    Effect of salinity on the biosynthesis of n-3 long-chain polyunsaturated fatty acids in silverside Chirostoma estor

    The genus Chirostoma (silversides) belongs to the family Atherinopsidae, which contains around 150 species, most of which are marine. However, Mexican silverside (Chirostoma estor) is one of the few representatives of freshwater atherinopsids and is only found in some lakes of the Mexican Central Plateau. However, studies have shown that C. estor has improved survival, growth and development when cultured in water conditions with increased salinity. In addition, C. estor displays an unusual fatty acid composition for a freshwater fish with high docosahexaenoic acid (DHA) : eicosapentaenoic acid (EPA) ratios. Freshwater and marine fish species display very different essential fatty acid metabolism and requirements and so the present study investigated long-chain polyunsaturated fatty acid (LC-PUFA) biosynthesis to determine the capacity of C. estor for endogenous production of EPA and DHA, and the effect that salinity has on these pathways. Briefly, C. estor were maintained at three salinities (0, 5 and 15 ppt) and the metabolism of 14C-labelled 18:3n-3 determined in isolated hepatocyte and enterocyte cells. The results showed that C. estor has the capacity for endogenous biosynthesis of LC-PUFA from 18-carbon fatty acid precursors, but that the pathway was essentially only active in saline conditions with virtually no activity in cells isolated from fish grown in freshwater. The activity of the LCPUFA biosynthesis pathway was also higher in cells isolated from fish at 15 ppt compared to fish at 5 ppt, The pathway was around 5-fold higher in hepatocytes compared to enterocytes, although the majority of 18:3n-3 was converted to 18:4n-3 and 20:4n-3 in hepatocytes whereas the proportions of 18:3n-3 converted to EPA and DHA were higher in enterocytes. The data were consistent with the hypothesis that conversion of EPA to DHA could contribute, at least in part, to the generally high DHA:EPA ratios observed in the tissue lipids of C. estor

    Radiocarbon evidence for the mining of organic nitrogen from soil by mycorrhizal fungi

    Organic nitrogen use by mycorrhizal fungi and associated plants could fuel productivity in nitrogen-limited systems. To test whether fungi assimilated soil-derived organic nitrogen, we compared the 14C signal (expressed as Δ14C) from 1950s to 1960s thermonuclear testing in protein and structural carbon of ectomycorrhizal fungi. As expected, structural carbon had Δ14C similar to recent photosynthesis; however, protein Δ14C was either higher or lower than structural carbon depending on the fungal taxa. This suggests that some protein carbon derived from uptake of organic nitrogen with different Δ14C signals. Specimens from two taxa (Lactarius and Russula) adapted to taking up soluble nutrients had protein higher than structural carbon in Δ14C, indicating uptake of young, post-bomb organic nitrogen, whereas two taxa (Cortinarius and Leccinum) adapted to using insoluble, complex organic nutrients had protein lower than structural carbon in Δ14C, indicating uptake of old, pre-bomb organic nitrogen. Tuber, a genus common in mineral soil, was also consistently lower in Δ14C for protein than for structural carbon, with an estimated 10 % of protein carbon originating from old, deep organic nitrogen for this taxon. Our results indicate that radiocarbon can provide evidence of organic nitrogen use in ectomycorrhizal fungi and reflects the exploration depth of different taxa

    Nutrition in relation to organic aquaculture: Sources and strategies

    Organic production is a system of farm management and food production that combines best environmental practices, a high level of biodiversity, the preservation of natural resources, the application of high animal welfare standards and a production method in line with the preference of certain consumers for products produced using natural substances and processes. Mie et al. (2017) reviewed existing evidence on the impact of organic food on human health and compared organic versus conventional food production with respect to parameters important to human health. The review emphasised several documented human health benefits associated with organic food production and production methods and concluded that it is likely to be beneficial within the conventional agriculture, for example, in integrated pest management and antibiotics. This chapter covers aspects of current use of formulated feeds, feed composition, aquafeed technology, sustainable alternatives to common feed ingredients, nutritional physiology and general nutritional principles and product quality in the context of the organic aquaculture. It reviews new knowledge and presents research results to update and may modify the criteria and standards for organic aquaculture in relation to nutrition and thus to provide high-quality products for the consumers. This chapter is based on the current European regulation on organic aquaculture, as well as on the proposed revision of the European regulation, which is currently being approved after a long process for getting the agreement of the European Parliament, European Council and the European Commission
