    A highly sensitive sandwich ELISA for the determination of glycomacropeptide to detect liquid whey in raw milk

    Milk processing industries and distributors have problems with adulteration of liquid milk by the addition of bovine cheese whey. Recently, the detection of fraudulent manipulation of milk with whey has focused on the identification of glycomacropeptide (GMP). Current non-immunological methods to detect GMP in dairy products are expensive and time-consuming or have low sensitivity. In this study, a novel sandwich enzyme-linked immunosorbent assay (ELISA) for the detection and quantification of whey in raw milk was developed, using a polyclonal rabbit anti-GMP antibody. Calibration curves were constructed by analyzing raw milk standards containing different known concentrations of liquid cheese whey (0.02–20%). The method had a detection limit of 0.047% (v/v) and a quantification limit of 0.14% (v/v). The antibody showed high specificity and no cross-reaction with milk components (other than κ-casein) and was successful in detecting GMP in dairy commercial products. The recovery ratio was between 95.62% and 113.88% for all matrices tested. The intra-assay and interassay coefficients of variation were <6% and <7%, respectively. Finally, it can be stored for 3 months in the form of a ready-to-use kit, while maintaining its accuracy and reproducibility

    A multiscale data-driven approach for bone tissue biomechanics

    The data-driven methodology with application to continuum mechanics relies upon two main pillars: (i) experimental characterization of stress–strain pairs associated to different loading states, and (ii) numerical elaboration of the elasticity equations as an optimization (searching) algorithm using compatibility and equilibrium as constraints. The purpose of this work is to implement a multiscale data-driven approach using experimental data of cortical bone tissue at different scales. First, horse cortical bone samples are biaxially loaded and the strain fields are recorded over a region of interest using a digital image correlation technique. As a result, both microscopic strain fields and macroscopic strain states are obtained by a homogenization procedure, associated to macroscopic stress loading states which are considered uniform along the sample. This experimental outcome is here referred as a multiscale dataset. Second, the proposed multiscale data-driven methodology is implemented and analyzed in an example of application. Results are presented both in the macroscopic and microscopic scales, with the latter considered just as a post-process step in the formulation. The macroscopic results show non-smooth strain and stress patterns as a consequence of the tissue heterogeneity which suggest that a preassumed linear homogeneous orthotropic model may be inaccurate for bone tissue. Microscopic results show fluctuating strain fields – as a consequence of the interaction and evolution of the microconstituents – an order of magnitude higher than the averaged macroscopic solution, which evidences the need of a multiscale approach for the mechanical analysis of cortical bone, since the driving force of many biological bone processes is local at the microstructural level. Finally, the proposed multiscale data-driven technique may also be an adequate strategy for the solution of intractable large size multiscale FE2 computational approaches since the solution at the microscale is obtained as a postprocessing. As a main conclusion, the proposed multiscale data-driven methodology is a useful alternative to overcome limitations in the continuum mechanical study of the bone tissue. This methodology may also be considered as a useful strategy for the analysis of additional biological or technological hierarchical multiscale materials

    Modelo de gestión para un control de calidad eficiente en edificios de nueva planta

    The management of the quality control of each building process is usually set up in Spain from different levels of demand. This work tries to obtain a model of reference, to compare the quality control of the building process of a specific product (building), and to be able to evaluate its warranty level. In the quest of this purpose, we take credit of specialized sources and 153 real cases of Quality Control were carefully revised using a multi-judgment method. Applying different techniques to get a specific valuation (impartial) of the input parameters through Delphi’s method (17 experts query), whose matrix treatment with the Fuzzy-QFD tool condenses numerical references through a weighted distribution of the selected functions and their corresponding conditioning factors. The model thus obtained (M153) is useful in order to have a quality control reference to meet the expectations of the quality.La gestión del control de calidad en cada proceso edificatorio se plantea habitualmente en España desde distintos niveles de exigencia. El presente trabajo tiene por objeto obtener un modelo de referencia para comparar el control del proceso edificatorio realizado sobre un producto concreto (edificio) y poder evaluar su nivel de garantía. Para ello recurrimos a fuentes especializadas así como al estudio de 153 casos reales de expedientes de control de calidad utilizando una metodología del tipo multicriterio. Se aplican técnicas conducentes a una valoración concreta (objetiva) de los parámetros de entrada mediante el método Delphi (consulta a 17 expertos), cuyo tratamiento matricial con la herramienta Fuzzy-QFD condensa referencias numéricas a través de un reparto de pesos entre las funciones seleccionadas y sus correspondientes factores intervinientes. El modelo así obtenido (M153) es útil como referente óptimo de las actuaciones de control que conllevan el cumplimiento de las expectativas de calidad

    Factors influencing academic achievement: the mediating role of motivation in learning strategies and school climate

    El objetivo principal de esta investigación consiste en analizar la relación existente entre estrategias de aprendizaje, motivación, clima escolar y el rendimiento académico y establecer cuáles son los mejores predoctores de este rendimiento. Para ello hemos utilizado una muestra de 101 alumnos de 4º ESO de centros de Granada y Málaga, a los que se les administró el CEAM II, para medir estrategias de aprendizaje y motivación; y el CECSCE para medir el clima escolar. Los resultados indican que altas puntuaciones en estrategias de aprendizaje, una mayor motivación y un mejor clima escolar se relacionan con un mejor rendimiento académico. A su vez, los resultados muestran que los factores que mejor predicen el rendimiento académico son la valoración de la tarea y percepción de autoeficacia (motivación), organización y esfuerzo (estrategias de aprendizaje) y percepción del centro (clima escolar). Por último, se observa que la motivación tiene un papel mediador entre las estrategias de aprendizaje y el clima escolar sobre el rendimiento académico.
