    Stock assessment of Haliporoides triarthrus (Fam. Solenoceridae) off Mozambique: a preliminary analysis

    The pink shrimp, Haliporoides triarthrus, is an important species in the deep-water shrimp fishery in Mozambique. Total catches are in the range of 1,500 to 2,700 tons, with the pink shrimp accounting for 70-90%. Estimates of growth parameters and of natural mortality are used for a preliminary assessment of the fishery, based on length-structured virtual population analysis and yield-per-recruit analyses. With an arbitrarily chosen terminal fishing mortality F, the results indicate a situation of overfishing, but realistic parameters must be acquired in order to diagnose the state of the stoc

    Fatty acid metabolism in Atlantic salmon (Salmo salar L.) hepatocytes and influence of dietary vegetable oil

    Isolated hepatocytes from Atlantic salmon (Salmo salar), fed diets containing either 100% fish oil or a vegetable oil blend replacing 75% of the fish oil, were incubated with a range of seven 14C-labelled fatty acids. The fatty acids were; [1-14C]16:0, [1-14C]18:1n-9, [1-14C]18:2n-6, [1-14C]18:3n-3, [1-14C]20:4n-6, [1-14C]20:5n-3, and [1-14C]22:6n-3. After 2 hours of incubation the hepatocytes and medium were analyzed for acid soluble products, incorporation into lipid classes, and hepatocytes for desaturation and elongation. Uptake into hepatocytes was highest with [1-14C]18:2n-6 and [1-14C]20:5n-3 and lowest with [1-14C]16:0. The highest recovery of radioactivity in the cells was found in triacylglycerols. Of the phospholipids the highest recovery was found in phosphatidylcholine, with [1-14C]16:0 and [1-14C]22:6n-3 being the most prominent fatty acids. The rates of β-oxidation were as follows: 20:4n-6 > 18:2n-6 = 16:0 > 18:1n-9 > 22:6n-3 = 18:3n-3 = 20:5n-3. Of the fatty acids taken up by the hepatocytes, [1-14C]16:0 and [1-14C]18:1n-9 were subsequently exported the most, with the majority of radioactivity recovered in phospholipids and triacylglycerols, respectively. The major products from desaturation and elongation were generally one cycle of elongation of the fatty acids. Diet had a clear effect on the overall lipid metabolism, with replacing 75% of the fish oil with vegetable oil resulting in decreased uptake of all fatty acids and reduced incorporation of fatty acids into cellular lipids, but increased β-oxidation activity, and higher recovery in products of desaturation and elongation of [1-14C]18:2n-6 and [1-14C]18:3n-3

    Effect of Marine Omega 3 Fatty Acids on Methylmercury-Induced Toxicity in Fish and Mammalian Cells In Vitro

    Methylmercury (MeHg) is a ubiquitous environmental contaminant which bioaccumulates in marine biota. Fish constitute an important part of a balanced human diet contributing with health beneficial nutrients but may also contain contaminants such as MeHg. Interactions between the marine n-3 fatty acids eicosapentaenoic acid (20:5n-3, EPA) and docosahexaenoic acid (22:6n-3, DHA) with MeHg-induced toxicity were investigated. Different toxic and metabolic responses were studied in Atlantic salmon kidney (ASK) cell line and the mammalian kidney-derived HEK293 cell line. Both cell lines were preincubated with DHA or EPA prior to MeHg-exposure, and cell toxicity was assessed differently in the cell lines by MeHg-uptake in cells (ASK and HEK293), proliferation (HEK293 and ASK), apoptosis (ASK), oxidation of the red-ox probe roGFP (HEK293), and regulation of selected toxicological and metabolic transcriptional markers (ASK). DHA was observed to decrease the uptake of MeHg in HEK293, but not in ASK cells. DHA also increased, while EPA decreased, MeHg-induced apoptosis in ASK. MeHg exposure induced changes in selected metabolic and known MeHg biomarkers in ASK cells. Both DHA and MeHg, but not EPA, oxidized roGFP in HEK293 cells. In conclusion, marine n-3 fatty acids may ameliorate MeHg toxicity, either by decreasing apoptosis (EPA) or by reducing MeHg uptake (DHA). However, DHA can also augment MeHg toxicity by increasing oxidative stress and apoptosis when combined with MeHg

