    Evidence of instability in previously-mapped landslides as measured using GPS, optical, and SAR data between 2007 and 2017: A case study in the Portuguese Bend Landslide Complex, California

    Velocity dictates the destructive potential of a landslide. A combination of synthetic aperture radar (SAR), optical, and GPS data were used to maximize spatial and temporal coverage to monitor continuously-moving portions of the Portuguese Bend landslide complex on the Palos Verdes Peninsula in Southern California. Forty SAR images from the COSMO-SkyMed satellite, acquired between 19 July 2012 and 27 September 2014, were processed using Persistent Scatterer Interferometry (PSI). Eight optical images from the WorldView-2 satellite, acquired between 20 February 2011 and 16 February 2016, were processed using the Co-registration of Optically Sensed Images and Correlation (COSI-Corr) technique. Displacement measurements were taken at GPS monuments between September 2007 and May 2017. Incremental and average deformations across the landslide complex were measured using all three techniques. Velocity measured within the landslide complex ranges from slow (\u3e 1.6 m/year) to extremely slow (\u3c 16 mm/year). COSI-Corr and GPS provide detailed coverage of m/year-scale deformation while PSI can measure extremely slow deformation rates (mm/year-scale), which COSI-Corr and GPS cannot do reliably. This case study demonstrates the applicability of SAR, optical, and GPS data synthesis as a complimentary approach to repeat field monitoring and mapping to changes in landslide activity through time

    Building the Foundation for a Necessary Debate: Projectification of Society

    Projectification is the phenomenon whereby activities traditionally carried out in a functional manner are approached as projects. It also includes the transformation processes of organisations as project management and non-functional structures. It is a phenomenon that has become important in recent years. It has brought great benefits to organisations and public administration, and it has optimised the use of economic resources. On the other hand, projectification also brings undesirable effects, known as the dark side of projectification. Several years after the first time projectification was coined, a deep debate about projectification has been necessary to make the most of all possible levels. This research, through a bibliometric analysis and a review of the most outstanding literature, identifies those aspects that need to be discussed and where there is room for improvement. The results, with an important set of disadvantages of projectification, sometimes not taken into account, especially at the individual level, establish a solid basis for the debate on projectification and the possible points of improvement from all perspectives (individual, organisational and societal). These perspectives should be observed as different but complementary, forming a holistic understanding of projectification

    Effects of High-Mobility Group A Protein Application on Canine Adipose-Derived Mesenchymal Stem Cells In Vitro

    Multipotency and self-renewal are considered as most important features of stem cells to persist throughout life in tissues. In this context, the role of HMGA proteins to influence proliferation of adipose-derived mesenchymal stem cell (ASCs) while maintaining their multipotent and self-renewal capacities has not yet been investigated. Therefore, extracellular HMGA1 and HMGA2 application alone (10–200 ng/mL) and in combination with each other (100, 200 ng/mL each) was investigated with regard to proliferative effects on canine ASCs (cASCs) after 48 hours of cultivation. Furthermore, mRNA expression of multipotency marker genes in unstimulated and HMGA2-stimulated cASCs (50, 100 ng/mL) was analyzed by RT-qPCR. HMGA1 significantly reduced cASCs proliferation in concentrations of 10–200 ng/mL culture medium. A combination of HMGA1 and HMGA2 protein (100 and 200 ng/mL each) caused the same effects, whereas no significant effect on cASCs proliferation was shown after HMGA2 protein application alone. RT-qPCR results showed that expression levels of marker genes including KLF4, SOX2, OCT4, HMGA2, and cMYC mRNAs were on the same level in both HMGA2-protein-stimulated and -unstimulated cASCs. Extracellular HMGA protein application might be valuable to control proliferation of cASCs in context with their employment in regenerative approaches without affecting their self-renewal and multipotency abilities

    Plk1 overexpression induces chromosomal instability and suppresses tumor development

    Polo-like kinase 1 (Plk1) is overexpressed in a wide spectrum of human tumors, being frequently considered as an oncogene and an attractive cancer target. However, its contribution to tumor development is unclear. Using a new inducible knock-in mouse model we report here that Plk1 overexpression results in abnormal chromosome segregation and cytokinesis, generating polyploid cells with reduced proliferative potential. Mechanistically, these cytokinesis defects correlate with defective loading of Cep55 and ESCRT complexes to the abscission bridge, in a Plk1 kinase-dependent manner. In vivo, Plk1 overexpression prevents the development of Kras-induced and Her2-induced mammary gland tumors, in the presence of increased rates of chromosome instability. In patients, Plk1 overexpression correlates with improved survival in specific breast cancer subtypes. Therefore, despite the therapeutic benefits of inhibiting Plk1 due to its essential role in tumor cell cycles, Plk1 overexpression has tumor-suppressive properties by perturbing mitotic progression and cytokinesis.We are indebted to Stephen Taylor for the Sgo1 antibody.     Mesenchymal Stem Cells and Extracellular Vesicles: An Emerging Alternative to Combat COVID-19

    The global SARS-CoV-2 outbreak has been accompanied with severe socio-economic and health burdens that will ripple through history. It is now known that SARS-CoV-2 induces a cytokine storm that leads to acute respiratory distress syndrome and systemic organ damage. With no definitive nor safe therapy for COVID-19 as well as the rise of viral variants the need for an urgent treatment modality is paramount. Mesenchymal stem cells (MSCs) and their extracellular vesicles (EVs) have long been praised for their anti-viral, anti-inflammatory and tissue regenerative capabilities. MSCs and their EVs are now being studied for their possible use as a treatment modality for COVID-19. In this review we explore their capabilities and outline the evidence of their use in ALI, ARDS and COVID-19

    Plastid phylogenomics resolves ambiguous relationships within the orchid family and provides a solid timeframe for biogeography and macroevolution

    Recent phylogenomic analyses based on the maternally inherited plastid organelle have enlightened evolutionary relationships between the subfamilies of Orchidaceae and most of the tribes. However, uncertainty remains within several subtribes and genera for which phylogenetic relationships have not ever been tested in a phylogenomic context. To address these knowledge-gaps, we here provide the most extensively sampled analysis of the orchid family to date, based on 78 plastid coding genes representing 264 species, 117 genera, 18 tribes and 28 subtribes. Divergence times are also provided as inferred from strict and relaxed molecular clocks and birth–death tree models. Our taxon sampling includes 51 newly sequenced plastid genomes produced by a genome skimming approach. We focus our sampling efforts on previously unplaced clades within tribes Cymbidieae and Epidendreae. Our results confirmed phylogenetic relationships in Orchidaceae as recovered in previous studies, most of which were recovered with maximum support (209 of the 262 tree branches). We provide for the first time a clear phylogenetic placement for Codonorchideae within subfamily Orchidoideae, and Podochilieae and Collabieae within subfamily Epidendroideae. We also identify relationships that have been persistently problematic across multiple studies, regardless of the different details of sampling and genomic datasets used for phylogenetic reconstructions.
