    An assessment of the mantle and slab components in the magmas of an oceanic arc volcano: Raoul Volcano, Kermadec arc

    Raoul Volcano occupies a simple oceanic subduction setting in the northern part of the Kermadec arc on the Pacific–Australian convergent plate boundary. The primary inputs to the magmatic system that feeds the volcano are a subduction component derived from the subducting old Pacific oceanic lithosphere and its veneer of pelagic sediment, and the overlying peridotitic mantle wedge. Conservative trace elements that are very incompatible during mantle melting are relatively depleted in Raoul lavas indicating a source that has been depleted during an earlier melting event. Major element co-variations indicate magma genesis by 25% near fractional melting of a mantle source that is weakly depleted (2% melt extraction) relative to a fertile MORB source. An important influence on the composition of the mantle component is progressive melt extraction coupled with minimal advection of fresh material into the sub-arc zone followed by melt extraction from a melting column beneath the spreading centre of an adjacent back arc basin. High field strength element and rare earth element systematics indicate involvement of a subduction-related component of constant composition. Two fluid components can be distinguished, one enriched in large ion lithophile elements inferred to be an aqueous fluid that is continuously added to the ascending melt column and the other a less mobile fluid that transfers Th. A homogeneous subduction-related component of constant composition and magnitude arises if the slab-derived flux migrates from the slab–mantle interface to the sub-arc melting column by repeated episodes of amphibole formation and decomposition its composition is then governed by the distribution coefficients of pyroxene and its magnitude by the degree of amphibole saturation of mantle peridotite. The results from Raoul Volcano are comparable to those from other oceanic subduction-related arcs such as South Sandwich and Marianas suggesting that this is a general model for oceanic arcs

    Solving the Hamilton-Jacobi Equation for General Relativity

    We demonstrate a systematic method for solving the Hamilton-Jacobi equation for general relativity with the inclusion of matter fields. The generating functional is expanded in a series of spatial gradients. Each term is manifestly invariant under reparameterizations of the spatial coordinates (``gauge-invariant''). At each order we solve the Hamiltonian constraint using a conformal transformation of the 3-metric as well as a line integral in superspace. This gives a recursion relation for the generating functional which then may be solved to arbitrary order simply by functionally differentiating previous orders. At fourth order in spatial gradients, we demonstrate solutions for irrotational dust as well as for a scalar field. We explicitly evolve the 3-metric to the same order.     A within farm clinical trial to compare two treatments (parenteral antibacterials and hoof trimming) for sheep lame with footrot

    From observational studies, farmers who use parenteral antibacterials to promptly treat all sheep with footrot (FR) or interdigital dermatitis (ID) have a prevalence of lameness of <2% compared with a prevalence of 9% lameness reported by farmers who treat lame sheep by trimming affected feet. We tested the hypothesis that prompt treatment of sheep lame with naturally developing FR or ID with parenteral and topical antibacterials reduces the prevalence and incidence of lameness with these conditions compared with less frequent treatment with trimming of hoof horn and applying topical antibacterials. A further hypothesis was that reduction of ID and FR would improve productivity. A lowland sheep flock with 700 ewes was used to test these hypotheses in an 18-month within farm clinical trial with four groups of ewes: two intervention and two control. The duration and severity of lameness was used to categorise sheep into three weighted scores of lameness (WLS): never lame (WLS0), mildly lame/lame for <6 days (WLS1) and severely or chronically lame (WLS2). The intervention reduced the prevalence of lameness due to FR and ID in ewes and lambs and the incidence of lameness in ewes. The WLS was also significantly lower in sheep in the intervention groups. Ewes with a higher WLS were subsequently significantly more likely to have a body condition score <2.5 and to have lame lambs. Significantly more ewes lambed and successfully reared more lambs that were ready for slaughter at a younger age in the intervention versus control groups. There was an increase in the gross margin of ÂŁ630/100 ewes mated in the intervention group, including the cost of treatment of ÂŁ150/100 ewes mated. We conclude that prompt parenteral and topical antibacterial treatment of sheep lame with ID and FR reduced the prevalence and incidence of these infectious conditions and led to improved health, welfare and productivity

    Haploinsufficiency of human APOE reduces amyloid deposition in a mouse model of amyloid-ÎČ amyloidosis