    Towards Fish Lipid Nutrigenomics: Current State and Prospects for Fin-fish Aquaculture

    Lipids are the predominant source of energy for fish. The mechanisms by which fish allocate energy from lipids, for metabolism, development, growth and reproduction are critical for understanding key life history strategies and transitions. Currently, the major lipid component in aquaculture diets is fish oil (FO), derived from wild capture fisheries that are exploited at their maximum sustainable limit. The increasing demand from aquaculture for FO will soon exceed supply and threaten the viability of aquaculture. Thus, it is essential to minimize FO use in aquaculture diets. This might be achieved by a greater understanding of lipid storage and muscle growth, or the identification of alternatives to FO in feeds. This review focuses on recent research applying molecular and genomic techniques to the study of fin-fish lipid metabolism from an aquaculture perspective. Accordingly, particular emphasis will be given to fatty acid metabolism and to highly unsaturated fatty acid (HUFA) biosynthesis, and to the transcriptional mechanisms and endocrine factors that regulate these processes in fish. Comparative studies of gene function and distribution are described which, when integrated with recent fish genome sequence information, provide insights into lipid homeostasis and the outcomes associated with the replacement of FO in fish diets

    ICES coordinated acoustic survey of ICES divisions IIIa, IVa, IVb AND Via (North) 2002 Results and long term trends

    Six surveys were carried out during late June and July covering most of the continental shelf north of 54oN in the North Sea and to the west of Scotland to a northern limit of 62oN. The eastern edge of the survey area was bounded by the Norwegian and Danish, Swedish and German coasts, and to the west by the shelf edge between 200 and 400 m depth. The surveys are reported individually in the report of the planning group for herring surveys, and a combined report has been prepared from the data from all surveys. The combined survey results provide spatial distributions of herring abundance by number and biomass at age by statistical rectangle; and distributions of mean weight and fraction mature at age. The estimates of North Sea autumn spawning herring are consistent with previous years at 2.9 million tonnes and 17,200 million herring. The survey also shows two exceptional year classes of herring (the 1998 and 2000 year classes) in the North Sea, which is consistent with the observation of exceptionally large year classes observed in the MIK and IBTS surveys. The estimates of Western Baltic spring spawning herring SSB are 255,000 tonnes and 2.9 millions (Table 2) and show a large increase compared with the previous year. The Western Baltic survey produces a rather noisy signal but the indications are of a stock that is higher now than between 1996 to 2000. The West of Scotland survey estimates of 548,000 tonnes and 2,900 million and shows the high 1995 year class again this year. The 1998 year class now (3 ring) is also a large one. Total adult mortality shows much lower mortality than last year (0.1 compared to 0.5 ) but the mean mortality over the last 4 years has been 0.3: this is consistent with the 2002 assessment that the stock is lightly exploited. The overall time series of abundance by age from 1989 to 2002 are summarised by simple models describing the spatial distribution over time. The changes over time with latitude, longitude and area occupied are compared with changes in abundance

    Fatty acid utilisation and metabolism in caecal enterocytes of rainbow trout (Oncorhynchus mykiss) fed dietary fish or copepod oil

    A combined fatty acid metabolism assay was employed to determine fatty acid uptake and relative utilisation in enterocytes isolated from the pyloric caeca of rainbow trout. In addition, the effect of a diet high in long-chain monoenoic fatty alcohols present as wax esters in oil derived from Calanus finmarchicus, compared to a standard fish oil diet, on caecal enterocyte fatty acid metabolism was investigated. The diets were fed for 8 weeks before caecal enterocytes from each dietary group were isolated and incubated with [1-14C]fatty acids: 16:0, 18:1n-9, 18:2n-6, 18:3n-3, 20:1n-9, 20:4n-6, 20:5n-3, and 22:6n-3. Uptake was measured over 2 h with relative utilisation of different [1-14C]fatty acids calculated as a percentage of uptake. Differences in uptake were observed, with 18:1n-9 and 18:2n-6 showing the highest rates. Esterification into cellular lipids was highest with 16:0 and C18 fatty acids, accounting for over one-third of total uptake, through predominant incorporation in triacylglycerol (TAG). The overall utilisation of fatty acids in phospholipid synthesis was low, but highest with 16:0, the most prevalent fatty acid recovered in intracellular phosphatidylcholine (PC) and phosphatidylinositol (PI), although exported PC exhibited higher proportions of C20/C22 polyunsaturated fatty acids (PUFA). Other than 16:0, incorporation into PC and PI was highest with C20/C22 PUFA and 20:4n-6 respectively. Recovery of labelled 18:1n-9 in exported TAG was 3-fold greater than any other fatty acid which could be due to multiple esterification on the glycerol ‘backbone’ and/or increased export. Approximately 20-40% of fatty acids taken up were β-oxidised, and was highest with 20:4n-6. Oxidation of 20:5n-3 and 22:6n-3 was also surprisingly high, although 22:6n-3 oxidation was mainly attributed to retroconversion to 20:5n-3.     Contaminant levels in Norwegian farmed Atlantic salmon (<i>Salmo salar</i>) in the 13-year period from 1999 to 2011

