    Analytical Method Validation of Benzene Using High Performance Liquid Chromatography in Beverage Containing Sodium Benzoate and Ascorbic Acid

    Several countries reported discovering benzene in beverages containing benzoic acid and ascorbic acid. Benzoic acid decarboxylation by ascorbic acid will form benzene. American Beverage Association (ABA) recommended the use of accelerated testing to test benzene in beverages. High Performance Liquid Chromatography (HPLC) is one of several methods to analyze benzene in a wide variety of samples, but there is no much information provided regarding the validation of analysis method of benzene. Therefore, developing analysis method of benzene and its validation becomes a current need. The HPLC system consists of Hitachi L-2130 pump, sample injector with 20 µL sample loop, and UV detector L-2420 operating at 205 nm. The analytical column is a LiChrosorb Phenomenex RP-18 (250 x 4 mm, 10 µm, 100 Ǻ), the mobile phase is acetonitrile:aquabidest (60:40 v/v) and pumped at a flow rate of 0,8 mL/min.Benzene separated from the matrix and follows the validation requirements. The developed analytical method showed that resolution was 8.37, r = 0.995 with LOD and LOQ 6.52 ppb and 19.75 ppb, with a precise of ≤11% and recovery of 80-110%. Accelerated testing indicated that benzene levels increased with increasing of the temperature. Beverages containing 400 mg/mL of ascorbic acid and benzoic acid formed benzene which was detected as 699.38 ppb at 25 oC, 799.61 ppb at 40 oC, and 808.94 ppb at 60 oC in 48 hours. In conclusion, the method was fully validated and can be utilized to analyze benzene in beverages with the accelerated testing at 60 oC in 48 hours, so that benefits the producers and consumers in the end

    Validation for the Quantification of Andrographolide Isolated From Andrographis Paniculata Nees Plant Using HPLC

    The aim of study was to develop quantitative analysis of isolated andrographolide from Andrographis paniculata and different solvent for prelimenary studies to preperation Self Nano Emulsifying Drug Delivery System (SNEDDS) using HPLC. The separation was acquired on Sunfire C18 column with an isocratic mixture of methanol and water at a ratio of 6:4, v/v as a mobile phase. The method to determine the content of isolated andrographolide showed an adequate precision, with a RSD smaller than 1%. The accuracy was analyzed by adding the standard andrographolide, and good recovery values were obtained for all concentrations used. The HPLC method developed in this study showed specificity and selectivity with linearity in the working range and good precision and accuracy, making it very suitable for the quantification of isolated andrographolide. Compared to the standard, the purity of the isolated andrographolide was 95.74 ± 0.29 %. Prelimenary study to determined the highest solubility of isolated andrographolide in oil, surfactant and co-surfactant phases for preperation of SNEDDS were obtained 1.226 ± 0.009 of Capryol-90, 2.965 ± 0.014 of tween 20, and 6.074 ± 0.101 mg mL-1 of PEG 400, respectively.     Vitamin C, Vitamin A And Alpha Hydroxy Acid In Bengkoang (Pachyrhizus Erosus)

    Bengkoang or Pachyrhizus erosus has been used as a traditional cosmetic material since many years ago. Until now, many cosmetic preparations using bengkoang as a main material have been produced by cosmetic industries, mainly for skin care, whitening and sunscreen preparation. However, content of vitamin A, vitamin C and alpha hydroxyl acid has not been discovered yet. These compounds are essential in skin care process because of their activities in cell regeneration, antioxidant as well as peeling of dead skin cell layer. The aim of this research is to provide information about the content of three compounds in bengkoang. Vitamin A has been analysed by spectrophotometer, vitamin C analysed using titration method and alpha hydroxyl acid analysed using gas chromatography. The result showed that concentrations of vitamin A and vitamin C are 179.21 ± 8.19 ppm and 0.31 ± 0.06 %, respectively. Meanwhile the content of alpha hydroxyl acid was 0.80 ± 0.01% (measured as glycolic acid)

    Glycaemic Index Of Uwi, Gadung, And Talas Which Were Given On Rat

    The aim of the study was the determination of glycaemic index (GI) of local tubers such as Uwi (Dioscorea alata), Gadung (Dioscorea hispida), dan Talas (Colocasia esculenta). Tubers were prepared as raw flour which were administered orally to the rats. Blood were taken at 0; 0,5; 1 and 2 hour after tubers administration. The dose was 2,5 kg/kg body weight (BW) whereas glucose was administered as a standart treatment. Area Under Curve (AUC) was calculated after plotting the graph of time versus glucose levels. Glucose levels were determined using GOD-PAP kit. GI values were calculated as a ratio of AUC (0-2hr) of tubers to glucose. Porang was moderate while the other tubers were low GI

    ABSTRACT Polyaromatic Hydrocarbons (PAHs) are a class of organic chemicals, composed of fused benzene rings, occured in the environment due to incomplete combustion of fossil fuels. Health concerns are focussed to the metabolite transformation of PAHs, which is carcinogenic, mutagenic and terratogenic. Due to their hydrophobidty. so there are probable into human food chain. The aims of this research is to study resolution of eight component mixture PAHs (fluoranthene, pyrene, benzo(a)anthrasene, chrysene, benzo(k)fluoranthene, benzo(a)pyrene, perylene, and dibenz(a,h) anthrasene) by High Performance Liquid Chromatography (HPLC). The result shown that optimally condition of HPLC as follows: stationary phase is Clg with size particle 5 mm, column length 20 cm and inside diameter lcm. Mobile phase is acetonitrile-water with gradien elution programme from 70% of acetonitrile in water to 80%. Detector is uv-vis spectrophotometer at 254nm.
