    Sähköpotkulaudoilla tuotettu liike kestävässä kaupunkitilassa

    Tiivistelmä. Ihmisen tarve liikkua on aina ollut keskeisessä osassa elämää. Suuret liikevirrat kohdistuvat ennen kaikkea kohti kaupunkeja. Kiihtynyt urbanisaatio ja yhä keskeisemmässä osassa olevat kaupunkitilat ovat monitilaisuutensa ja jatkuvan muovautuvuutensa kautta erityisen ajankohtaisia tutkimusympäristöjä. Vaikka liike kohdistuukin kohti kaupunkeja, myös kaupunkien sisällä on liikettä. Moderni liike ja liikenne ovat asioita, joita ilman emme voisi edes kuvitella elämää. Paljon mahdollistavalla liikenteellä on kuitenkin myös omat huonot puolensa. Liikenteen ja liikkeen suorat ja epäsuorat vaikutukset heikentävät kaupunkitilojen hyvinvointia. Kaupunkien heikentynyt tila on huomattu ja kaupunkitilojen uudelleen inhimillistämisen näkökulmasta on alettu tehdä erilaisia suunnitelmia. UNESCO:n laatimien Agenda 2030 -tavoitteiden kohdan 11 tarkoituksena on taata kaikille oikeus hyviin sekä kestäviin kaupunkeihin ja asuinympäristöihin. Kaupunkimme pitäminen elinvoimaisina ja inhimillisinä tiloina vaatii meiltä jo olemassa olevien liikennemuotojen tarkastelua. Myös kestävämpien liikkumistapojen innovointi on välttämätöntä. Vuokraus- ja jakamispalveluverkostoissa olevat sähköpotkulautapalvelut ovat vakiintuneet nopeasti osaksi kaupunkiemme liikennejärjestelmiä. Uuden sukupolven sähköpotkulautaverkostot ovat tulleet vastaamaan niin lyhyiden alku- kuin loppumatkojenkin kysynnäntarpeeseen. Sähköpotkulautaverkostot muuttavat ja ovat muuttaneet kaupunkitiloissa tapahtuvaa liikettä. Laitteilla tuotetun liikkeen ristiriitaisuus muun kaupunkitilan liikkeen kanssa voi heikentää kaupunkiemme tilaa. Tämä vaikuttaa mahdollisuuksiimme saavuttaa kestävän kaupunkikehityksen tavoitteet sekä uudet inhimillisemmät kaupunkitilat. Tutkielmassani tarkastelen sitä, kuinka hyvin sähköpotkulaudat ovat löytäneet paikkansa osana kaupunkitilojen liikettä, ja onko liike ristiriidassa muun kaupunkitilojen liikkeen kanssa

    Incidence and management of patients with colorectal cancer and synchronous and metachronous colorectal metastases : a population-based study

    Background This population-based study aimed to examine the incidence, patterns and results of multimodal management of metastatic colorectal cancer. Methods A retrospective population-based study was conducted on patients with metastatic colorectal cancer in Central Finland in 2000-2015. Clinical and histopathological data were retrieved and descriptive analysis was conducted to determine the pattern of metastatic disease, defined as synchronous, early metachronous (within 12 months of diagnosis of primary disease) and late metachronous (more than 12 months after diagnosis). Subgroups were compared for resection and overall survival (OS) rates. Results Of 1671 patients, 296 (17.7 per cent) had synchronous metastases, and 255 (19.6 per cent) of 1302 patients with resected stage I-III tumours developed metachronous metastases (94 early and 161 late metastases). Liver, pulmonary and intraperitoneal metastases were the most common sites. The commonest metastatic patterns were a combination of liver and lung metastases. The overall metastasectomy rate for patients with synchronous metastases was 16.2 per cent; in this subgroup, 3- and 5-year OS rates after any resection were 63 and 44 per cent respectively, compared with 7.1 and 3.3 per cent following no resection (P <0.001). The resection rate was higher for late than for early metachronous disease (28.0versus17 per cent respectively;P = 0.048). Three- and 5-year OS rates after any resection of metachronous metastases were 78 and 62 per cent respectivelyversus42.1 and 18.2 per cent with no metastasectomy (P <0.001). Similarly, 3- and 5-year OS rates after any metastasectomy for early metachronous metastases were 57 and 50 per centversus84 and 66 per cent for late metachronous metastases (P = 0.293). Conclusion The proportion of patients with metastatic colorectal cancer was consistent with that in earlier population-based studies, as were resection rates for liver and lung metastases and survival after resection. Differentiation between synchronous, early and late metachronous metastases can improve assessment of resectability and survival.Peer reviewe