    Apolipoprotein E Δ4 (APOE Δ4) is the strongest genetic risk factor for Alzheimer’s disease (AD). Evidence suggests that the effect of apoE isoforms on amyloid-ÎČ (AÎČ) accumulation in the brain plays a critical role in AD pathogenesis. Like in humans, apoE4 expression in animal models that develop AÎČ-amyloidosis results in greater AÎČ and amyloid deposition than with apoE3 expression. However, whether decreasing levels of apoE3 or apoE4 would promote or attenuate AÎČ-related pathology has not been directly addressed. To determine the effect of decreasing human apoE levels on AÎČ accumulation in vivo, we generated human APOE isoform haploinsufficient mouse models by crossing APPPS1-21 mice with APOE isoform knock-in mice. By genetically manipulating APOE gene dosage, we demonstrate that decreasing human apoE levels, regardless of isoform status, results in significantly decreased amyloid plaque deposition and microglial activation. This differences in amyloid load between apoE3 and apoE4 expressing mice were not due to apoE4 protein being present at lower levels than apoE3. These data suggest that current therapeutic strategies to increase apoE levels without altering its lipidation state may actually worsen AÎČ amyloidosis, while increasing apoE degradation or inhibiting its synthesis may be a more effective treatment approach

    Time to improve informed consent for dialysis: an international perspective

    The literature reveals that current nephrology practice in obtaining informed consent for dialysis falls short of ethical and legal requirements. Meeting these requirements represents a significant challenge, especially because the benefits and risks of dialysis have shifted significantly with the growing number of older, comorbid patients. The importance of informed consent for dialysis is heightened by several concerns, including: (1) the proportion of predialysis patients and patients on dialysis who lack capacity in decision making and (2) whether older, comorbid, and frail patients understand their poor prognosis and the full implications to their independence and functional status of being on dialysis. This article outlines the ethical and legal requirements for a valid informed consent to dialysis: (1) the patient was competent, (2) the consent was made voluntarily, and (3) the patient was given sufficient information in an understandable manner to make the decision. It then considers the application of these requirements to practice across different countries. In the process of informed consent, the law requires a discussion by the physician of the material risks associated with dialysis and alternative options. We argue that, legally and ethically, this discussion should include both the anticipated trajectory of the illness and the effect on the life of the patient with particular regard to the outcomes most important to the individual. In addition, a discussion should occur about the option of a conservative, nondialysis pathway. These requirements ensure that the ethical principle of respect for patient autonomy is honored in the context of dialysis. Nephrologists need to be open to, comfortable with, and skillful in communicating this information. From these clear, open, ethically, and legally valid consent discussions, a significant dividend will hopefully flow for patients, families, and nephrologists alike

    Light-Front Approach for Heavy Pentaquark Transitions

    Assuming the two diquark structure for the pentaquark state as advocated in the Jaffe-Wilczek model, there exist exotic parity-even anti-sextet and parity-odd triplet heavy pentaquark baryons. The theoretical estimate of charmed and bottom pentaquark masses is quite controversial and it is not clear whether the ground-state heavy pentaquark lies above or below the strong-decay threshold. We study the weak transitions of heavy pentaquark states using the light-front quark model. In the heavy quark limit, heavy-to-heavy pentaquark transition form factors can be expressed in terms of three Isgur-Wise functions: two of them are found to be normalized to unity at zero recoil, while the third one is equal to 1/2 at the maximum momentum transfer, in accordance with the prediction of the large-Nc approach or the quark model. Therefore, the light-front model calculations are consistent with the requirement of heavy quark symmetry. Numerical results for form factors and Isgur-Wise functions are presented. Decay rates of the weak decays Theta_b+ to Theta_c0 pi+ (rho+), Theta_c0 to Theta+ pi- (rho-), Sigma'_{5b}+ to Sigma'_{5c}0 pi+ (rho+) and Sigma'_{5c}0 to N_8+ pi- (rho-) with Theta_Q, Sigma'_{5Q} and N_8 being the heavy anti-sextet, heavy triplet and light octet pentaquarks, respectively, are obtained. For weakly decaying Theta_b+ and Theta_c0, the branching ratios of Theta_b+ to Theta_c0 pi+, Theta_c0 to Theta+ pi- are estimated to be at the level of 10^{-3} and a few percents, respectively.Comment: 33 pages, 3 figures, version to be published in Phys.     Template-stripped gold surfaces with 0.4 nm rms roughness suitable for force measurements. Application to the Casimir force in the 20-100 nm range

