studies reveal that the lamin proteinase and the fragmentation nuclease function in independent parallel pathways during the final stages of apoptotic execution. Neither pathway alone is sufficient for completion of nuclear apoptosis. Instead, the various activities cooperate to drive the disassembly of the nucleus
Essentially English : Sherlock Holmes at the BBC
Sir Arthur Conan Doyle’s Sherlock Holmes stories, currently enjoying renewed popularity on television via the BBC’s Sherlock (2010- ), have been adapted for the screen countless times around the world. Arguably best remembered are Granada’s long-running strand with Jeremy Brett (1984-94), and the Universal film series of the 1940s, featuring Basil Rathbone and Nigel Bruce. Less frequently cited, however, are the two series produced by the BBC between 1965 and 1968, in which first Douglas Wilmer and later Peter Cushing took on the mantle of the Baker Street detective. Typically for the time – but against the wishes of the Conan Doyle estate - these programmes adopted a multi-camera studio model; a mode of production which made them less attractive to US networks than the single camera 35mm film output of commercial rivals such as ITC. Drawing upon material from the BBC’s Written Archives Centre, this article investigates the motivations underpinning the Corporation’s refusal to accommodate the estate’s exhortations to seek American co-production and utilise single camera filming, not least of which was the BBC’s stated desire to maintain the ‘essentially English’ quality of Sherlock Holmes. This decision would have significant repercussions for the series’ overseas saleability, and – despite impressive viewing figures and positive audience reaction at home in the UK – helped contribute to its ‘forgotten’ status with regard to the television canon
Dynamics of viscous amphiphilic films supported by elastic solid substrates
The dynamics of amphiphilic films deposited on a solid surface is analyzed
for the case when shear oscillations of the solid surface are excited. The two
cases of surface- and bulk shear waves are studied with film exposed to gas or
to a liquid. By solving the corresponding dispersion equation and the wave
equation while maintaining the energy balance we are able to connect the
surface density and the shear viscocity of a fluid amphiphilic overlayer with
experimentally accessible damping coefficients, phase velocity, dissipation

Supersymmetric Electroweak Cosmic Strings
We study the connection between supersymmetry and a topological bound
in a two-Higgs-doublet system with an gauge group. We derive the Bogomol'nyi equations from
supersymmetry considerations showing that they hold provided certain conditions
on the coupling constants, which are a consequence of the huge symmetry of the
theory, are satisfied. Their solutions, which can be interpreted as electroweak
cosmic strings breaking one half of the supersymmetries of the theory, are
studied. Certain interesting limiting cases of our model which have recently
been considered in the literature are finally analyzed.Comment: 20 pages, RevTe
Scattering from Electroweak Strings
The scattering of a charged fermion from an electroweak string is studied.
Owing to an amplification of the wave function at the core radius, the cross
sections for helicity flip processes can be largely enhanced. For (where is the Weinberg angle), and , we show that the helicity flip differential cross section
for electrons is of the order and is independent of angle. We
compare our results with those obtained in calculations of rates for baryon
number violating processes in the core of a cosmic string. In that case, while
the enhancement is a general phenomenon, its actual magnitude is extremely
sensitive to the fractional flux carried by the string core. Apart from showing
the existence of a similar enhancement effect for non-topological strings, our
results indicate that in some models the magnitude of enhancement can be
rendered much less sensitive to the value of the parameters in the theories.
Scattering of particles off semi-local strings and axion strings are also

Cryo-electron microscopy of viruses
Thin vitrified layers of unfixed, unstained and unsupported virus suspensions can be prepared for observation by cryo-electron microscopy in easily controlled conditions. The viral particles appear free from the kind of damage caused by dehydration, freezing or adsorption to a support that is encountered in preparing biological samples for conventional electron microscopy. Cryo-electron microscopy of vitrified specimens offers possibilities for high resolution observations that compare favourably with any other electron microscopical method
The PreVOCA experiment: modeling the lower troposphere in the Southeast Pacific
The Preliminary VOCALS Model Assessment (PreVOCA) aims to assess contemporary atmospheric modeling of the subtropical South East Pacific, with a particular focus on the clouds and the marine boundary layer (MBL). Models results from fourteen modeling centers were collected including operational forecast models, regional models, and global climate models for the month of October 2006. Forecast models and global climate models produced daily forecasts, while most regional models were run continuously during the study period, initialized and forced at the boundaries with global model analyses. Results are compared in the region from 40&deg; S to the equator and from 110&deg; W to 70&deg; W, corresponding to the Pacific coast of South America. Mean-monthly model surface winds agree well with QuikSCAT observed winds and models agree fairly well on mean weak large-scale subsidence in the region next to the coast. However they have greatly differing geographic patterns of mean cloud fraction with only a few models agreeing well with MODIS observations. Most models also underestimate the MBL depth by several hundred meters in the eastern part of the study region. The diurnal cycle of liquid water path is underestimated by most models at the 85&deg; W 20&deg; S stratus buoy site compared with satellite, consistent with previous modeling studies. The low cloud fraction is also underestimated during all parts of the diurnal cycle compared to surface-based climatologies. Most models qualitatively capture the MBL deepening around 15 October 2006 at the stratus buoy, associated with colder air at 700 hPa