    Immune cell profiles of metastatic HER2-positive breast cancer patients according to the sites of metastasis

    Purpose Recent works have characterized that metastatic site can affect the tumour immune profiles and efficiency of cancer immunotherapies. The prognosis of HER2-positive breast cancer is associated with the characteristics of the tumour immune microenvironment, with immunological cells playing a central role in efficiency of HER2-targeted antibodies. Here we investigated the prognostic significance of different metastatic sites and their correlation to tumour immune profiles in HER2-positive breast cancer treated with trastuzumab. Methods We collected all (n = 54) HER2-positive metastatic breast cancer patients treated with trastuzumab containing regimens at Oulu University Hospital 2009-2014. Pathological and clinical data were collected from electronic patient records. The tumour immune profiles were analysed from pre-treatment primary tumours using well-characterized immunological markers with computer-assisted immune cell counting. Results Of the metastatic sites, only liver metastases were associated with poor prognosis (hazard ratio 1.809, 95% confidence interval 1.004-3.262), especially when presented as the primary site of metastases. Of the other sites, pulmonary metastases characterized a patient profile with trend to improved survival. Of the studied tumour immunological markers, patients with liver metastases had low densities of CD3(+) T cells (p = 0.030) and M1-like macrophages in their primary tumours (p = 0.025). Of the other studied markers and sites, patients with pulmonary metastases had low STAB1(+)-immunosuppressive macrophage density in their primary tumours. Conclusion Our results suggest that the site of metastasis is associated with prognosis in HER2-positive breast cancer, highlighted by the poor prognosis of liver metastases. Furthermore, liver metastases were associated with adverse tumour immune cell profiles.Peer reviewe

    Effects of ionizing radiation and HPSE1 inhibition on the invasion of oral tongue carcinoma cells on human extracellular matrices in vitro

    Chemoradiation is an established approach in the treatment of advanced oral tongue squamous cell carcinoma (OTSCC), but therapy may cause severe side-effects due to signal interchanges between carcinoma and the tumour microenvironment (TME). In this study, we examined the potential use of our human 3D myoma disc and Myogel models in in vitro chemoradiation studies by analysing the effects of ionizing radiation (IR) and the combined effect of heparanase 1 (HPSE1) inhibitors and IR on OTSCC cell proliferation, invasion and MMP-2 and - 9 production. Finally, we analysed the long-term effects of IR by studying clones of previously irradiated and invaded HSC-3 cells. We found that in both human uterine leiomyoma-based extracellular matrix models IR inhibited the invasion of HSC-3 cells, but blocking HPSE1 activity combined with IR induced their invasion. Low doses of IR increased MMP expression and initiated epithelial-mesenchymal transition in cells cultured on myoma discs. We conclude that myoma models offer consistent methods for testing human carcinoma cell invasion and phenotypic changes during chemoradiation treatment. In addition, we showed that IR had long-term effects on MMP-2 and - 9, which might elicit different HSC-3 invasion responses when cells were under the challenge of HPSE1 inhibitors and IR.Peer reviewe

    Microwave studies of the fractional Josephson effect in HgTe-based Josephson junctions

    The rise of topological phases of matter is strongly connected to their potential to host Majorana bound states, a powerful ingredient in the search for a robust, topologically protected, quantum information processing. In order to produce such states, a method of choice is to induce superconductivity in topological insulators. The engineering of the interplay between superconductivity and the electronic properties of a topological insulator is a challenging task and it is consequently very important to understand the physics of simple superconducting devices such as Josephson junctions, in which new topological properties are expected to emerge. In this article, we review recent experiments investigating topological superconductivity in topological insulators, using microwave excitation and detection techniques. More precisely, we have fabricated and studied topological Josephson junctions made of HgTe weak links in contact with two Al or Nb contacts. In such devices, we have observed two signatures of the fractional Josephson effect, which is expected to emerge from topologically-protected gapless Andreev bound states. We first recall the theoretical background on topological Josephson junctions, then move to the experimental observations. Then, we assess the topological origin of the observed features and conclude with an outlook towards more advanced microwave spectroscopy experiments, currently under development.Comment: Lectures given at the San Sebastian Topological Matter School 2017, published in "Topological Matter.