    Using a template-stripping method, macroscopic gold surfaces with root-mean-square (rms) roughness less than 0.4 nm have been prepared, making them useful for studies of surface interactions in the nanometer range. The utility of such substrates is demonstrated by measurements of the Casimir force at surface separations between 20 and 100 nm, resulting in good agreement with theory. The significance and quantification of this agreement is addressed, as well as some methodological aspects regarding the measurement of the Casimir force with high accuracy.Comment: 7 figure

    Methods for suspension culture, protoplast extraction, and transformation of high-biomass yielding perennial grass Arundo donax

    Arundo donax L. is a promising biofuel feedstock in the Mediterranean region. Despite considerable interest in its genetic improvement, Arundo tissue culture and transformation remains arduous. The authors developed methodologies for cell- and tissue culture and genetic engineering in Arundo. A media screen was conducted, and a suspension culture was established using callus induced from stem axillary bud explants. DBAP medium, containing 9 \ub5M 2,4-D and 4.4 \ub5M BAP, was found to be the most effective medium among those tested for inducing cell suspension cultures, which resulted in a five-fold increase in tissue mass over 14 days. In contrast, CIM medium containing 13 \ub5M 2,4-D, resulted in just a 1.4-fold increase in mass over the same period. Optimized suspension cultures were superior to previously-described solidified medium-based callus culture methods for tissue mass increase. Suspension cultures proved to be very effective for subsequent protoplast isolation. Protoplast electroporation resulted in a 3.3 \ub1 1.5% transformation efficiency. A dual fluorescent reporter gene vector enabled the direct comparison of the CAMV 35S promoter with the switchgrass ubi2 promoter in single cells of Arundo. The switchgrass ubi2 promoter resulted in noticeably higher reporter gene expression compared with that conferred by the 35S promoter in Arundo

    Restoring habitat for fire-impacted species' across degraded Australian landscapes

    In the summer of 2019-2020, southern Australia experienced the largest fires on record, detrimentally impacting the habitat of native species, many of which were already threatened by past and current anthropogenic land use. A large-scale restoration effort to improve degraded species habitat would provide fire-affected species with the chance to recover and persist in burnt and unburnt habitat. To facilitate this, decision-makers require information on priority species needs for restoration intervention, the suite of potential restoration interventions, and the priority locations for applying these interventions. We prioritize actions in areas where restoration would most likely provide cost-effective benefits to priority species (defined by each species proportion of habitat burned, threat status, and vulnerability to fires), by integrating current and future species habitat suitability maps with spatially modelled costs of restoration interventions such as replanting, removing invasive species, and implementing ecologically appropriate fire management. We show that restoring the top similar to 69% (112 million hectares) of the study region (current and future distributions of priority species) accounts for, on average, 95% of current and future habitat for every priority species and costs similar to AUD73billionyr(−1)(AUD73 billion yr(-1) (AUD650 hectare(-1) yr(-1)) annualized over 30 years. This effort would include restoration actions over 6 million hectares of fire-impacted habitat, costing similar to AUD8.8billion/year.Largescalerestorationeffortsareoftencostlybutcanhavesignificantsocietalco−benefitsbeyondbiodiversityconservation.Wealsoshowthatupto291MtCO2(similarto150MtDM)ofcarboncouldbesequesteredbyrestorationefforts,resultinginapproximatelyAUD8.8 billion/year. Large scale restoration efforts are often costly but can have significant societal co-benefits beyond biodiversity conservation. We also show that up to 291 MtCO2 (similar to 150 Mt DM) of carbon could be sequestered by restoration efforts, resulting in approximately AUD253 million yr(-1) in carbon market revenue if all carbon was remunerated. Our approach highlights the scale, costs, and benefits of targeted restoration activities both inside and outside of the immediate bushfire footprint over vast areas of different land tenures

    Gauge-ready formulation of the cosmological kinetic theory in generalized gravity theories

    We present cosmological perturbations of kinetic components based on relativistic Boltzmann equations in the context of generalized gravity theories. Our general theory considers an arbitrary number of scalar fields generally coupled with the gravity, an arbitrary number of mutually interacting hydrodynamic fluids, and components described by the relativistic Boltzmann equations like massive/massless collisionless particles and the photon with the accompanying polarizations. We also include direct interactions among fluids and fields. The background FLRW model includes the general spatial curvature and the cosmological constant. We consider three different types of perturbations, and all the scalar-type perturbation equations are arranged in a gauge-ready form so that one can implement easily the convenient gauge conditions depending on the situation. In the numerical calculation of the Boltzmann equations we have implemented four different gauge conditions in a gauge-ready manner where two of them are new.