    AbstractBackgroundEnvironmental pollutants such as dioxins and PCBs, heavy metals, and organochlorine pesticides are a global threat to food safety. In particular, the aquatic biota can bioaccumulate many of these contaminants potentially making seafood of concern for chronic exposure to humans.ObjectivesThe main objective was to evaluate trends of contaminant levels in Norwegian farmed Atlantic salmon in light of the derived tolerable intakes.MethodsThrough an EU-instigated surveillance programme, the Norwegian Food Safety Authority (NFSA) has between 1999 and 2011 collected more than 2300 samples of Norwegian farmed Atlantic salmon (Salmo salar) for contaminant analyses. The fillets of these fish were homogenised and analysed for dioxins, PCBs, heavy metals and organochlorine pesticides.ResultsThe levels of the contaminants mercury, arsenic, dioxins, dioxin-like PCBs and DDT in Norwegian farmed salmon fillet have decreased during our period of analyses. The levels of cadmium, lead and several organochlorine pesticides were too close to the limit of quantification to calculate time trends. For PCB6 and quantifiable amounts of pesticides, except DDT, stable levels were observed.ConclusionThe contaminant levels in Norwegian farmed salmon have generally decreased between 1999 and 2011. Excluding other dietary sources, the levels of dioxins and dioxin-like PCBs in 2011 allowed consumption of up to 1.3kg salmon per week to reach the tolerable weekly intake. The group of contaminants which was the limiting factor for safe consumption of Norwegian farmed salmon, based on currently established TWI values, is the sum of dioxins and dioxin-like PCBs

    Diet x genotype interactions in hepatic cholesterol and lipoprotein metabolism in Atlantic salmon (Salmo salar) in response to replacement of dietary fish oil with vegetable oil

    The present study investigates the effects of genotype on responses to alternative feeds in Atlantic salmon. Microarray analysis of the liver transcriptome of two family groups, lean or fat, fed a diet containing either a fish oil (FO) or a vegetable oil (VO) blend indicated that pathways of cholesterol and lipoprotein metabolism might be differentially affected by the diet depending on the genetic background of the fish, and this was further investigated by real-time quantitative PCR, plasma and lipoprotein biochemical analysis. Results indicate a reduction in VLDL and LDL levels, with no changes in HDL, when FO is replaced by VO in the lean family group, whereas in fat fish fed FO, levels of apoB-containing lipoproteins were low and comparable with those fed VO in both family groups. Significantly lower levels of plasma TAG and LDL-TAG were measured in the fat group that was independent of diet, whereas plasma cholesterol was significantly higher in fish fed the FO diet in both groups. Hepatic expression of genes involved in cholesterol homeostasis, β-oxidation and lipoprotein metabolism showed relatively subtle changes. A significantly lower expression of genes considered anti-atherogenic in mammals (ATP-binding cassette transporter A1, apoAI, scavenger receptor class B type 1, lipoprotein lipase (LPL)b (TC67836) and LPLc (TC84899)) was found in lean fish, compared with fat fish, when fed VO. Furthermore, the lean family group appeared to show a greater response to diet composition in the cholesterol biosynthesis pathway, mediated by sterol-responsive element-binding protein 2. Finally, the presence of three different transcripts for LPL, with differential patterns of nutritional regulation, was demonstrated
